Abstract

ABSTRACT :Hair loss affects one’s self image and emotional well-being. It is a very real and serious aesthetic concern for everyone. Leaves and flowers of Hibiscus are traditionally known to potentiate hair growth promotion. In this study we determine the hair growth promotion activity of hibiscus leaves using 2.5%, 5% and 10% of hibiscus leaf extract in the form of hair tonic. We also assessed the physical stability and safety of the hair tonic. Hair growth promotion activity assay was carried out by applying hair tonic on the rabbit’s back and measured hair length, hair thickness, hair weight and hair density. Physical stability assessment was performed at low (4°C ± 2°C), room (25°C ± 2°C) and high (40°C ± 2°C) temperature as well as the cycling test. Safety test was done by eye irritation test with HET-CAM and skin irritation test with patch test method. The results showed that hair tonic contains 10% of leaf extract have hair growth activity promotion better than the minoxidil 2%. Physical stability test showed extract of hibiscus leaf hair tonic has good physical stability. Results of safety test showed that there’s no skin irritation occurred while eye irritation test showed positive eyes iritation. Daun dan bunga kembang sepatu telah diakui memiliki aktivitas pertumbuhan rambut berdasarkan penggunaan tradisional. Pada penelitian ini, 2,5%, 5% dan 10%  ekstrak daun kembang sepatu diformulasikan dalam bentuk hair tonic karena penggunaannya lebih mudah dan tidak lengket seperti sediaan semisolid. Tujuan penelitian ini adalah untuk mengetahui aktivitas pertumbuhan rambut ektrak etanol daun kembang sepatu  stabilitas fisik dan keamanannya. Uji aktivitas pertumbuhan rambut dilakukan dengan mengoleskan sediaan hair tonic pada punggung kelinci dan diukur panjang rambut, ketebalan rambut (diameter rambut), kelebatan rambut (bobot rambut) dan kepadatan rambut (densitas rambut). Uji stabilitas fisik dilakukan pada penyimpanan suhu rendah (4oC±2oC), suhu ruang (25oC±2oC) dan suhu tinggi (40oC±2oC) serta  cycling test. Uji keamanan dilakukan dengan uji iritasi mata dengan metode HET-CAM dan uji iritasi kulit dengan metode patch test. Hasil menunjukkan bahwa sediaan hair tonic ekstrak daun kembang sepatu 10% memiliki aktivitas pertumbuhan rambut yang lebih baik dibandingkan kontrol positif minoksidil 2%. Hasil uji stabilitas fisik menunjukkan sediaan hair tonic ekstrak daun kembang sepatu memiliki stabilitas fisik yang baik. Dari hasil uji keamanan iritasi kulit  tidak terjadi iritasi, sedangkan hasil uji iritasi mata menunjukkan sediaan mengiritasi mata.Kata Kunci: daun kembang sepatu, pertumbuhan rambut, hair tonic, HET-CAM, patch test

Abstract

A total of 21 aquatic plant species were collected from Bantimurung waterfall areas, South Sulawesi, Indonesia, in October 2017. These plant materials were subjected to both species and phytochemicals identification. The aims of this study were to determine the species or taxonomic rank of Indonesian aquatic plants collected from Bantimurung waterfall, South Sulawesi, Indonesia and to identify their chemical compounds (phytochemicals) as a candidate for new herbal medicine. Plant genetic materials used in this study were collected from Bantimurung Bulusaraung waterfall and were then identified based on standard botanical techniques for species identification in the Herbarium Bogoriense, Research center for Biology Indonesian Institute of Science (LIPI), Cibinong, West Java. The samples were subjected to the phytochemistry screening such as alkaloids, flavonoids, tannins, saponins, glycosides, terpenoids and anthraquinone followed the procedures of Indonesian Materia Medika and Harborne.  Results showed that all collected aquatic plant samples were able to be identified, including their species names. Phytochemical screening of each sample revealed the presence of glycoside in all of the tested species. However, no alkaloids, anthraquinones, and terpenoids were observed in those tested plant samples. Of the total 21 aquatic plants, 14 species contained flavonoids, 8 species contained phenol compound, and 10 species contained saponins. Among these species Donnax canniformis possessed good antioxidant activity, which correlated to its total phenolic and flavonoid contents. Our results would be beneficial for any future effort in the development of new herbal drugs derived from aquatic plants.

Abstract

Several of Calophyllums genus have been searched and proven as medicinal plants and Calophyllum macrophyllum is one of its genus. We have been isolated a compound from ethyl acetate fraction of the stem-bark. The compound was flavan-3-ol (5,7,2’,5’-tetrahydroxy flavan-3-ol). This research aimed to determine inhibition antidiabetic activity and affinity of its compound on α-glycosidase enzyme. In vitro antidiabetic effect shown by IC50 9.10 µg/ml and docking by Arguslab 4.01 shown by ΔG -10,24 kkal/mol as in silico test. The inhibition activity of flavan-3-ol against the α-glucosidase enzyme that is very strong shows its promising potential as a candidate for antidiabetic drugs.

Abstract

Gambir (Uncaria gambir Roxb.) is a member of family of Rubiaceae. In Asia including Indonesia, extracts of gambir are empirically used daily to weed out. The high content of catechin flavonoids in gambir has a pharmacological effect in the treatment of hyperlipidemia that potential to be developed into traditional medicine. This literature review aimed to examine the potency of pharmacological effect of gambir as hyperlipidemia treatment therapy based on the results of studies in silico, in vitro, in vivo pharmacological effects and its safety to provide evidence of scientific information to the community. The literatures used for analysis in this study including evidence-based articles on both pharmacology and safety which are available in Pubmed and Google Scholar. The results showed a very strong potency of gambir plants in the treatment of hyperlipidemia with catechin as bioactive compounds. In silico study revealed mechanism action of catechin as antihyperlipidemic using 2 pathways, inhibition of the enzyme HMG-CoA reductase and increase the LDL receptors. In vitro studies of catechin are able to inhibit lipid absorption in the intestine through inhibition of pancreatic lipase activity, lipid hydrolysis and emulsification, micelle cholesterol deposition. Pre-clinical tests on animals showed that the ethyl acetate fraction of gambir leaves was able to reduce the levels of total cholesterol, triglycerides, LDL and increase blood plasma HDL. The long-term use of gambir leaves has been proven to be safe, not mutagenic, no hematological, clinical biochemical abnormalities and no abnormalities in the vital organs of the animal models. Abstrak Gambir (Uncaria gambir Roxb) adalah tumbuhan perdu dari suku Rubiaceae. Di Asia termasuk Indonesia, secara empiris ekstrak gambir digunakan sehari-hari untuk menyirih. Kandungan flavonoid katekin yang tinggi dalam gambir memiliki efek farmakologi dalam pengobatan hiperlipidemia yang berpotensi untuk dikembangkan menjadi obat tradisional. Kajian literatur ini bertujuan untuk mengkaji potensi farmakologi gambir sebagai terapi pengobatan hiperlipidemia berdasarkan hasil studi efek farmakologi in silico, in vitro, in vivo efek farmakologi dan keamanannya, sehingga memberikan bukti informasi ilmiah kepada masyakarat. Literatur yang digunakan dalam proses review meliputi literatur dengan berbasis bukti baik farmakologi maupun keamanan yang tersedia di Pubmed dan Google Scholar. Hasil kajian menunjukkan potensi yang sangat kuat dari tanaman gambir dalam pengobatan hiperlipidemia dengan katekin sebagai senyawa bioaktif utama. Studi in silico menunjukkan mekanisme aksi katekinsebagai antihiperlipidemia menggunakan dua jalur yaitu penghambatan enzim HMG-CoA reduktase dan peningkatan reseptor LDL. Studi in vitro katekin mampu menghambat penyerapan lipid di usus melalui penghambatan aktivitas lipase pankreas, hidrolisis lipid dan emulsifikasi, serta pengendapan kolesterol misel. Studi in vivo menunjukkan fraksi etil asetat daun gambir mampu menurunkan kadar kolesterol total, trigliserida, LDL dan meningkatkan HDL plasma darah. Penggunaan daun gambir dalam jangka panjang terbukti aman, tidak mutagen, tidak ditemukan kelainan hematologi, biokimia klinis dan tidak menyebabkan kelainan organ vital hewan uji.

Abstract

Rubus fraxinifolius can be founded in the mountainous regions of West Java. Previous studies have shown that this plant has potent antioxidant activity. The content of polyphenol compounds in this species contributes substantially to antioxidant activity, and the extraction method will affect the content of the compound. In this study an evaluation of the extraction method effect on leaves, young fruits and stems of R. fraxinifolius on the content of polyphenols and their antioxidant activity, have been done. Method: The leaves, fruits, and stems of R. fraxinifolius were extracted by maceration, reflux, and soxhlet using methanol as a solvent. Each extract was determined the total polyphenol content using Folin-Ciocalteu reagent and antioxidant activity test using DPPH reduction method. Result: The highest extract yield was R. fraxinifolius leaf reflux extract (30.20%). The highest total polyphenol content was given by the Soxhlet extract of leaf R. fraxinifolius (48.79 mg GAE / g extract). The highest antioxidant activity was leaf Soxhlet extract of R. fraxinifolius (98.29% at 100 ppm). Conclusion: From this study it was shown that the methanol extract of R. fraxinifolius leaves extracted by Soxhlet gave the highest polyphenol content and antioxidant activity.

Abstract

Senyawa Etil P-Metoksi Sinamat (EPMS) merupakan senyawa yang tidak stabil dalam suhu panas dan mudah terhidrolisis menjadi asam p-metoksi sinamat (APMS). Senyawa EPMS dengan penyimpanan yang cukup lama dikhawatirkan dapat mempengaruhi pH sediaan krim dan dapat menyebabkan kulit menjadi iritasi yang berbahaya bagi pasien. Penelitian ini bertujuan untuk mengetahui stabilitas kimia senyawa EPMS pada sedian krim anti jerawat tipe O/W yang diukur tiap 2 minggu selama penyimpanan 3 bulan, dengan metode Kromatografi Lapis Tipis Densitometer (TLC Scanner). Dari penelitian ini diketahui stabilitas kimia krim EPMS selama penyimpanan 3 bulan mengalami sedikit perubahan dengan penurunan kadar EPMS dalam krim sebesar 4,89%.

Abstract

The previous research showed that the fraction C from active extract of Garcinia latissima Miq. fruit was active against Bacillus subtilis. This study aims to isolate and identify the active compound as an antibacterial agent from the fraction C. Fraction C was purified by recrystallization using chloroform and n-hexane solvents and then isolated using preparative-thin layer chromatography-silica gel 60 GF254 to give a yellow compound. The antibacterial activity was determined using microdilution with thiazolyl blue tetrazolium bromide indicator against B. subtilis American Type Culture Collection 6633. The isolate was identified using UV-Vis, IR, MS, Proton Nuclear Magnetic Resonance (1H-NMR) and carbon NMR (13C-NMR), and NMR-2D techniques including HMQC and HMBC. Based on the spectroscopic analysis and literature review, the compound was identified as 6-deoxyjacareubin, which is a new compound from Garcinia latissima Miq. The 6-deoxyjacareubin showed antibacterial activity with MIC value of 156.25 ppm and was categorized as a weak antibacterial agent because the MIC value was more than 100 ppm. According to in silico approach to the docking study, 6-deoxyjacareubin showed similar hydrophobic interaction with several amino acid residues including C2565, C2589, G2484, U2590, and U5588 between a native ligand.

Abstract

Garcinia kydia Roxburgh merupakan salah satu spesies dari genus garcinia, yang secara khusus belum banyak diketahui aktivitas senyawa bioaktif, namun pada penelitian sebelumnya terbukti berpotensi sebagai agen penghambat α-glukosidase. Pada penelitian ini, akan dilakukan investigasi aktivitas antioksidan terhadap berbagai fraksi yang diperoleh dari daun G. kydia Roxb. Metode penelitian dilakukan dengan menentukan aktivitas antioksidan melalui analisis in-vitro menggunakan microplete reader dengan metode radical scavenging (Metode DPPH) dan ferric reducing antioxidant power (Metode FRAP). Hasil menunjukkan bahwa fraksi yang paling aktif dari ekstrak etil asetat adalah FEA8 (DPPH) dan FEA7 (FRAP), sedangkan fraksi paling aktif dari ekstrak metanol adalah FMT4 (DPPH) dan FMT6 (FRAP). Aktivitas antioksidan tertinggi di antara empat fraksi adalah FEA7 dan FEA8. Aktivitas fraksi FEA7 dan FEA8 ditentukan oleh nilai EC50, sehingga diperoleh masing-masing 9,73 dan 9,26 μg/mL (DPPH); 10,28 dan 13,94 μg/mL (FRAP). Garcinia kydia Roxb. berpotensi sebagai agen antioksidan dengan mekanisme yang berbeda. Sehingga hasil penelitian terkait terdapatnya potensi antioksidan tersebut dapat memberikan alternatif terapi diabetes yang disertai komplikasi stres oksidatif.
