Abstract

Cisplatin is one of chemotherapy agent for long cancer, ovarium cancer, gastric cancer, breast cancer, head-neck cancer. However, in the fact, the role of cisplatin does not always provide an optimal effect because it often appears cancer cell resistance phenomenon to cisplatin. This resistance condition occurs partly due to the inactive metabolite cause of conjugation reaction between cisplatin and GSH in cancer cells. Therefore, gluthathione (GSH) has an important role in controlling cisplatin resistance. This study aims to analyze some combination of cisplatin and the depletion agent of gluthathione (GSH) as a support for cisplatin activity in several types of cancer cells within in vitro scope. This study is prepared using systematic literature review method. Library search were carried out on two accredited international journals databases, namely PubMed and Science Direct with interval years of publication in 2011-2020. From 10 selected journals, it was shown that the use of GSH depletion agents could enhance the cytotoxic effect of cisplatin. This was analyzed based on data of the number measured GSH cells and the number of living cells (% cell viability) which gave a significant decrease. The result of research are expected to be able to provide information for the development of therapeutic agents on cisplatin as chemotherapy.

Abstract

Demam tifoid merupakan salah satu penyakit yang menjangkit masyarakat Indonesia dengan 500 kasus per 100.000 penduduk dengan angka kematian mencapai 5%. Pengobatan utama demam tifoid adalah menggunakan berbagai macam antibiotik. Namun masyarakat Indonesia cenderung lebih memilih pengobatan alternatif atau tradisional. Salah satu obat tradisional demam tifoid adalah ekstrak cacing (Lumbricus rubellus) yang mempunyai kandungan protein sampai 71%. Kandungan protein sangat tergantung pada proses produksinya. Untuk itu diperlukan metode preparasi dan analisis untuk menentukan kadar protein yang bisa digunakan untuk menjamin mutu produk ekstrak cacing. Metode preparasi yang optimum  adalah dengan pemanasan pada temperatur 50°C selama 5 menit kemudian dianalisis kadar proteinnya dengan metode Lowry menggunakan spektrofotometer UV-Vis pada panjang gelombang 650,8 nm. Metode linier, akurat dan teliti dengan batas deteksi sebesar 0,18 mg/mL dan batas kuantifikasi sebesar 0,58 mg/mL. Kata Kunci: Protein, Cacing Tanah, Spektrofotometri, Metode Lowry  Typhoid fever is a disease that affects Indonesian people with 500 cases per 100,000 population with a mortality rate of 5%. The main treatment for typhoid fever is the use of various antibiotics. However, Indonesian people prefer to use alternative or traditional medicine. One of the traditional medicines for typhoid fever is earthworm (Lumbricus rubellus) extract which has a protein content of up to 71%. Protein content is highly dependent on the production process. For this reason, preparation and analysis methods are needed to determine protein levels that can be used to guarantee the quality of the worm extract products. The optimum preparation method is by heating at 50°C for 5 minutes and then the protein is analyzed using the Lowry's method using a UV-Vis spectrophotometer at wavelength of 650.8 nm. The method is linear, accurat and presice with  detection limit of 0.18 mg/mL and quantification limit of 0.58 mg/mL.

Abstract

Spirulina merupakan salah satu spesies cyanobacteria yang mulai dikembangkan dalam bidangkomersial dan pengobatan karena mengandung banyak zat bergizi seperti beberapa mineral danvitamin. Penelitian terdahulu menyebutkan bahwa spirulina mampu mengakumulasi logam-logamberat dari perairan yang tercemari logam berat seperti logam Pb dan Cd. Tujuan penelitian ini adalahmengetahui ada tidaknya cemaran logam Pb dan Cd dalam sediaan spirulina menggunakan metodespektroskopi serapan atom (SSA). Metode tersebut terlebih dahulu divalidasi meliputi parametersensitivitas, liniearitas, ripitabilitas dan akurasi. Sampel spirulina diambil dari dua macam produkdari dua produsen. Spirulina didestruksi menggunakan kombinasi HNO3 dan H2O2, kemudianselanjutnya cemaran logam dikuantifikasi dengan SSA pada λ 283,3 nm (logam Pb) dan λ 228,8 nm(logam Cd). Hasil validasi metode menunjukkan bahwa metode yang digunakan cukup peka, linier,dan teliti untuk kuantifikasi logam Pb dan Cd dalam spirulina. Hasil pengukuran dengan metodetersebut menunjukkan adanya cemaran logam Pb dan Cd dalam kedua produk spirulina dengankadar rata-rata logam Pb 2,642±0,559 μg/mL dan 4,253±1,922 μg /mL, sedangkan untuk logam Cd0,569±0,357 μg/mL dan 0,163±0,062 μg/mL.

Abstract

Earthworms are widely used by Indonesian people as an alternative treatment of typhoid fever in addition to the main treatment using antibiotics. One of the main ingredients of earthworms (Lumbricus rubellus) is protein. The amount of protein content of the extracts is highly dependent on the production process. For this reason, a spectrophotometric analysis was carried out to determine the protein content of the worm extracts in the market. As a result of the analysis using the Lowry method at a maximum wavelength of 650.8 nm, the protein content of a sample of earthworm extract obtained from the market was (10.41 ± 0.47) %.

Abstract

Hewan laut berupa teripang atau sea cucumber telah popular dimanfaatkan sebagai bahan pangan maupun bahan obat. Tujuan dari penelitian ini adalah mengevaluasi kemungkinan adanya cemaran logam berat dalam produk teripang. Sampel diambil dari tiga macam produk komersil yang berasal dari tiga industri berbeda. Sampel jamu dianalisis kandungan logam timbal (Pb) dan kadmium (Cd) secara spektrometri serapan atom dengan proses digesti basah menggunakan campuran asam nitrat dan asam perklorat. Hasil penelitian menunjukan rerata konsentrasi logam timbal pada tiga produk tersebut adalah 1,66; 4,47; dan 3,36 ppm. Rerata kadar logam kadmium sebesar 0,15; 0,473; dan 0,29 ppm. Konsentrasi logam cadmium masih di bawah dari batas maksimum yang diperbolehkan, namun untuk kadar logam timah relatif cukup tinggi, sudah melebihi batas maksimum yang diperbolehkan.

Abstract

Glutathione (GSH) is one of the targets to overcome chemotherapy resistance. Cancer cells adapt by increasing the production of GSH which acts as an antioxidant to reduce the reactive oxygen species (ROS)-inducing of chemotherapy agents. GSH can bind to chemotherapy agents that are electrophile resulting a conjugate of electrophile-GSH which is more polar so it is easily excreted out of the body. Butionine sulfoximine (BS) is a GSH inhibitor that decrease the concentration of GSH in cells. BS is a γ-glutamyl- cysteine ligase (GCL) enzyme inhibitor that catalyzes the first steps of GSH synthesis. This literature review aims to evaluate the use of BS to sensitize cancer cells against chemotherapy in vivo. The literature search was carried out of two databases (Pubmed and Sciendirect) resulting in 357 journals. Furthermore, a selection process was carried out which yielded in 10 journals. BS synergize with chemotherapy of alkylating groups, vincristine, geftinib, and cytarabine+doxorubicin and significantly increases the effectiveness of therapy compared to chemotherapy only. However, BS delay cancer in initiation phase, and not in tumor progression phase. In vivo, the combination of BS and chemotherapy of alkylating groups, vincristine, geftinib, and cytarabine+doxorubicin reduce the tumor volume, cancer cell proliferation and also increasing the survival of experimental animals.
