Abstract

 ABSTRACT Phytochemical study and antiacetylcholinesterase assay by using bioautography method were carried out on polar fraction of tabat barito (Ficus deltoidea jack) leaves. The result of this studies showed that the leaves contained flavonoid, terpenoid, steroid and fenolic compounds. This extract gave positive result on antiacetylcholinesterase. Keywords: Antiacetylcholinesterase, Ficus deltoidea Jack

Abstract

Generally, Gluta renghas L. is known because of its very toxic latex which can cause hard irritations to skin. Although, rengas latex has efficacy as an antibacterial agent. Related to previous research, it has been reported that there are urisol, rengol, glutarengol, laccol, and thitsiol in rengas latex. The woody trunk was reported to contain flavonoid, benzenoid, lipid, and steroid compounds. The leaves, were reported shows that the leaves of G. renghas contain an anticholinesterase substance and can be reduced the rate of hydrolysis of acetylcholine. Extraction of rengas leaves has been carried out. It has been shown phenols, steroids, and coumarins compounds in methanol extract, flavonoids, phenols, saponins and alkaloids compounds in ethyl acetate extract, steroids and alkaloids compounds in hexane extract. In the present, cytotoxicity and antibacterial activity have been tested. The results show that the great cytotoxicity activity by ethyl acetate extracts which have an LC50 value is 123,718 µg/mL (R2 0.9822), while the great antibacterial activity shown by methanol extract 1,000 µg/mL with a diameter of inhibition zone 19.02 mm (S. aureus) and 16.06 mm (E. coli).

Abstract

Lantana camara Linn is a family of Verbenaceae which grows wild and is widespread in various both tropical and sub-tropical countries. Isolation essential oil of the L. camara leaves extracted by hydrodistillation and were analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). The results of GC-MS analysis show isocaryophyllene (14.39%), ρ-cymene (8.17 %), β-cubebene (7.8 %), α-pinene (7.64 %), and β-elemene (5.51 %) as the main compound. The cytotoxic activity of the isolated essential oil was highly toxic both to Artemia salina shrimp larvae (LC50 15.92 µg/mL) and to breast cancer cells T-47D (IC50 10.67 µM).

Abstract

Dates are one of the plants that grow in the Middle East, one of them is the Golden Valley variety. Dates are usually consumed directly and can also be processed into date juice drinks. Dates refined process would produce waste in the form of date seeds are discarded and not processed into other products. This study used lipid-free Golden valley date seed extract to determine the secondary metabolite profile by using LC-MS/MS method, determining phenolic content using the Folin-Ciocalteau method, determining total flavonoids using the aluminium chloride method, determining antioxidant activity by DPPH method, and determination of antibacterial activity by the disc diffusion method. The results showed that the compounds identified from the lipid-free date seed extract were catechin-(4α→8)-catechin, bis((1S,2R,5S)-5-isopropyl-2,3-dimethylcyclo-hexyl)-2,6-dimethylpyridine-3,5-dicarboxylate, 3,5,6-trihydroxy-4',7-dimethoxyflavone, ethyl-2-amino-3-oxobutanoate, and bis(2-isopropyl-5-methylcyclohexyl)-2,6-dimethyl-1,4-dihydro-3,5-pyridine dicarboxylate. Lipid-free date seed extract contains total phenolic and flavonoid totals of 4364.704 mgGAE/100 grams of dried extract and 17200 mgQE/100 grams of dried extract. Its antioxidant activity is very strong in counteracting DPPH free radicals indicated by IC50 value of 10.1951 mg/L. Antibacterial activity shows strong inhibition of Staphylococcus aureus bacteria compared to inhibition of Escherichia coli bacteria that are categorized as weak.

Abstract

A triterpenoid compound has been isolated from the ethyl acetate fraction of Lantana camaraLinn leaves. The isolation was performed by chromatography of gravitation column with a gradient elution system using hexane, ethyl acetate and methanol. Purification by recrystallization produces a white solid (20 mg) with a melting point of 288-289 °C. The results of elucidation of UV, IR, 1H-NMR, 13C-NMR, HMBC, HSQC, DEPT and COSY spectral data, it is known that the isolated triterpenoid compound is 22β-angeloyloxy-9-hydroxy-3-oxo-olean-12- en-28-oicacid with molecular formula C35H52O6. Keywords—Lantana camaraLinn, Chromatography of gravitation column, Triterpenoid, C35H52O6

Abstract

This research aimed to isolate chemical constituents from ethyl acetate and n-hexane fractions of Ficus aurata (Miq.) fruits and investigated their antibacterial activity. Chemical constituents were separated using vacuum liquid chromatography, gravity column chromatography, and thin-layer chromatography. The structures of the isolated compounds 3,4-dihydroxybenzoic acid (1) and β-sitosterol (2) were elucidated using various spectroscopic methods, including UV, IR, and NMR (1H-NMR, 13C-NMR, and DEPT). While, antibacterial activity was done using resazurin microtiter assay. These 2 compounds were first time reported from F. aurata fruit. In addition, analysis of the bioassay showed that compound 1 had a notable antibacterial activity against Gram negative (Escherichia coli ATCC 25922 and Pseudomonas aeruginosa ATTC 27853), and a gram-positive bacterium (Staphylococcus aureus ATCC 23235) with the minimum inhibitory concentration (MIC) value of 0.1563 µg/ml, and it indicated the same MIC as cefadroxil (positive control). Compound 2 has weak antibacterial activity compared to cefadroxil (positive control). The results found that F. aurata (Miq.) fruits metabolites have antibacterial activity and show potency as antibacterial agents. Different kinds of bacteria and assay could be needed to investigate and support its antibacterial activity in the future.

Abstract

Sambuang (Etlingera elatior) is a member of the Zingiberaceae family which is widely distributed in Indonesia. Traditionally, sambuang is used for cooking spices and to cure various diseases, namely diseases of the ears, stomach, skin wounds, and stomach pain due to poisoning. In this study, essential oils were isolated from sambuang leaves and antibacterial and antioxidant activities were also determined. Isolation of essential oil was carried out by steam distillation and the chemical content of the isolated essential oil was analyzed by Gas Chromatography-Mass Spectrometry (GC-MS). It is known to contain 98 chemical compounds, with the main ingredients being a-Pinene (19.39%), Gadoleyl Alcohol (7.37), Myristoleyl Alcohol (7.16%), Palmitoleyl Alcohol (5.87%), Sabinene (5 ,48%). The results of the antibacterial activity of essential oils using the disc diffusion method showed strong activity against Escherichia coli, Salmonella typhi (50% concentration) and Staphylococcus aureus (70% concentration) with inhibition zones of 13.6 mm, 12 mm, respectively. and 11 mm. The results of the antioxidant activity test using the DPPH method (1,1-diphenyl-2-picrylhydrazyl) showed that the essential oil of sambuang leaf essential oil was not effective as an antioxidant because it was very weak with an IC50 value of 6939.18 g/mL.

Abstract

Caraway (Carum carvi L.) is one of the oldest medicinal plants found in various countries such as West Asia, North Africa, Pakistan, India, North America, Northern Europe, Iran, Russia, and Indonesia. This plant has been use for traditional medicines to treat digestive disorders, flatulence, colic in infants, headaches, coughs, hypertension, eczema, pneumonia, diabetes, scabies, mouthwash and as an antiseptic. This plant is reported to contain around 1-9% essential oil in the seeds with various components and potential as bioactive compounds. In this study, essential oils were isolated from caraway seeds and determined their chemical content as well as their antimicrobial and toxicity. Isolation of essential oils was carried out by hydrodistillation and analysis of the chemical components of the isolated essential oils was carried out using the Gas Chromatography-Mass Spectrometry (GC-MS) method. The results of the GC-MS analysis showed the presence of 35 compounds with four main compounds, namely carvone (62.54%), limonene (23.39%), and trans-dihydrocarvone (8.20%). The antimicrobial activity test was carried out using the disc diffusion method, which showed strong activity against Staphylococcus aureus at a concentration of ≥75%, and against the fungus Candida albicans. The results of the toxicity test using the Brine Shrimp Lethality Test (BSLT) method showed that caraway seed essential oil was low toxic against Artemia salina Leach shrimp larvae with an LC50 value 568.3292 mg/L.

Abstract

Pucuk merah (Syzygium myrtifolium Walp.) is an ornamental plant from the Myrtaceae family. Several phytochemical tests from previous researchers reported that pucuk merah contains secondary metabolites that have benefits such as antioxidants, antibacterials, and anti-cancer. In addition, the distinctive aroma produced from pucuk merah  leaves indicates the presence of essential oils whose bioactivity can be studied. Still, there has been no report on this plant's chemical components or bioactivity. Therefore, this study aims to determine the content of chemical components of pucuk merah  essential oil and their bioactivity as antibacterial and toxicity. The isolation of essential oils was carried out by the hydro distillation method. The chemical components of essential oils were analyzed using the Gas Chromatography-Mass Spectrometry (GC-MS) method. GC-MS results showed the presence of 42 compounds with four main compounds, namely caryophyllene (23.45%), 3-carene (15.67%), α-terpineol (10.74%), and α-pinene (5.98%) ). Antibacterial test of pucuk merah  essential oil using the disc diffusion method showed strong activity against Staphylococcus aureus and Escherichia coli with inhibition zones of 16.15 ± 2.03 and 16.13 ± 1.74 mm at 100% concentration. The results of the toxicity test of pucuk merah  essential oil using the BSLT (Brine Shrimp Lethality Test) method on Artemia salina Leach shrimp larvae showed strong toxic properties with an LC50 value of 3.99 µg/mL.
