Abstract

Characterization of morphophysiology and oil yield of 10 patchouli genotypes (Pogostemon cablin Benth.) aims to test the morphology and physiology character of nine genotypes Aceh patchouli and one comparative clone (Lhokseumawe), obtain high yield oil clones (weight plant and essential oil content), and obtain genetic and phenotypic variabilities that can be used as a new genotype selection indicators. The research was conducted at Experimental Field and Plant Laboratory of the State Polytechnic of Lampung in April until November 2016. This research used Randomized Block Design (RBD) with 10 treatment levels and 3 replications.The LSI test results on the physiological character of the nine genotypes and one comparative clone showed that NPL 9 genotypes superior to the variables of the angle of petiole, chlorophyll content, leaf area, specific leaf area, and leaf area index. The NPL 9 genotypes resulted higher yield and rendement yields than other genotypes tested and one comparator, that was 2,36%. The character of stem production and rendement has a genetic variant value greater than the environmental variant. For genetic variability and phenotypic variability in the variables observed relatively uniform so that can not be used as an indicator of selection.   Keywords: Aceh patchouli, leaf character, selection indicator, variability of genotype, variability of phenotype

Abstract

The importance of patchouli plants to produce essential oil has established this medicinal herb as one of the Indonesia’s main economic contributor. To date, Indonesia can only provide 5 varieties i.e. Sidikalang, Tapak Tuan, Lhokseumawe, Patchoulina 1 and Patchoulina 2. In order to increase national patchouli production, several efforts needs to be done by providing new superior patchouli lines. This experiment was undertaken to reveal diversity of mutant progenies derived from gamma ray irradiation in patchouli. Six SSR primers were used to determine genetic diversity on MV3 generation of the mutants. Molecular analysis revealed a moderate to high polymorphism based on tested primers, which amplified 73 bands, and resulting of 0.705 PIC value. Dendrogram analysis based on Euclidean dissimilarity showed broad diversity that ranged from 0.23 to 0.82. Phenotypic tree gave clear separation that all mutant clones were clustered into three different group, with Eucledian dissimilarity coefficient ranging from 0.02 to 0.13. This report indicates that Patchouli vegetatively-propagated mutants were able to maintain their variability continuously from their parental lines (M0). Furthermore, our findings offer comprehensive information on breeding strategies of patchouli plants, giving an opportunity to assess important traits at early generation.

Abstract

This study aims to get a higher cup lump production in rubber plants that are given a stimulant treatment compared to rubber plants that are not given a stimulant. Higher levels of dry rubber in PB 260 that has been given a stimulant compared to the RRIC 100 who has been given a stimulant and clone that provide the best responses to the stimulant. This research was conducted at Afdeling 5 PTPN VII Padang Pelawi starting from March to May 2019. The method used a t-test with PB 260 and RRIC 100 clones. The observed variables were stem girth, tapping angle, tapping depth, tapping thickness, skin consumption, cup production lumps, and dry rubber content. The results obtained by the use of stimulants increased cup lump production in two clones studied, RRIC 100 clones gave the best response to stimulants compared with PB 260 clones.
