Abstract

Background Formaldeyde is main substance used for organ embalming and even for foods preservations. However, long and high exposure of formaldeyhde lead to various organs irritations. Honey is well-known for its natural preservative activity expected to be the soultion for the problem so there is safe alternative for organ preservations.Aim Observation and examintaion the difference of macroscopic and microscopics feature of Wistar rats liver after immersion of formaldehyde and randu honey.Methods Experimental with post-test only control group design using 24 male Wistar rats as sample, randomly divided into 4 groups consist of: Group C (preserved with 10% formaldehyde solutions), P1 (preserved with 10% randu honey solutions), P2 (preserved with 20% randu honey solutions) and P3 (preserved with 30% randu honey solutions). The sampels were preserved for 24 hours then macroscopic (smell,color and size) and microscopic study with Hematoxylin-Eosin (HE) stain  using 400x observation performed. Manja Roenigk score were used to accessed the degree of live microsopic damage.Results There are normal cells, mild damage, moderate damage and severe damage respectively in the group C: 36%,47%,17%,0% ; P1: 0%,27%,46%,27% ; P2: 0%,77%,17%,6% ; P3: 47%,33%,17%,3%. Kruskal-Wallis test showed significant results with p=0.000. With Mann-Whitney test, significant results were found between group C with P1 (0.000), C with P2 (0.006), P1 with P2 (0.000), P1 with P3 (0.000) and P2 with P3 (0.003). However, test result were not significant berween group C with P3 (0.684).Conclusions There were differences in macroscopics and microscopic features between Wistar rats liver after immersion in formaldehyde and randu honey. The best features showed in samples preserved in 30% Randu Honey solutions.

Abstract

Background: Basal cell carcinoma is one of the most common cancers in the world, and its incidence is increasing over time. It is estimated that it accounts for three-quarters of all non-melanoma skin cancers, and the incidence increases exponentially in predominantly white areas. The p53 gene is the gene that most undergoes mutations due to sun exposure and is the cause of skin cancer, especially basal cell carcinoma. This finding explains that p53 expression can be used as marker analysis in cases of basal cell carcinoma. Methods: Observational analytic design was carried out with a cross-sectional. The population and samples were paraffin blocks which histopathologically diagnosed Basal Cell Carcinoma in the anatomical pathology laboratory, Dr. Kariadi General Hospital Semarang. Samples were processed by the p53 immunohistochemical staining method. The p53 expression was given a quantitative score to be 0 = negative, 1-2 = low, 3-4 = high. The expression of p53 was considered positive when it was colored in the cell nucleus. The correlation between variables was analyzed by Spearman and Mann Whitney correlation test. Results: There is no correlation between p53 expression and the age of p53 patients with p = 0.390 (p > 0.05). There is no correlation between p53 expression and lesion location with p = 0.817 (p > 0.05) There is no difference significant, between p53 expression and gender with p = 0.576 (p > 0.05). Conclusion: There is no correlation between high and low-risk Basal Cell Carcinoma subtype associated with age, gender, and lesion location.

Abstract

Latar Belakang : Kanker payudara merupakan kanker terbanyak pada wanita yang menyebabkan kematian di Indonesia. Ekspresi HER2 yang overexpressed pada kanker payudara merupakan penanda prognosis yang buruk. Tumor Associated Macrophage (TAM) berperan dalam pertumbuhan, invasi dan metastasis tumor dan berkaitan erat dengan prognosis yang buruk. Penelitian ini bertujuan mengetahui hubungan antara ekspresi TAM/CD68 dengan status ekspresi HER2 pada karsinoma payudara invasif.Metode : Penelitian ini merupakan penelitian observasional analitik dengan desain case control, dengan jumlah sampel total 50 blok paraffin dari pasien yang telah didiagnosis invasive carcinoma of no special type di RSUP dr. Kariadi Semarang pada tahun 2015-2016, yang kemudian dibagi menjadi 2 kelompok : HER2 positif (+) dan HER2 negatif (-). Data yang diambil adalah distribusi dan intensitas ekspresi CD68 menggunakan Allred immunohistochemistry score pada 5 lapangan pandang besar pada masing-masing sampel.Hasil : Jumlah distribusi ekspresi CD68 kelompok HER2 positif lebih banyak daripada kelompok HER2 negatif pada skor 3, 4 dan 5, dengan perbedaan yang bermakna (p = 0,001). Untuk intensitas ekspresi CD68, kelompok HER2 positif memiliki jumlah sel dengan intensitas kuat lebih banyak secara bermakna dibandingkan kelompok HER2 negatif. (p = 0,005).Kesimpulan : Karsinoma payudara invasif HER2 positif memiliki distribusi dan intensitas ekspresi TAM/CD68 yang lebih banyak dan lebih kuat secara bermakna dibanding kelompok karsinoma payudara invasif HER2 negatif. Kata Kunci : Tumor Associated Macrophage (TAM), CD68, HER2, Kanker Payudara Invasif

Abstract

Latar Belakang: Lebih dari 90% keganasan pada kulit merupakan tumor keratinocyte,dimana 70%-nya merupakan Basal cell carcinoma (BCC) dengan angka kejadian yang semakin meningkat.Diagnosis BCC yang tepat dengan menyingkirkan diagnosis bandingnya sangatlah diperlukan, menggunakan pemeriksaan CD10 dan pengecatan PAS. Penelitian ini bertujuan untuk membandingkan pemeriksaan CD10 dan PAS pada BCC untukpenegakan diagnosis BCC.Metode: Penelitian observasional analitik dengan desain case control.Sampel adalah 36 blok paraffin dari pasien tumor kulit yang telah didagnosis basal cell carcinoma di RSUP dr. Kariadi Semarang pada Januari 2014 hingga Desember 2016.Data yang diambil adalah ekspresi CD10 dan PAS.Hasil: Sebanyak 30 dari 36 sampel menunjukkan ekspresi CD10 positif (83,3%). Pengecatan PASdari 36 sampel karsinoma sel basal, sebanyak 28 ekspresi PAS positif (77,8%). Uji chi square, menunjukkan perbedaan tidak bermakna antara ekspresi CD10 dan PAS pada karsinoma sel basal (p = 0,473).Simpulan: Tidak terdapat hubungan bermakna antara CD 10 dan PAS pada BCC, sehingga keduanya dapat dipakai untuk menyokong diagnosis BCC dan membedakan BCC dengan diagnosis bandingnya. Kata Kunci: BCC, CD10, PAS

Abstract

Latar Belakang: CEA merupakan penanda tumor yang klasik pada kanker kolorektal dan telah digunakan untuk penegakan diagnosis, evaluasi rekurensi dan sebagai faktor prognostik. Penelitian ini bertujuan membandingkan perbedaan kadar serum CEA saat diagnosis awal ditegakkan dan hubungan dengan adanya rekurensi lokal pada pasien dengan kanker rektum.Metode: Penelitian retrospektif ini mengikutsertakan 42 pasien dengan kanker rektum yang diambil dari rekam medis sejak Januari 2013 sampai Desember 2014, terdiri dari 23 wanita dan 19 pria, lokasi tumor rektum, kadar serum CEA preoperasi, tindakan operasi, jenis histopatologi dan adanya rekurensi lokal.Hasil: Tidak didapatkan perbedaan yang signifikan antara kadar serum CEA preoperasi terhadap lokasi tumor (p=0.106 vs p=0.100), tidak berbeda signifikan antara rekurensi lokal terhadap lokasi (p=0.525 vs p=0.200). Kecenderungan rekurensi lokal pada kadar serum CEA preoperasi dengan cut off point 8,75 mg/dl (sensitivitas 66.7%, spesifisitas 33.3%).Kesimpulan: Perbedaan lokasi tumor secara anatomis terhadap refleksi peritoneal tidak mempengaruhi perbedaan kadar serum CEA preoperasi dan kecenderungan adanya rekurensi lokal pada kadar serum CEA preoperasi dengan cut off point 8,75 mg/dl (sensitivitas 66.7%, spesifisitas 33.3%).  Kata Kunci: CEA, rekurensi lokal, kanker rektum

Abstract

Background: Systemic inflammatory response syndrome (SIRS) is a state of systemic inflammatory activation by various causes. SIRS have a high mortality rate. Prolactin is known to regulate cellular function of immune system.  Neutrophil-lymphocyte ratio (NLR) is simple, cost effective and easy parameter that currently used as inflammation marker.Objective: The aims of this study is to determine the correlation between prolactin serum with NLR in SIRS patients.Methods: A cross sectional study was conducted on 50 clinically SIRS patients. Prolactin serum was measured by enzyme linked immunosorbent assay (ELISA) and NLR was calculated manually from absolute neutrophil and lymphocyte count measured by automatic hematology analyzer. Non-parametric Spearman test was used to analyze the correlation between prolactin with NLR.Results: Median value of serum prolactin level was 11.32 ng/mL (2.76-194.81), whereas the mean value NLR was 16.36 ± 11.58. The correlation between prolactin levels with NLR was r = 0.345, p = 0.014.Conclusion: There is a weak positive significant correlation between prolactin with neutrophil lymphocyte ratio in SIRS

Abstract

ABSTRACT Background:Intra ocular pressure (IOP) elevationis one of topical steroids’ side effects. Corticosteroid will initiate matrix metalloproteinase cascade that leads to extra cellular matrix (ECM)of trabecular meshwork (TM) turnover(remodeling) and its acumulationin TM. Objective: To analyze the reversibility of ECM TM in Wistar rat after discontinuation oftopical prednisolone1% at different periode(14 days and 28 days). Methods:This was an experimental study with post test only controlgroup design. Total samples of 28 rats were divided into 4 groups: Treatment groups 1 was treated with  topical prednisolone for 28 days, and was terminated 14 days after discontinuation of the drug.Treatment goups2 was treated topical prednisolone for 28 days and was terminated28 days after discontinuation of the drug. Control 1 was treated withtopical prednisolone for 28 days, and Control 2 was treated with topical saline for 28 days and terminated without periode of discontinuation. Histopathological grading score was used to evaluate ECM TM deposition. Mann-Whitney test and Kruskal-Wallis test were used to analyze the data. Result: Deposition of ECM in TM was not statistically different intreamentgroup 1 and treatment group 2 (p>0.05). Deposition of ECM in TM were statistically differentbetween treatment group 2 and control group 1 (p<0.05). Comparative test showed p<0.001,which means that there was a change in the  thickness of ECM after discontinuation of instillation. Conclusion: ECM TM was thinner in the experimental animals with a longer duration of topical prednisolondiscontinuation, which demonstrate that maintenance remodeling of ECM was happen.
