Abstract

Antioxidant activity and identification of antioxidative compounds of Callyspongia sponge from Seribu Island (Kepulauan Seribu) were investigated. The sponge was extracted with acetone and the extract was concentrated using rotary vacuum evaporator. DPPH and tiocyanate methods were used to examine the antioxidant activity of the extract. The extract exhibited strong antioxidant activity in DPPH method with IC50 of 41.21 µg/ml. Chemical analysis indicated that the antioxidative compound in the sponge was alkaloid group.

Abstract

Rosella calyx (Hibiscus sabdariffa) has been reported exhibited on decreasing high blood pressure activity. Celery herb (Apium graveolens) has been acknowledged as an antihypertension herb with vasodilator and diuretic effect. Combination of the extracts was expected to provide synergism effect on decreasing high blood pressure. This study was intended to prepare the combination of herbal tea from standardized extracts of rosella calyx and celery herb. Standardization of rosella calyx and celery herb included determination of several specific and non-specific parameters. Three different formulas of herbal tea were prepared that would be hedonically tested to obtain the most favorable herbal tea formula from the 30 panelists. The most favorable formula was formula C which contained rosella calyx (2 gram) and celery herb (0.5 gram).

Abstract

Leaves of tamarind (Tamarindus indica L.) has been traditionally used to treat some diseases such as: constipation, dyspepsia, flatulence and urinary tract infection. The leaves extract also reported has antibacterial and antidiabetic activities. It is essential to determine its specific and non specific parameters in assesing the quality and safety. The samples collected from three different perferctures. Grounded samples were macerated with ethanol. The extracts were subjected to determine its characteristic parameters. Parameter of quality for the extracts consisted of water content, ethanol-extractive, water-extractive, loss on drying, total ash, acid-insoluble ash, solvent residue and heavy metal content. Phytochemical identification showed that the ex-tract contains flanovoid, tannin, glycosides, and saponin. TLC chromatogram using chloroform-methanol-water (80:12:2) as mobile phase, exhibited 4 spots after sprayed with iron(III) chloride. Total phenolic was determined pectrophotometrically using Folin Ciocalteu reagent, gave 0.35-8.24% total phenolic as gallic acid.

Abstract

Antioxidant has been known to have an important role in the prevention and treatment of diseases. In recent years, many plants have been explored to obtain new antioxidants from natural products. Rice has been known rich in antioxidative compounds. However, there is little attention to the co- or by- products from rice (Oryza sativa) milling plant. One of the products is rice bran. In this study ten varieties of rice bran, which are commonly cultivated varieties in Indonesia were studied their antioxidant activity and total phenolic content. The antioxidant activity was examined by DPPH scavenging assay and total reducing power, while total phenolic content was determined by Folin-Ciocalteu reagent. The results of this study showed that the methanolic extract of IR-64 variety demonstrated the strongest DPPH radical scavenger activity with IC50 350.64 ppm, while in reducing power activity was exhibited by methanolic extract of IR-42 variety. However, the highest total phenolic content (71.85 mg gallic acid equivalent/gram) was found in methanolic extract of OM-4495 rice bran. These result indicated that the methanolic extract of rice bran might potentially be natural antioxidants.

Abstract

Cordia sebestena L. (Boraginaceae) atau Cordia merupakan tanaman hias dengan kandungan flavonoid. Penelitian ini bertujuan menganalisis senyawa flavonoid pada fraksi hasil hidrolisat ekstrak etanol 70% daun Cordia. Penelitian terbagi menjadi 7 tahapan, yaitu (1) maserasi daun menggunakan etanol 70%, (2) hidrolisis asam pada ekstrak dengan HCl 2N pada 95 °C selama 2 jam, (3) fraksinasi hidrolisat dengan etil asetat, (4) penentuan kadar flavonoid total dengan kolorimetri, (5) pengujian aktivitas antiradikal terhadap 2,2’-difenil-1-pikrilhidrazil (DPPH), (6) pemisahan flavonoid menggunakan kromatografi lapis tipis (KLT), dan (7) identifikasi flavonoid menggunakan spektrofotometri UV-Vis dan infra merah. Hasil menunjukan kadar flavonoid total fraksi etil asetat sebesar 8,20 mgQE/g dengan nilai IC50 terhadap radikal DPPH sebesar 296,91 µg/mL. Kromatogram KLT menunjukkan keberadaan satu bercak senyawa pada fraksi etil asetat dengan nilai hRf adalah 85,5. Hasil spektrum UV-Vis pada bercak tersebut diperoleh serapan pada 340 nm (pita I) dan 240 nm (pita II). Sedangkan spektrum infra merah menunjukan adanya sinyal pada 3363 cm-1 (O-H ulur), 1727 cm-1 (C=O), 1459 cm-1 (cincin aromatik) dan 1060 cm-1 kuat (C-O). Dengan demikian, dapat disimpulkan bahwa fraksi etil asetat hidrolisat ekstrak etanol daun Cordia mengandung senyawa flavonoid yang mirip dengan kelompok kalkon.

Abstract

Tiga spesies dari genus Syzygium, seperti jambu air (Syzygium aqueum (Burm.f.) Alston), jambu bol (Syzygium malaccense (L.) Merr. & L.M. Perry), dan jambu semarang (Syzygium samarangense (Blume) Merr. & L. M. Perry), telah dimanfaatkan daunnya dalam pengobatan tradisional karena kandungan polifenolnya. Penelitian ini bertujuan untuk mempelajari anatomi jaringan, mikroskopis, kadar senyawa fenol, flavonoid, dan tanin ekstrak etanol daun dari tiga jenis jambu tersebut. Daun jambu biji (Psidium guajava) digunakan sebagai pembanding. Anatomi jaringan daun dan identifikasi mikroskopis dilakukan menggunakan mikroskop. Penentuan kadar senyawa pada ekstrak etanol 70% ditentukan dengan metode kolorimetri yang absorbansinya diukur pada spektrofotometer UV- Vis. Anatomi jaringan daun dan fragmen pengenal secara mikroskopis ada kemiripan pada ketiga jenis daun jambu genus Syzygium. Secara statistik, ada perbedaan signifikan antara kadar fenol, flavonoid, dan tanin pada ekstrak etanol 70% daun jambu air, daun jambu bol, dan daun jambu semarang dibandingkan dengan ekstrak etanol 70% daun jambu biji. Daun jambu air merupakan spesiesterpilih dari genus Syzygium yang diuji pada penelitian ini dengan kandungan polifenol yang tinggi. Leaves of three guava species of the genus Syzygium, such as water apple/jambu air (Syzygium aqueum (Burm.f.) Alston), malay apple/jambu bol (Syzygium malaccense (L.) Merr. & L.M. Perry), and wax apple/jambu Semarang (Syzygium samarangense (Blume) Merr. & L. M. Perry), have been used in traditional medicine because of its polyphenol content. This study aims to investigate the anatomy of leaf tissues, powder microscopical characteristic, and total phenolic, flavonoids, and tannins content in the ethanol extract of the leaves of the three types of selected guava species of the genus Syzygium. Guava leaves (Psidium guajava) were used as a comparison. Leaf tissue anatomy and microscopic identification were carried out using a microscope. Determination of the polyphenol content in 70% ethanol extract was determined by the colorimetric method in which the absorbance was measured on a UV-Vis spectrophotometer. Leaf tissue anatomy and microscopic recognition fragments are similar in the three types of Syzygium guava leaves. Statistically, there were significant differences between the total phenols, flavonoids, and tannins content in the 70% ethanol extract of water apple, Malay apple, and wax apple compared to the 70% ethanol extract of guava leaves. Water apple leaves are selectedspecies from the genus Syzygium tested in this study with high polyphenol content.

Abstract

Piper retrofractum Vahl. (java chili or cabe jawa) and Piper nigrum L. (black pepper or lada hitam) are parts of the Piperaceae. One of the chemical constituents found in the fruit of the two plants is piperine, which has properties as antimicrobial, antidiabetic, anti-inflammatory, anticancer etc. This study aims to determine the piperine levels contained in 95% ethanol extract of java chili and black pepper fruits from regions with different altitude using UV-Visible spectrophotometer. Each sample were extracted by maceration method using 95% ethanol. Qualitative identification of piperine was carried out using thin layer chromatography (TLC). Determination of piperine levels was carried out with UV-Visible spectrophotometer at maximum wavelength of 253.8 nm. The results showed that the levels of piperine contained in the 95% ethanol extract of java chili fruit from low altitude (Lampung), medium altitude (Madura) and high altitude (Bogor) are 1.54±0.02%; 1.44±0.02%; and 1.41±0.02% (w/v), respectively. Whereas the levels of piperine contained in the 95% ethanol extract of black pepper fruit from low altitude (East Luwu), medium altitude (Central Lampung) and high altitude (Bogor) are 5.63±0.14%; 5.38±0.05%; and 4.87±0.09% (w/v), respectively.
