Abstract

Oxidative stress induced by the radicals have been known to affect the occurrence of various degenerative disease. In order to search natural antioxidant compounds, it had been studied the isolation and identification of antioxidant compounds in fern stems (Alsophila glauca J.Sm) using DPPH method (1,1-diphenyl-2-picrylhydrazyl). Fern stem extraction was done by maceration with wasbenzen and metanol. Extracts was obtained by evaporating the wasbenzen solvent with a rotavapor. Material was re-macerated with methanol after wasbenzen evaporation. The same way was done to obtain the metanol extract. These extracts were tested for antioxidant activity using DPPH method with TLC. Active extract was partitioned in 80% methanol. Both 80% methanol soluble and insoluble extract were tested their antioxidant activity using DPPH method (TLC). Active extract was fractionated in vacuum liquid chromatography using a mobile phase with different polarity gradient and different concentrations (wasbenzen:chloroform). Active fraction was isolated by preparative TLC method and purity of obtained compounds was tested by TLC. Antioxidant activity of obtained isolates was tested by DPPH method using spectrophotometry. Compounds known to had antioxidant activity as radical catcher with IC50 178.4µg/mL.

Abstract

Free radical induced oxidative stress that influences the occurrence of various degenerative diseases such as cancer, coronary heart disease and premature aging. In the case that body's antioxidant defense system does not have excessive antioxidants, additional natural antioxidant via food or other nutrients intake is needed. Stems of ferns are known to contain phenolic compounds that are known to have antioxidant activity. A study has been carried out to determine antioxidant potential of stems of ferns of western Borneo origin compared to quercetin as positive control. Initially, material was macerated gradually with wasbenzen followed by methanol. The extract obtained was filtered and evaporated to dryness to give dried wasbenzen extract and methanol extract respectively. Antioxidant properties of extracts were performed by DPPH on TLC. Active (wasbenzen) extract was then partitioned with 80% methanol to give soluble and insoluble fractions. Active (insoluble) fraction was further fractionated by vacuum liquid chromatography using a gradient mobile phase with increasing polarity (wasbezen: chloroform). Fractions obtained were tested antioxidant activity, and isolation was done by preparative TLC. The purity of isolated antioxidant compound was confirmed by TLC, and antioxidant potential was measured spectroscopically with DPPH method. This compound displayed antioxidant activity, but its value was lower than that of quercetin used as a positive control (IC50 178.4 v / s 2.17 microg / mL)

Abstract

Antioxidant is a substance which in small concentrations can significantly inhibit or prevent the oxidation of the substrate. One plant is efficacious as an antioxidant is the persimmon (Diospyros kakiThunb.) where is cultivated widely in East Asia, Spain and Indonesia. In the search for natural antioxidant compounds, have been studied isolation and identification of antioxidant compound of persimmon leaves (DiospyroskakiThunb.) using DPPH (2,2-diphenyl-1-pikrilhidrazil) method. The extraction of persimmon leaves was carried out by maceration method with wasbenzen. Extracts obtained by evaporating the solvent wasbenzenon a rotary evaporator and were then re-macerated with methanol. Antioxidant activity of extracts was evaluated with DPPH 0.2% method. Active extract was partitioned with chloroform, methanol, distilled water and tested for antioxidant activity by DPPH 0.2% method. The active compound was purified byPTLC and the purified active isolates was then confirmed by TLC. The antioxidant activity (IC50) active isolates were analyzed using spectrophotometry. Analysisof active isolates resulted the antioxidant activity (IC50) of 100.00 ug / ml. UV-Vis spectrum of the active isolates showed absorption at λmax 285 and 401 nm. Infrared spectra (KBR) showed absorption at 3446 cm-1 (OH), 2926 cm-1 (CHaliphatic), 1456 cm-1 (CH2), 1384 cm-1 (CH3), 1255 cm-1, and 1115 cm-1 (C-0-C), 1631 cm-1 (C=Caromatic). GC-MS spectra gave two peaks with retention time 11.408 minutes with a similarity index of 79.243% with the molecular ion (M•)+ appeared at m/z 178, 163, 147. These spectra are expected to be methyl eugenol. Furthermore, the peak with retention time 12.982 minutes with a similarity index of 20.757%, producing a mass spectrum with molecular ion peaks (M•)+appeared at m/z 192, 177, 161. These spectra are expected to be myristicin compound.

Abstract

Red Cabbage (Brassica oleracea L. Var. Capitata f. rubra) has a high anthocyanin content therefore it can be potential as a natural antioxidant. In this research, the antioxidant activity of the red cabbage extract was evaluated quantitatively with a spectroscopy method using DPPH reagent to obtain the value of IC50 . This research was divided into 2 stages. First, the influence of solvent variant to the extract yield of red cabbage powder and the maximum wavelength (λ) using a maceration protocol. The kind of solvent (70%, 80%, 95%, and 96% of ethanol) with addition 3% of citric acid. Second, the influence of extraction methods in neutral condition to the antioxidant activity. The results show the red cabbage powder maceration with 96% ethanol solvent (acid condition) exhibits  the highest yield. The fresh red cabbage soxhletation with 96%  ethanol (neutral condition) exhibits 288,5 nm maximum wavelength (λ) and 168,78 mg/mL of IC50 value.

Abstract

Sumbawa horse milk is one of pure milk of wild horses farmed by organic farmers in the island of Sumbawa, West Nusa Tenggara, Indonesia. The communities claim that milk has many benefits. In this study Sumbawa horse milk was effective as an antibacterial against Staphylococcus epidermidis (one of the bacteria caused acne). Antibacterial activity test performed by spread plate method and quantified by standard plate count. Percentage of the number of colonies after contact with  Sumbawa horse milk for 5 minutes (used acid control). The result of 25% is 99.96%, 12.5% is 99.99%, 6.25% is 99, 67% and 3.125% is 99.73%. This suggests that  Sumbawa horse milk has good antibacterial activity until 3.125%.

Abstract

On Timor island, Nusa Tenggara Timur, faloak barks (Sterculia quadrifida R.Br.) has been used empirically to restore stamina. Faloak bark ethanolic extract proved to have immunomodulatory activity in vitro, which can increase macrophage phagocytosis activity. This research aimed: (i) to determine the immunomodulatory active fraction of faloak bark ethanolic extract, (ii) to determine the total flavonoid contents of faloak extract and fractions, and (iii) to evaluate the correlation of the total flavonoid contents of those extract and fractions with their macrophage phagocytosis activity. The simplisia powder is macerated with 96% ethanol. The extract was dissolved in methanol:water (9:1v/v) was then subsequently partitioned with n-hexane, ethyl acetate, and water to obtain n-hexane fraction, ethyl acetate fraction, water fraction, and insoluble fraction. Faloak extract and fractions at concentration 62,5; 125; 250; 500μg/mL were tested for their effect on the peritoneal macrophage phagocytosis of Balb/c mice in vitro by the latex beads method. Phagocytosis capacity and phagocytosis index were analyzed using one-way anova and post hoc Tukey HSD test with 95% confidence level. The results showed that ethyl acetate fraction had the highest macrophage phagocytosis capacity and the highest total flavonoid content compared to other fractions. The highest macrophage phagocytosis capacity of ethyl acetate fraction at concentration of 250 μg/mL was 51,94±4,67%, this value was significantly different from cell control (7,50±1,29%), negative controls of 0,0625% dimethylsulphoxide (6,25±0,36%), as well as positive control of 200 μg/mL echinaceae extract syrup® (9,97±0,33%). The total flavonoid content of ethyl acetate fraction determined by aluminum chloride method was 4,290±0.029 mg of quercetin equivalent/g fraction. There was a positive and strong correlation between the total flavonoid content of these extract and fractions with their macrophage phagocytosis capacity (Pearson correlation coefficient of 0,781) and showing linear relationship y=4,721x+19,663; R2=0,61.

Abstract

In Indonesia, beras (Oryza sativa) has become the main ingredient in natural beauty products such as masks, scrubs, and cold powder. γ-oryzanol and phenolic compounds are the major components in beras that have many benefits, one of which as antioxidants. This study aims to determine the content of γ-oryzanol and total phenolic compounds in different beras cultivars in the area of Yogyakarta. The leveling was done by a TLC-densitometry method and Folin Ciocalteu. The results showed that the largest γ-oryzanol content found in red beras was 0.0843% by weight of the sample. The highest total phenolic content was found in red beras as much as 1.92% by weight of the sample. The antioxidants activity IC50 Red Beras is 0.591.

Abstract

Green tea (Camellia sinensis L.) is known to have ability to protect skin against free radicals. This is supported by polyphenol compound catechin. This research aims to determine the optimum Hydrophilic-Lipophilic Balance (HLB) value of Tween 60 and Span 80 compositions on the optimum cream formula of ethanol extract of green tea leaves. Tea leaves are extracted by macerating using 70% ethanol. Catechin in extract is known from Thin Layer Chromatography (TLC) test with silica gel 60 F254 as stationary phase and ethyl acetate:aquadest:formic acid (18:1:1 v/v) as mobile phase. Antioxidant activity is determined by 2,2-Diphenyl-1-picryhydrazyl (DPPH) method and value of Inhibition Concentration 50% (IC50) is then calculated. Formula optimization using Design Expert® version 7.1.5 (DX 7) software, Simplex Lattice Design (SLD) method with two components Tween 60 and Span 80. Cream is characterized according to physical properties organoleptic, homogeneity, viscosity, pH, spreadability, adhesiveness, and cream type. The optimum formula obtained is then tested for physical stability for 4 weeks at room temperature (28±2°C) and data are statistically analyzed using one-way ANOVA. The extract contains catechin proved with Retention factor (Rf) value 0.8 and has antioxidant activity with IC50 value 56.35 ppm. 6.4% Tween 60 and 3.6% Span 80 result an optimum HLB value 11.1. It has viscosity 2897.50±35.94 mPa.s, spreadability 18.44±0.06 cm2, adhesiveness 0.85±0.05 seconds, and pH 4.530±0.002. Statistical test shows that the cream is significantly altered at pH, but does not significantly change in viscosity, spreadability, and adhesiveness after being stored for 4 weeks.
