Abstract

Trigonella  foenum-graecum  (TFG)  is  one  of  medicinal  plants  contains  some steroidal sapogenin  such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, also  alkaloid  trigonellin,  which  is  have  many  activity  as  antidiabetic,  estrogenic  and  also as  anti  cancer.    This  experiment  was  done  to  explore  the  activity  of  some  extract  of TFG on some cell lines such as MCF7 (Human Breast Cancer Cell-line), T47D (Human Breast  Cancer  Cell-line),  PC3  (Human  Prostate  Cell-line)  and  SKOV  (Human  Ovarian Carcinoma Cell-line).  This assay was done using MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium)  methods.    Results  showed  that  ethyl  acetate  fraction  gives  the lowest  IC50  than  another  extracts.  IC50  for  PC3  is  66.24  ppm,  IC50  for  MCF7  is  41.81 ppm,  IC50  for  T47D  is  58.63  ppm.    These  datas  can  be  used  for  further  research  to isolate the active compound from TFG. Abstract

Phytoestrogen is the chemical compound contains in plant which has estrogenic like effect. Estrogen has important function on woman’s sexual organ, such as proliferation of uterine and vaginal cornification. Phytoestrogen are weak agonists for estrogen and illicit statistic significantly increases in uterine wet weight, at definite dose, in uterothrophic bioassay. Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that suspected having an estrogen-likeeffect or as phytoestrogen. The aim of this research was to investigate the estrogenic effect of fenugreek’s ethanolic extract in immature rat models. These models represent the climacteric/menopause phase, where estrogen level is verylow because ovary produces no estrogen. The testing animals were divided into five groups, namely normal group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW). Theresult indicated that on 120mg/200gBW, fenugreek extract significantly (=0.05) increasing wet weight uterine and proliferation of uterine, such as uterine thickness and uterine diameters. Fenugreek extract, start on 60mg/200gBW, also induces vaginal lubrication and estrus cycle.

Abstract

Hematology Profile of Sprague-Dawley Male Rats (Rattus norvegicus) Aged 7 and 10 WeeksCurrently, the normal hematology profile has not been yet available for outbred rats in Indonesia. Most of the researchers use the data of hematology profile from references outside Indonesia in which the breeding environment is different. This study aimed to obtain a hematology profile of Sprague-Dawley male rats aged 7 and 10 weeks. The animals were obtained from the Laboratory of Animal Experiment, NADFC. Blood was drawn from the retro-orbital sinus and then analyzed to obtain the hematology profile including erythrocytes, hemoglobin, hematocrit, MCV, MCH, MCHC, leukocytes. Statistically significant differences for erythrocytes, hemoglobin, MCV, MCHC, thrombocyte, and leukocytes parameters were observed between rats aged 7 and 10 weeks. However, there were no statistically significant differences observed on MCH, thrombocyte, and body weight parameters. Data presented here would be useful as a reference for hematology research as well as in evaluating alterations in blood parameters.Keywords: erythrocyte, hematology, leukocyte, Sprague-Dawley, thrombocyteABSTRAKSaat ini belum banyak tersedia informasi profil hematologi untuk hewan coba tikus yang dikembangbiakkan di Indonesia. Kebanyakan peneliti menggunakan data profil hematologi dari referensi luar Indonesia. Penelitian ini bertujuan untuk mendapatkan data hematologi tikus jantan usia 7 dan 10 minggu.  Hewan coba yang digunakan adalah tikus jantan galur Sprague-Dawley yang diperoleh dari Laboratorium Hewan Percobaan-BPOM. Sebanyak 60 ekor tikus jantan yang terdiri dari 30 ekor usia 7 minggu dan 30 ekor usia 10 minggu diambil darahnya melalui sinus orbitalis. Selanjutnya, dianalisa jumlah sel eritrosit, hemoglobin, hematokrit, MCV, MCH, MCHC, leukosit. Hasil analisis statistik menunjukkan terdapat perbedaan signifikan antara tikus jantan usia 7 dan 10 minggu untuk parameter jumlah eritrosit, hemoglobin, hematokrit, MCV, MCHC dan leukosit. Tetapi tidak terdapat perbedaan signifikan untuk jumlah MCH, trombosit dan berat badan tikus. Data yang diperoleh dapat digunakan sebagai referensi penelitian yang berhubungan erat dengan profil hematologi dan penyakit kelainan darah lainnya.

Abstract

Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that are suspected having an estrogen-like effect or as phytoestrogen. The objective of this study was to evaluate the estrogen-like effect of biji klabet alcohol extracts. This research used immature Wistar female rats, age of 19 days, which were represent the climacteric/menopause phase. The testing animals were divided into five groups, namely normal group, estradiol control group and three level dose groups of fenugreek extract (30 mg/200 gBW; 60 mg/200 gBW and 120 mg/200 gBW). The result showed that on 120 mg/200 gBW fenugreek extract significantly (a=0.05) increased estradiol and FSH level on immature rat. Analysis of vaginal smear showed vaginal lubrication effect, surprisingly on 120 mg/200 gBW could induce estrous cycle.

These models represent the climacteric/menopause phase, where estrogen level is very low because ovary produces no estrogen. The testing animals were divided into normal group, ovariectomized control group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW). The result indicated that start on 60mg/200gBW, fenugreek extract significantly could increasing mammary gland proliferation. Empirically fenugreek containing diosgenin, that caused breast enhancement. This research showed that treatment with fenugreek extract can caused proliferation of mammary gland, both on imma-ture and ovariectomized rats.

Abstract

Hiperuresemia prevalence tends to increase in society. A combined extract of Uncaria gambir (gambir) and Caesalinia sappan (secang), had been proven to reduce blood uric acid level in vivo. This study aimed to evaluate the subchronic toxicity of this combination in male and female Sprague Dawley rat strain. Animals were randomly grouped into four groups, namely, DOSE-1 (75 mg/kg bw), DOSE-2 (300 mg/kg bw), DOSE-3 (1200 mg/kg bw) and control group gavaged with carrier. The tested sample was given for 7 weeks orally. The result of blood biochemical parameters were not different significantly compared to control (p> 0.05), as well as the results of hematology analysis. However, the NEUT level of female of DOSIS-2 showed lower and significantly different compared to control (p 0,05). Furthermore, this dose did not influence feed intake and body weight of animals in each sex. From this study, it could be concluded that the combination administrated at the dose of 75 mg/kg bw for 7 consecutive weeks did not affect blood biochemistry and hematology and also organ histopathology of kidney, liver, heart, small intestine, and stomach

Abstract

The development of science has found a way to prevent skin aging in menopausal women. One of the therapies is estrogen replacement therapy (17β estradiol). Skin aging in menopausal women is associated with decreased ERα expression. Leunca fruit (Solanum nigrum L) is known to contain phytoestrogens and has the potential as an antiaging agent. The purpose of this study was to predict the anti-aging effect of Solanum nigrum ethanol extract compounds through the Erα 1A52 [A] activation pathway. The research method of active substance analysis metabolite profiling was carried out by analyzing physicochemical properties using pkCSM. This analysis was carried out as an initial selection of compound safety for therapy. Molecular docking with the MVD (Free trial) application. The RMSD value ≤ 2Å indicates a valid docking and the application can be used. The results of the in silico study showed that the results of metabolite profiling showed that diosgenin was an agonist against ERα. The compound has physicochemical properties according to the Lipinski standard so that it can bind to ERα. The conclusion is that diosgenin has the potential and can act as a 1A52 [A] inhibitor so it can be used as an antiaging agent.

Abstract

Present investigation shows that the extract of C. aeruginosa attenuates DMBA-induced spleen carcinogenesis in Wistar rats. Three-week female Wistar rats were treated with three different C. aeruginosa extract doses (CA1: 40 mg/200 g body weight, BW; CA2: 80 mg/200 g BW; CA3: 160 mg/200 g BW) and were induced with DMBA after one-week administration of these doses. A commercial immunostimulant, and DMBA only were also given to each group as positive and negative control, respectively. The development of tumors was evaluated by investigating the incidence of tumor and tumor multiplicity during the experiment. Spleen mass index and histological parameters such as white pulp, centrum germinativum, and marginalis zone were also examined. Based on our study, the administration of C. aeruginosa extract during and after carcinogen induction gave several impacts on rat carcinogenesis. At the extract dose of 80 mg/200 g BW, tumor incidence of animals were least (P<0.05). However, all doses did not show any effect to the spleen mass index, though the highest dose (160 mg/200 g BW) was found to cause changes in white pulp and marginalis zone boards. This trend indicates that it takes higher dose to cause an immune response effect reaching the organs.
