Abstract

Phytoestrogen is the chemical compound contains in plant which has estrogenic like effect. Estrogen has important function on woman’s sexual organ, such as proliferation of uterine and vaginal cornification. Phytoestrogen are weak agonists for estrogen and illicit statistic significantly increases in uterine wet weight, at definite dose, in uterothrophic bioassay. Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that suspected having an estrogen-likeeffect or as phytoestrogen. The aim of this research was to investigate the estrogenic effect of fenugreek’s ethanolic extract in immature rat models. These models represent the climacteric/menopause phase, where estrogen level is verylow because ovary produces no estrogen. The testing animals were divided into five groups, namely normal group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW). Theresult indicated that on 120mg/200gBW, fenugreek extract significantly (=0.05) increasing wet weight uterine and proliferation of uterine, such as uterine thickness and uterine diameters. Fenugreek extract, start on 60mg/200gBW, also induces vaginal lubrication and estrus cycle.

Abstract

Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that are suspected having an estrogen-like effect or as phytoestrogen. The objective of this study was to evaluate the estrogen-like effect of biji klabet alcohol extracts. This research used immature Wistar female rats, age of 19 days, which were represent the climacteric/menopause phase. The testing animals were divided into five groups, namely normal group, estradiol control group and three level dose groups of fenugreek extract (30 mg/200 gBW; 60 mg/200 gBW and 120 mg/200 gBW). The result showed that on 120 mg/200 gBW fenugreek extract significantly (a=0.05) increased estradiol and FSH level on immature rat. Analysis of vaginal smear showed vaginal lubrication effect, surprisingly on 120 mg/200 gBW could induce estrous cycle.

The testing animals were divided into normal group, ovariectomized control group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW). The result indicated that start on 60mg/200gBW, fenugreek extract significantly could increasing mammary gland proliferation. Empirically fenugreek containing diosgenin, that caused breast enhancement. This research showed that treatment with fenugreek extract can caused proliferation of mammary gland, both on imma-ture and ovariectomized rats.

Abstract

Renal dysfunction can be caused by several factors, including hypertension, urinary tract obstruction, autoimmune disorders, urinary tract infection, diabetes mellitus, consumption of drugs that affect nephrotoxic and antibiotic from aminoglycoside class. Breadfruit leaves (Artocarpus altilis (Park.) Fsb.) is a traditional plant that hasbeen used empirically to treat kidney diseases. The research has been done to figure out the nephroprotective effect of breadfruit leaves infusion on white male rats strain Sprague-Dawley previously induced by carbon tetrachloride. There were 25 rats which were divided randomly into five groups. Group I which was the normal control group received CMC 0,5%. Group II which was the carbon tetrachloride control group was induced with carbon tetrachloride that was dissolved in the coconut oil 0,4 mL/kg bw rat. Group III, IV and V were administered doses of infusion 13,5 g/kg bw/day; 27 g/kg bw/day and 54 g/kg bw/day for 7 days. Two hours later, the animals were given carbon tetrachloride induction. At the 8th day, the blood was collected from the orbital sinus and then the rats were performed a surgery to collect the kidney. The urea and creatinine plasma level measurement has been done by colorimetric method and histology of kidney was observed. One way ANOVA (α=0,1) of the study showed that nephroprotective effect of breadfruit leaves infusion were significantly different among groups and dose of 54 g/kg bw/day has the best nephroprotective effect.

Abstract

Estrogenic Effect of 70% Ethanol Turmeric (Curcuma domestica Val.) extract on ovariectomized Female Mice (Mus musculus L.). The influence of extract turmeric (Curcuma domestica Val.) on endometrium thickness, vaginal epithelium, mammary gland, and protein of estrogen receptor of ovariectomized mice was examined. Twenty five ovariectomized mice which were divided into five groups, were treated by ethynilestradiol (8,4 x 10-3 g), aquades (10 ml), and turmeric extract at doses 230 mg/kg b.w.; 310 mg/kg b.w.; and 390 mg/kg b.w. for eight days. At the end of experiments the mice were killed, then the uterus, vagina, and mammae were removed and the wet weight of uterus was recorded. Uterus, vagina, and mammae were examined histologically. Estrogen receptor protein from uterus were analized by using SDS-PAGE. One way anava test showed that turmeric extract at doses 310 mg/kg b.w. and 390 mg/kg b.w give estrogenic effect on vaginal ephitelium, endometrium thickness, and diametre of mammary glands. SDS-PAGE analysis showed there were differences in protein concentration between control and treatment groups which were seen in the thickness of the bands. Estrogen receptor band could be detected in sampel of treatment groups at molecular weight 45 kDa

Abstract

Spermatozoa Quality from Polyscias guilfoylei. In our study, we investigated the effect of extract Polyscias guilfoylei L.H. Bailey leaves on spermatozoa quality in rabbits. The experiment used 18 male rabbits from New Zealand white strain which were devided randomly into three groups. Group I was normal control group, Group II and III were given the extract of Polyscias guilfoylei leaves in these following doses: 327.56 mg/1.5 kg body weight and 655.2 mg/1.5 kg bw. The extract was given orally once a day, continuously for 8 weeks. Semens from the rabbit was taken every two weeks and were observed over the quality and consentration of spermatozoa. The result of the research showed that the extract of Polyscias guilfoylei leaves with dosage of 327.56 mg/1.5 kg bw and 655.2 mg/1.5 kg bw could decrease the consentration and the quality of spermatozoa of the male rabbits
