Abstract

Celery (Apium graveolens L.) is commonly used to lower blood pressure, antirheumatic, relaxant, mild diuretic, antiseptic for the urinary tract, antioxidant, and anti-inflammation. According to previous studies, a number of the phytochemicals found in the plant show cytotoxicity toward some types of cancer cells. However, studies on the cytotoxic effects of celery herb ethanolic extract (CEE) on breast cancer cell (T47D), colon cancer cell (WiDr), and cervix cancer cell (HeLa), however, has not been done yet. Our research aims at doing so. Cytotoxicity test was conducted using MTT assay and its absorbance was read using ELISA reader at λ = 595 nm. Results of the assay show that CEE reduces cell viability at concentrations of 100-750 µg/ml on HeLa cells, while reduction of T47D and WiDr cell viability was not achieved until concentrations of 500-750 µg/ml. Based on these results, we conclude that CEE hold many potentials for further developments as preventive and therapeutive agent in cancer treatment.

Abstract

Schoutenia ovata Korth. atau Walikukun merupakan tumbuhan yang banyak ditemukan di Indonesia. Kayu dari tumbuhan ini digunakan oleh masyarakat sebagai gagang perkakas dan juga sebagai bahan kerajinan tangan. Namun demikian, hingga saat ini, penelitian mengenai potensi farmakologis Walikukun belum dieksplorasi lebih jauh. Oleh karena itu, pada penelitian kali ini dilakukan skrining aktivitas antibakteri dan antioksidan dari daun dan batang Walikukun yang dikoleksi dari Mandalika, Nusa Tenggara Barat. Simplisia kering dari daun dan batang Walikukun diekstraksi secara bertingkat dengan pelarut n-heksana, diklorometan, etil asetat dan metanol. Analisis profil kimia dari daun dan batang Walikukun dilakukan dengan metode kromatografi lapis tipis. Uji aktivitas antibakteri dilakukan terhadap Staphylococcus aureus dan Escherichia coli sedangkan uji aktivitas antioksidan dilakukan terhadap radikal bebas 1,1-diphenyl-2-picrylhydrazyl (DPPH). Penentuan kadar hambat minimum (KHM) antibakteri serta penentuan indeks aktivitas antioksidan (IAA) dilakukan dengan metode mikrodilusi. Hasil pengamatan menunjukkan bahwa ekstrak daun dan batang dari Walikukun menunjukkan aktivitas antibakteri yang lemah hingga moderat dengan nilai KHM terendah sebesar 128 µg/ml. Ekstrak batang Walikukun menunjukkan aktivitas antioksidan yang potensial. Ekstrak metanol batang Walikukun menunjukkan aktivitas antioksidan terbaik dengan nilai IAA 8.275 ± 0.012 (aktivitas antioksidan sangat kuat). Hasil penelitian ini menunjukkan bahwa batang Walikukun merupakan sumber senyawa antioksidan yang potensial.Kata kunci: Schoutenia ovata Korth., Staphylococcus aureus, Escherichia coli, DPPH

Abstract

Endophytic fungi are fungi that live in plant tissues that do not cause disease symptoms, do not harm the host plant, and can produce secondary metabolites such as antibacterial, antifungal, and antiviral compounds. This study was conducted to isolate and evaluate the antibacterial and antioxidant activities of endophytic fungi from Aloe vera (L.) Burm.f. against Staphylococcus aureus and Escherichia coli bacteria. The antibacterial activity test was carried out qualitatively by thin layer chromatography-direct-bioautography (TLC-DB), and microdilution methods were used for the antibacterial assay against Staphylococcus aureus and Escherichia coli. Antioxidant activity was determined against DPPH using TLC-DB and the microdilution assay. Antibacterial results showed that the extracts LBB-2.4, LBB-2.2, LBAu-1.1 and LBAu-2.1 could inhibit the growth of S. aureus. Meanwhile, extracts of endophytic fungi with the isolate codes LBB-2.4, LBB-2.1, LBAp-3, LBAp-1, LBAu-1.1 and LBAu-2.1 could inhibit the growth of E.coli. The results of Minimum Inhibitory Concentration (MIC) showed that isolate LBB-2.1 had moderate antibacterial activity with a MIC value of 256 μg/mL against S.aureus. The extracts showed weak antioxidant activity against DPPH. It can be concluded that endophytic fungi extracts from Aloe vera have potential activity as antibacterial agents.
