Abstract

We have successfully isolated and identified endophytic fungi from medicinal plant Mesua ferrea L. grown at Banyumas were for the first time. One of those fungi was MFD-01. In this study, we report the isolation, identification, antibacterial activity, and phytochemical screening of Aspergillus sp. MFD-01. The endophytic fungus was isolated from the leaves of M. ferrea by subsequent inoculation on potato dextrose agar (PDA). The identification was based on its morphology and ITS-DNA sequence. The antibacterial activity was determined by dilution method. The identification of compounds in ethyl acetate extract of it was conducted according to the standart phytochemical screening method. MFD-01 was identified as Aspergillus sp. MFD-01. The ethyl acetate extract of Aspergillus sp. MFD-01 inhibited the growth of both Staphylococcus aureus, and Escherichia coli. At concentration of 1000 ppm, its diameter of inhibitory zone against those bacteria was 10.53 and 11.84, respectively. The extract contained flavonoids, alkaloids, saponins, terpenoids, and tannins, which might responsible for its antibacterial activity against both tested bacteria.

Abstract

An ethnopharmacological study is an initial step for the development of jamu, which has been simultaneously conducted throughout Indonesia. This study aimed to explore the utilization of medicinal plants in Baturraden, a region with relatively rich plant biodiversity in Java. Our research is a quantitative ethnobotanical study reporting the utilization of medicinal plants as a single plant botanical preparation in the studied area. The data were collected through semi-structured interviews with 36 informants on the local names, indications, parts used, method of preparation, and application of the plants. The species use-value (SUV), relative frequency citation (RFC), and the fidelity level (FL) of each species were calculated accordingly. The study recorded the utilization of 47 species of medicinal plants, which were employed to treat 32 ailments. Leaves, decoctions, and oral were recorded as the most commonly used plant part, herbal preparation, and application. There were 24 botanical preparations with the FL value of 100%, which were indicated for the treatment of fourteen diseases/symptoms. Curcuma longa, Zingiber officinale, Piper betle, Cymbopogon citratus, Andrographis paniculata, Syzygium polyanthum, Citrus aurantiaca, and Zingiber montanum were considered as the most important and valuable plants by the local community. Hence, those plants should be evaluated further for their pharmacological activity and developed into standardized botanical preparations.

Abstract

Galangal (Alpinia galanga (L.) Willd.) has been long used for medicinal purposes. It showed promising effects on the food-spoilage microorganisms and is commonly used as spices in various cuisines. This study aimed to evaluate the microbial growth inhibitory activity and the preservation potential of galangal water extracts on tofu. The water extracts were prepared by infundation method in the concentrations of 5, 10, and 20%w/v. The phytochemical compounds in the extracts were screened as per standard methods. The tofu was preserved by immersion method for eight days under room temperature with an evaluation time point of two days. The microbial growth inhibitory activity was evaluated by indirect optical density (OD)-based enumeration, and the OD reduction by the extracts was calculated accordingly. The preservation potential was calculated from the changes of the physical characters on tofu evaluated by sensorial analysis. Galangal water extracts contained tannins and terpenoids. It concentration- dependently inhibited microbial growth on tofu, and the extract showed the best OD reduction at a concentration of 20%. It changed the color of tofu but maintained its texture. It also masked the odor of fresh tofu with the aromatic galangal scent. All three tested concentrations showed an equal preservation potential of 2 days. Our data suggested galangal water extract in the optimum concentration of 20% inhibited the microbial growth on tofu and potentially preserved it for two days. This moderate preservation effect might be attributable to its tannins and terpenoids content.

Abstract

Cattle manure contains high organic matter, so that this material is commonly used as plant nutrients in a form of organic fertilizer. In order to get a better understanding on the use of cow manure, the objectives of this research are (1) to determine the influence of liquid waste and solid livestock as fertilizer on soil quality and (2) to study the effect of both liquid and solid fertilizer on coffee plant growth. In this research, Completely Randomized Design (CRD) was employed as a research design, while data analysis was conducted using ANOVA. The F statistical test was applied to measure whether all independent variables could influence the dependent variable by using 5% and 1% (Alpha) degrees. The results showed that one time application of livestock wastewater improved C-organic chemical properties, N, P, CEC, Ca and Mg while soil chemical properties of pH, K, Na decreased. Two times application of livestock liquid fertilizer treatment improved soil chemical properties pH, N total, Ca, Mg and saturation bases. As for solid waste, one time treatment increased chemical properties of pH, C organic, N total, Ca and saturation of the base. The two times treatment could improve soil chemical properties of pH, N. total, P, Ca, Mg, base number and base saturation. Liquid waste fertilizer also found to provide significant effect on plant height and coffee plants quality. Additionally, (3) solid waste (compost) was found to have a significant effect on plant height, plant height rate and number of buds.

Abstract

Anthraquinones are phenolic compounds known best for their laxative activity. Rheum australe D. Don (Indian rhubarb, Polygonaceae) is a prominent medicinal plant with anthraquinones as the bioactive compounds, among a few others. The online literature search was carried out to collect data on the phytochemistry and pharmacological activity of anthraquinones, particularly those isolated from R. australe. Anthraquinones are commonly found in the genera of Rheum, Senna, Aloe, Frangula, and Rubia, which can be quantitatively and qualitatively determined using both conventional and advanced analytical methods. The anthraquinones of R. australe were found in free and glycosidic forms, which were best extracted by the microwave-assisted extraction method. Various chromatographic techniques were commonly conducted to isolate the pure compounds. In addition to its laxative activity, anthraquinones of R. australe also showed potential antibacterial, cytotoxic, and antioxidant properties.

Abstract

Daun mindi (Melia azedarach L.), buah kersen (Muntingia calabura L.), serta daun dan bunga tapakdara (Catharanthus roseus (L.) G.Don) digunakan masyarakat Baturraden dan Sumbang (Banyumas) untuk pengobatan tradisional diabetes. Penelitian ini mengkarakterisasi beberapa parameter mutu simplisia dan ekstrak ketiga tumbuhan tersebut.  Simplisia dibuat dari bahan tumbuhan yang dikumpulkan dari tiga desa di wilayah kecamata Baturraden dan Sumbang. Morfologi makroskopik dan mikroskopik, profil kromatografi lapis tipis (KLT), kadar fenolik total, dan kadar flavonoid total simplisia, serta kadar air, kadar abu total, dan kadar abu tidak larut asam ekstrak dianalisis dengan metode standar dalam Farmakope Herbal Indonesia (FHI) 2017. Karakter makroskopik, fragmen diagnostik, dan profil KLT dari simplisia hasil penelitian ini diusulkan untuk menjadi standar dalam memastikan kebenaran identitas simplisia. Kadar fenolik total dan kadar flavonoid total simplisia daun mindi, buah kersen, serta daun dan bunga tapakdara masing-masing sebesar 0,74±0,07 dan 0,99±0,31; 1,62±0,36 dan 1,39±0,11; dan 0,94±0,02 dan 1,98±0,04%. Nilai tersebut diusulkan sebagai standar untuk aspek kandungan masing-masing simplisia. Data kadar air, kadar abu total, dan kadar abu tidak larut asam diusulkan sebagai standar aspek kemurnian dari masing-masing ekstrak.

Abstract

Jasmine tea is a popular tea type obtained from scenting tea (Camellia sinensis (L.) Kuntze) leaves with jasmine (Jasminum sambac (L.) Aiton) flowers. This study aimed to evaluate the antioxidant properties and sensory aspects of jasmine and green tea brewed at different times. The commercial jasmine and green tea were brewed in boiling water at a ratio of 1:100 for 0.5, 1, 2.5, 5, 10, 30, and 60 min. The obtained tea was evaluated for colour, pH, total flavonoid content (TFC), total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, and ferric-reducing antioxidant power (FRAP) by standard methods. Thirty-three untrained consumers evaluated the preference for tea from different brewing times. Jasmine tea exerted darker colour, lower pH, higher TFC and TPC, and higher FRAP than green tea. The values of those parameters increased with longer brewing time, with the optimum time being 30 min. However, the longer brewing generated tea liked less by consumers. The most favoured tea was jasmine green tea brewed for 5 min with aroma as the most preferred attribute. Our study suggested that brewing tea for a longer time benefited with better antioxidant properties but disadvantaged in sensory aspects.

Abstract

Penggunaan formalin sebagai bahan pengawet sangat berbahaya bagi tubuh manusia, maka perlu mencari alternatif pengawet alami pada makanan. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri campuran minyak atsiri cengkeh (MC) dan kitosan (K) terhadap Total Viable Colony (TVC). Pengawetan dilakukan dengan memasukkan potongan kecil daging ayam mentah ke dalam rendaman air yang berisi pengawet (MC&K) dengan komposisi tertentu (1250 µg/mL:3 mL, 6250 µg/mL:1 mL, 250 µg/mL:5 mL, 0 µg/mL: 3 mL, 1250 µg/mL:0 mL, 1250 µg/mL :3 mL:lecitin 0,5%, kontrol positif, formalin 10%) dan lama pengawetan selama 3, 6, 9, 12, 15 hari. Uji kandungan senyawa kimia minyak atsiri cengkeh dilakukan dengan Gas Chromatography-Mass Spectrometer (GC-MS) dan perhitungan TVC dengan metode turbidity menggunakan spektrofotometri. Hasil penelitian ini menunjukkan bahwa minyak atsiri cengkeh hasil penyulingan memiliki yield 1,47% dan senyawa dominanya yaitu eugenol 67%, Trans-β-Caryophyllene 25,64%, dan Humulene 2,66%. Penggunaan campuran pengawet (MC&K) memiliki aktivitas antibakteri yang signifikan berbeda (P<0,05) sehingga berpotensi untuk mengawetkan daging ayam mentah dan memperpanjang masa simpan daging ayam selama 6 hari (1250 µg/mL:3 mL, 6250 µg/mL:1 mL) dan 3 hari (1250 µg/mL:0 mL). Dari penelitian ini dapat disimpulkan bahwa pengawetan daging ayam mentah menggunakan campuran (MC&K) menghasilkan masa simpan yang lebih lama dibandingkan minyak atsiri cengkeh tunggal maupun kitosan.
