Abstract

Background: Cataract is a serious problem in the world because it is the biggest cause of blindness in the world. Cataract healing can only be done with operative measures, one of them is phacoemulsification. The phacoemulsification technique is the latest operative technique for cataract healing but both conventional techniques or phacoemulsification techniques can cause a decrease in tear secretion after cataract surgery. The reduction in tear secretion can be evaluated using the Schirmer I test. In this study an analysis of differences in tear secretion before and after phacoemulsification surgery.Aims: Knowing, comparing, and analyzing the differences in tear secretion before and after phacoemulsification surgery using the Schirmer I test.Methods: 22 cataract eyes consisting of 15 patients who will undergo phacoemulsification surgery were collected by consecutive sampling at the Sultan Agung Eye Center, Sultan Agung Hospital, Semarang from August to September 2019, examined using the Schirmer I test. After the data was collected, data analyzed with Wilcoxon rank test.Results: A total of 15 cataract patients consisting of 10 female patients and 5 male patients with a total of 22 eye samples. The average tear secretion before and after phacoemulsification surgery was 12.77 ± 2.74 mm and 11.00 ± 1.95 mm. Data were tested with the Wilcoxon rank test, p = 0,000 (p <0.05) which showed a difference in average tear secretion before and after phacoemulsification.Conclusion: There are differences in tear secretion before and after phacoemulsification surgery.

Abstract

Background: Myopia is one of the visual disorders that has a high prevalence in the world. Based on the severity degree, myopia is divided into three criteria, namely mild, moderate and severe. Severe-degree myopia patients are more likely to suffer from certain eye diseases, especially those associated with the retina, such as retinal detachment.Objective: To analyze the relationship between the degree of myopia and the incidence of retinal detachment.Methods: This research is an observational analytical with cross-sectional approach. The research data was obtained using consecutive sampling from the medical records of myopia patients in Diponegoro National Hospital during 2015-2020. Research subjects used in this research were 70 patients who fulfill the inclusion and not the exclusion criteria. The data were analyzed using Chi Square test.  Results: The prevalence of retinal detachment is dominated by the age group of 51-60 years old (18.6%), male (35.7%) and a severe degree of myopia (34.3%). The results of the Chi Square test show that there is a significant relationship between the severity degree of myopia and the incidence of retinal detachment.Conclusion: There is a significant relationship between the degree of myopia and the incidence of retinal detachment.

Abstract

Background: One modality type of burn wound therapy is topical silver sulfadiazine. Recently, research about topical herb medicine has grown. Moringa oleifera (MO) is a kind of herb plant that has an anti-inflammation effect and has an influence on angiogenesis, so it tends to faster burn wound healing. This study aimed to prove the effect of ethanolic extract of Moringa oleifera leaves on the macrophage count and VEGF expression in Wistar rats with burn wounds. Methods: The research design that was used is "Randomized post-test with a control group." The study population used 24 male Wistar rats that were induced burn wounds and randomly divided into 4 groups which were given medicine topically once daily for 7 days. Treatment groups included: I (MO leaves extract 10%), II (SSD + MO leaves extract 10%), III (SSD), dan IV (pure vehiculum). Exudation was assessed macroscopically, while PMN and macrophage amount were assessed microscopically with Hematoxylin-Eosin. Data were analyzed and processed using hypothesis test One Way ANOVA – Post Hoc Bonferroni, Kruskal Wallis – Mann Whitney, and correlation test Spearman and Pearson with SPSS 25.0 program. Results: Significant difference obtained in macrophage amount between II group and IV group (p 0,000). There was no significant difference in VEGF expression. There was also no correlation between macrophage amount and VEGF expression. Conclusion: Ethanolic extract of Moringa oleifera leaves extract proved to be effective in decreasing macrophage cell count on Wistar rats with burn wounds.

Abstract

Background: Burns are a major cause of morbidity, including prolonged length of stay and disability, which requires no small amount of treatment costs. Currently, there are many studies to accelerate the healing of wounds, one of which is with Moringa oleifera (MO). This study aimed to prove the effect of ethanolic extract from MO leaves on collagen density and new capillary vessel count on Wistar rats' burn wounds. Methods: This research is randomized post-test only with a control group design. Twenty-four male Wistar rats induced burn wounds and were randomly divided into 4 groups which were given topical medicine once daily for 10 days; P1 (MO leaves extract 10%), P2 (Silver Sulfadiazine + MO leaves extract 10%), P3 (SSD), and P4 (negative control). Collagen density was assessed using the Kruskal-Wallis test and continued with Mann-Whitney Test. New capillary vessel count was assessed using Hematoxylin-Eosin staining and One Way ANOVA. The correlation test between two variables was tested using the Spearman test. Results: Collagen Density shows significant difference in group P1 compared to P4 (p=0,016), group P2 compared to P3 (p=0,047), group P2 compared to P4 (p=0,009). A significant increase in new capillary blood vessel counts was seen in group P1 compared to P3 and P4 (p=0.001; 0,000) and group P2 compared to P3 and P4 (p=0,000;0,000). A positive correlation was found between the new capillary blood vessel count and epithelization percentage (p=0,001), with a strong correlation (rho=0,682). Conclusion: Ethanolic extract from Moringa oleifera leaves proved to be effective in increasing collagen density and new capillary vessel count.

Abstract

Latar Belakang: Trauma kimia pada permukaan bola mata merupakan masalah umum dengan derajat yang berbeda mulai ringan sampai berat. Trauma basa kuat meningkatkan pH jaringan, sehingga ion hidroksida (OH-) penetrasi pada jaringan epitel kornea menyebabkan saponifikasi membran sel dan hidrolisis jaringan. Cyclooxygenase-2 (COX-2) merupakan enzim yang penting dalam sintesis prekursor mediator inflamasi. Obat anti inflamasi steroid dapat menghambat hal ini, sehingga memiliki efek anti inflamasi yang poten, namun memiliki banyak efek samping. Phaleriamacrocarpa adalah tanaman obat asli Indonesia yang memiliki efek anti inflamasi. DLBS1425 merupakan sediaan farmasi ekstrak Phaleriamacrocarpa yang memiliki efek anti inflamasi. Peneliti ingin mengetahui efek anti inflamasi DLBS1425 di bidang mata, dinilai dari ekspresi COX-2 kornea tikus Wistar pasca trauma basa.Tujuan: Membuktikan DLBS1425 topikal berbagai konsentrasi memiliki efek terhadap ekspresi COX-2 kornea tikus Wistar pasca trauma basa.Metode: Merupakan penelitian post-test only randomized controlled group design. 24 mata tikus Wistar mendapat paparan NaOH 1M, dibagi menjadi 4 kelompok. Kelompok K diberi tetes Hyalub, sedangkan P1, P2, dan P3 diberi tetes DLBS1425 konsentrasi 1x101mg/ml, 1x100mg/ml,dan1x10-1mg/ml. Setelah 7 hari, dinilai ekspresi COX-2 kornea secara imunohistokimia. Analisis statistik menggunakan uji Kruskal Wallis dan Mann Whitney.Hasil: Rerata ekspresi COX-2 kelompok K=4,86, P1=3,30, P2=3,73, dan P3=4,13. Terdapat penurunan pada semua kelompok perlakuan,dibanding kontrol, dimana P1 berbeda signifikan secara statistik (p=0,015).Kesimpulan: DLBS1425 topikal konsentrasi 1x101mg/ml memiliki efek penurunan ekspresi COX-2 kornea tikus Wistar pasca trauma basa dibanding kontrol. Kata kunci: Trauma basa, inflamasi kornea, DLBS1425, COX-2

Abstract

Latar Belakang: Kornea merupakan jaringan mata yang bersifat transparan dan avaskuler, sifat tersebut diperlukan dalam fungsi penglihatan normal, dipertahankan oleh keseimbangan faktor angiogenik dan antiangiogenik. Vascular Endothelial Growth Factor (VEGF) berperan dalam angiogenesis, terdapat peningkatan ekspresi VEGF pada neovaskularisasi kornea. Penelitian sebelumnya menunjukkan bahwa fraksi bioaktif Mahkota Dewa (Phaleria macrocarpa), DLBS1425, memiliki efek antiangiogenesis dengan cara menghambat ekspresi mRNA VEGF-C pada sel kanker payudara. Penelitian ini ingin mengetahui efek antiangiogenesis DLBS1425 di bidang mata, dinilai dari ekspresi VEGF kornea tikus Wistar pasca trauma.Tujuan: Membuktikan DLBS1425 topikal berbagai konsentrasi memiliki efek terhadap ekspresi VEGF kornea tikus Wistar pasca trauma basa.Metode: Penelitian ini merupakan true experimental post-test only design. Dua puluh empat ekor tikus Wistar mendapat paparan NaOH 1M dengan diameter 1 mm, dibagi secara acak menjadi 4 kelompok. Kelompok K diberi tetes Hyalub, kelompok P1 diberi tetes DLBS1425 konsentrasi 1 x 101 mg/ml, kelompok P2 diberi tetes DLBS1425 konsentrasi 1 x 100 mg/ml, kelompok P3 diberi tetes DLBS1425 konsentrasi 1 x 10-1 mg/ml. Setelah 7 hari, dinilai ekspresi VEGF kornea secara imunohistokimia. Analisis statistik menggunakan uji Kruskal Wallis.Hasil Penelitian: Rerata ekspresi VEGF pada kelompok K=4,93, kelompok P1=4,33, kelompok P2=4,47, kelompok P3=4,77. Ekspresi VEGF kelompok perlakuan lebih rendah dibandingkan kelompok kontrol (p=0,134).Kesimpulan: DLBS1425 topikal konsentrasi 1x10-1, 1x100 dan 1x101 memiliki efek terhadap ekspresi VEFG kornea tikus Wistar pasca trauma basa. Ekspresi VEGF pada kelompok perlakuan lebih rendah dibandingkan dengan kelompok kontrol. Kata kunci: Phaleria macrocarpa, DLBS1425, VEGF, neovaskularisasi kornea

Abstract

Background: Phaleria macrocarpa (DLBS1425) is a medicinal plant that has the effects of anti-cancer, anti-inflammation, anti-angiogenesis, and antioxidants. The potency of its toxicity is low towards normal cell. DLBS1425 is expected to be useable for eye drop, but its toxic effect towards goblet cell proliferation and mucin secretion still remain unknown.Methods: This is experimental study with design of comparison test. Twenty four rabbits’ eyes divided into 4 groups were given placebo and topical DLBS1425 with concentration of 1×10-1 mg/ml, 1×100 mg/ml, and 1×101 mg/ml. Goblet cell proliferation degree and mucin secretion were evaluated using histopathology examination from rabits’ conjunctiva.Results: In the group of goblet cell proliferation, compared to control group, there was decreased proliferation degree with topical DLBS1425 of 1×10-1 mg/ml; but the concentration of 1×100 mg/ml and 1×101 mg/ml did not affect goblet cell proliferation. In the group of mucin secretion, DLBS1425 with any concentration level decreased mucin secretion significantly.Conclusion: There was significant difference in goblet cell proliferation with topical DLBS1425 of 1×10-1 mg/ml and placebo. There was also significant difference in mucin secretion with topical DLBS1425 in any concentration level and placebo. Keywords: DLBS1425, goblet cell, mucin secretion
