Abstract

Lais (Krypyoterus apogon) is highly economic important catfish in South Sumatera. Investigating fish diversity of catfish is of importance for species conservation. Many species or subspecies extincted due to environment decreasing caused by habitat loss, pollution and over fishing. Cytochrome C Oxidase subunit I (COI) is one of mitochondrial DNA markers used for species barcoding in freshwater, brackishwater and marine fish. This research aim to explore the use of COI gene for species barcoding in investigating the diversity of COI nucleotide, constructing phylogenetic tree of lais, and knowing physical and chemistry characteristic of habitat at Musi and Kelekar River, South Sumatera. The methods used in the research consisted of DNA  extraction, PCR (Polymerase Chain Reaction) amplification and sequencing mtDNA COI gene of Kryptopterus apogon obtained from Musi and Kelekar River (South Sumatera). A 615 bp (sample code CM1) and 693 bp sequences (sample code CK1 and CK2) of parsial coding were sequenced from this fish. Nucleotide BLAST analyzes showed COI gen can distinguish lais at Musi and Kelekar River from others fish at GenBank database with high identity (92.29% and 92.21%) to Phalacronotus bleekeri (Thailand) and Phalacronotus apogon (Australia).Phylogenetic analyzes indicated that obviously, lais was at the same cluster with others genus in Siluridae family, but different cluster from Hemibagrus nemurus (Bagridae), Chanidae and Oreochomis niloticus (Cichlidae). Water quality criteria (water tranparancy, temperature, ammonia, pH and alkalinity) at Musi and Kelekar River were still in tolerance for maintainning survival rate of lais. Further study using more species in Siluridae family and various habitat are needed to investigate the diversity of DNA of Siluridae at South Sumatera water resources.Keywords: Catfish, DNA barcode, COI gene, Musi River, Kelekar River

Abstract

Permasalahan yang sering dialami oleh pembudidaya ikan termasuk yang dihadapi oleh kelompok pembudidaya ikan di Desa Pandan Arang saat ini belum mampu memproduksi benih ikan lele secara mandiri sehingga ketersediaan benih untuk usaha pembesaran budidaya ikan lele masih mengandalkan dari daerah lain. Tujuan pengabdian ini dilakukan untuk memberikan edukasi pada masyarakat, sehingga bisa diterapkan untuk membantu meningkatkan produksi benih ikan lele secara mandiri dan berkelanjutan. Kegiatan dilaksanakan di Desa Pandan Arang, Kecamatan Kandis Kabupaeten Ogan Ilir, Sumatera Selatan. Waktu pelaksanaan sosialisasi hingga pendampingan produksi pada bulan Oktober-Desember tahun 2020 dan dilanjutkan monitoring evaluasi pasca kegiatan pada tahun 2021. Demplot dilakukan pada satu indukan ikan lele yang menjadi percontohan sekaligus Praktik Lapangan mahasiswa program studi budidaya perairan. Hasil kegiatan menunjukan bahwa, tingkat serapan pengetahuan peserta berdasarkan indikator yaitu 60-80%, dari tidak tahu menjadi tahu tentang pemijahan semi alami ikan lele. Tingkat keterampilan peserta berdasarkan indikator 75-100 %, dari tidak bisa menjadi bisa membenihkan ikan lele

Abstract

AbstrakPermintaan komoditi perikanan di pasar internasional maupun domestik terus mengalami perubahan mulai dari produk dalam bentuk baku ke bentuk segar dan dalam bentuk hidup. Beberapa komoditas perikanan yang memiliki nilai ekonomis tinggi dan banyak diperdagangkan dalam keadaan hidup antara lain ikan nila, udang, lobster dan beberapa jenis ikan laut. Salah satu alasan permintaan konsumen terhadap komoditi hidup perikanan adalah keinginan konsumen untuk memperoleh kepuasan cita rasa dan tekstur daging yang lebih baik. Salah satu metode yang dapat dilakukan dalam upaya menurunkan aktifitas metabolisme ikan selama pengiriman adalah dengan pembiusan.tujuan dari penelitian ini adalah menguji toksisitas konsentrasi ekstrak akar tuba pada ikan nila merah.Mengetahui nilai LC50 96 jam pada ekstrak akar tuba pada ikan nila merah (Oreocromis sp). Memberikan informasi konsentrasi yang aman untuk penggunaan ekstrak akar tuba dalam menurunkan aktifitas metabolisme pada ikan nila merah (Oreocromis sp). Penelitian ini dilaksanakan selama 6 hari pada bulan Januari 2015. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL), terdiri 6 taraf perlakuan dengan 3 kali ulangan. Hasil penelitian ini menunjukkan bahwa semakin tinggi konsentrasi ekstrak akar tuba semakin banyak ikan uji yang mati dengan kematian hingga (77,1%) sudah dapat dicapai pada konsentrasi perlakuan P5 0,0036 ml/L dan kematian terendah (6,27%) terjadi pada perlakuan P1 (0,0013 ml/L) sedangkan pada konsentrasi (0 ml/L) tidak terjadi kematian. Sedangkan untuk kelangsungan hidup benih Ikan Nila Merah (Oreochromis sp) yang tertinggi yaitu pada perlakuan P0dengan tingkat SR sebesar 100% serta terendah pada perlakuan P5 dengan tingkat SR sebesar 22,9%. Sedangkan Nilai tingkat kelangsungan hidup pada perlakuan P1 sebesar 93,73 %, diikuti perlakuan P2 (86,3 %) , perlakuan P3  (85,8 %) serta  perlakuan P4  (62,9%). Nilai kualitas air tiap perlakuan yaitu suhu berkisar antara 25-260C, dan pH 6,0-7,0. Kata Kunci : Ikan Nila Merah, Ekstrak Akar Tuba, Nilai LC50 96 Jam, Kelangsungan Hidup, Mortalitas.

To overcome this, digital marketing was held, the Fishket application was a solution in marketing and selling crops, where this application can be accessed directly via a Smartphone. This application aims to market online aquaculture products, namely seeds, consumption fish, broodstock and various kinds of ornamental fish. This application is designed using hardware and software where in this application several features have been provided that can make it easier for users to buy the fish they want with a very easy payment method, you can use via transfer or direct payment on the spot (delivery order). This application provides features that facilitate the buying and selling process, such as product image features, product prices, product information and payment methods. The application of the application can be accessed anywhere with coverage throughout South Sumatra. So, with the Fishket application, farmers can determine the selling price of fish without a third party. This application is expected to increase sales and income of farmers.

Palembang: Penerbit & Percetakan Universitas Sriwijaya (UNSRI).  The high demand for fishery products has resulted in cultivators implementing intensive and even super intensive cultivation systems. The intensification of fish farming is characterized by an increase in stocking density followed by an increase in the use of artificial feed which has the potential to become waste and cause disease. To overcome this, alternative actions that are safer and also effective for controlling fish diseases are needed. One alternative is to use herbal plants that are anti-bacterial. By using herbal plants, it will be safer, easier to obtain and also not pose a danger to the surrounding environment. This study aimed to determine the effectiveness of lemongrass stem extract to treat bacterial disease in fish caused by Edwarsiella tarda. Some research results showed that Cymbopogon citratus extract is rich in benefits in herbal medicine. Cymbopogon citratus extract had antibacterial properties that could inhibit the growth of pathogenic bacteria in fish. Phytochemical test results, lemongrass extract contained secondary metabolites such as alkaloids, steroids, terpenoids, phenolics or tannins or flavonoids and saponins. Based on the results of in vitro tests on lemongrass stem extract, it was able to inhibit the growth of Edwarsiella tarda bacteria where the inhibition zone formed after the media was incubated for 24 hours with a size of 21 mm. This proved that lemongrass stem extract can inhibit the growth of Edwarsiella tarda bacteria based on the results of the inhibition test.

Abstract

Minyak cengkeh telah banyak digunakan oleh pembudidaya sebagai anestesi pada transportasi ikan. Akan tetapi penggunaannya pada beberapa jenis ikan dengan kepadatan yang berbeda menunjukkan kebutuhan dosis penggunaan yang berbeda. Tujuan penelitian ini adalah untuk menentukan dosis minyak cengkeh sebagai bahan pembius ikan tambakan (Helostoma temminckii) dan untuk mengetahui pengaruh pemberian minyak cengkeh pada transportasi ikan tambakan. Penelitian ini membandingkan kondisi transportasi yang menggunakan minyak cengkeh dan tanpa minyak cengkeh (kontrol) dengan kepadatan berbeda. Kepadatan yang digunakan adalah 10, 12, dan 14 ekor L-1. Ikan tambakan yang digunakan berukuran 15 ± 0,5 cm. Penelitian ini dibagi menjadi dua tahap yang terdiri dari penentuan konsentrasi efektif-100 10 menit (EC100 10 min) dan pengaruh minyak cengkeh terhadap kepadatan selama 12 jam transportasi. Hasil penelitian menunjukkan bahwa nilai EC100 10 min adalah 0,026 mL L-1. Sesaat setelah transportasi, tingkat kelangsungan hidup dan tingkat konsumsi oksigen tidak berbeda nyata antara perlakuan dan kontrol pada kepadatan yang berbeda. Akan tetapi kadar glukosa darah pada kepadatan 10 ekor L-1 dan 12 ekor L-1 lebih rendah daripada kontrol. Pemantauan kesehatan ikan setelah 7 hari pascatransportasi menunjukkan bahwa tidak ada perbedaan nyata antara kontrol dan perlakuan baik kelangsungan hidup maupun kadar glukosa darah. Oleh karena itu, pemanfaatan minyak cengkeh pada dosis 0,026 mL L-1 untuk transportasi ikan tambakan dapat diaplikasikan dengan kondisi kepadatan 14 ekor L-1 selama 12 jam. Pemanfaatan minyak cengkeh pada ikan tambakan perlu dilakukan kajian lebih lanjut terkait penambahan kepadatan dan waktu transportasi atau aplikasinya untuk transportasi benih ikan tambakan.Clove oil has been widely used by farmers as an anesthetic agent in fish transportation. However, its use on several fish with different densities indicates the need for different doses. The aim of this study was to determine the dose of clove oil as an anesthetic agent for kissing gourami (Helostoma temminckii) and to determine the effect of administering clove oil on the transportation of kissing gourami. This study compared transportation conditions using clove oil and without clove oil (control) with different densities. The densities used were 10, 12, and 14 L-1. The kissing gourami used measured 15 ± 0.5 cm. This study was divided into two stages consisting of determining the effective concentration-100 10 minutes (EC100 10 min) and the effect of clove oil on density during 12 hours of transportation. The results showed that EC100 10 min value was 0.026 mL L-1. Immediately after transportation, survival rates and oxygen consumption levels were not significantly different between treatments and control at different densities. However, blood glucose levels of 10 fish L-1 and 12 fish L-1 were lower than the control. Fish health status monitoring after 7 days post-transportation showed that there was no significant difference between control and treatment in terms of survival rate or blood glucose levels. Therefore, the use of clove oil at a dose of 0.026 mL L-1 for the transportation of kissing gourami can be applied at a density of 14 fish L-1 for 12 hours. Further studies are required to determine the effects of clove oil as an anesthetic agent applied at denser stocking densities and longer transportation period of kissing gourami seeds.
