Abstract

Forty healthy and non-pregnant Fries Holland dairy cows which had received oestrus synchronization were divided randomly into four experimental groups. Artificial inseminations (AI) were implemented 8-10 hours post estrous using frozen semen from the same breed. The first three groups were injected with 100, 150 and 200 mg medroxy progesterone acetate (MPA) i.m., respectively, three days post AI, while the other group was injected with placebo as control group. Madura cattle embryos were transfered contralaterally seven days post AI. The percentage of pregnancy in groups which had received 100 and 150 mg MPA was 37.5%r, and in group received 200 mg MPA and control was 55.6%, and 60%, (P>0.05), respectively. Calving percentages of the 100, 150, 200 mg MPA and the control groups were 25,37.5,55.6 and 60% respectively (P>0.05). All birth gave single calve.

Abstract

The researchs of immunocontraception have done in ZP3 of several species, but have not been done in ZP3 of goat. In preliminary study, gZP3 protein was effective prohibited of graviditation of mice. The aim of this study was to prove the potency of gZP3 protein to prohibit in vitro fertilization of mice as an animal model. Antibody of gZP3 produced on mice. Abstract

The aim of this study was to evaluate the efectiveness of PGF2? on the decrease level of progesterone after treated with PGF2?  that extracted from bali cattle vesicula seminalis monolayercell that stored for 0 month, two months and four months at temperature 40C and -4oC. Sample that used in this study was PGF2? producted from bali cattle vesicula seminalis monolayer cell. Acomplete random sample with two factorial pattern was used i.e; factor I was the period stored (0month, two months, and four months) and factor II was the temperature storage  (4oC).Each treatment was completely with repetitions. Result showed that PGF2? that resulted from balicattle monolayer cell vesikula seminalis that stored for two months was able to decrease level ofprogesterone to 49.06% at -4oC  and 48.82% at temperature 40C. Similarly. The PGF2? that storedfor four months affected on the decrease of progesterone level to 36.33% at temperature -4C and35.81% at temperature 40C. It can be concluded that PGF2? that resulted in the study that stored forfour months at temperature of -40C and 40C may have capability to regress luteal cell.

Abstract

Osteoporosis in postmenopausal women happened due to estrogen deficiency which leads imbalancebone-formation and bone-resorption process. Recently, phytoestrogen as an alternative hormon replacementtherapy in postmenopausal women could overcome estrogen deficiency. The objectives of this study was toevaluate the effect of tomato juice administration and combination of physical exercise on bone density inestrogen deficiency condition. Twenty eight postovarectomy female rats (Rattus novergicus) were randomizedinto four experiment groups : the controlled group ( 2 mL aquades administration); the exercised group(swimming for 30 minutes, three times in one week), the tomato juice administration every day (44 mg/200g body weight), and the combination exercise and tomato juice administration. Four weeks after treatmentthe samples were collected from os femure for morphological examination. The intervention groups showeda significant difference in bone density with the control group (p<0.05). Bone density highest on the tomatojuice administration group. There is significant difference bone density between tomato juice administrationgroup and the combination group (p<0.05), but no significant difference between physical exercise groupand the combination group. Tomato juice showed an increasing bone density on estrogen deficiencycondition and the combination group have the same effect with the exercised group.

Abstract

The aims of this research were to determine PGF2? concentration the produced by bali cattlesendometrial and seminal vesicle monolayer cell culture and in vitro luteolytic ability on luteal monolayercell culture. The endometrial and seminal vesicle epithelial cell of bali cattle were cultured in tissueculture medium (TCM) 199 growth medium supplemented with 10% fetal calf serum and 10% EstrusMare Serum. The cells were cultured at 1.9 x 106 density per ml medium. Then Followed by incubation at38.50 C in 5% CO2 atmosphere for 12 days. The level of PGF2? in the cell culture medium were assayed byRadioimmnuassay (RIA) technique. The luteal cells were cultured in 9 days incubation and divided into 2groups. Group I were added with 10% of cell culture product and group II were added with 1,25 mgdinoprost/ml. The level of progesterone produced by luteal cell culture was measured at day 9th and 11thincubation. The result showed concentration of PGF2? cell product of seminal vesicle cell culture wassignificantly higher (P < 0.05) compared to endometrial cell culture. There was no significant difference(P>0.05) in luteolytic ability between PGF2? cell culture product and dinoprost. In conclusion, the PGF2?could be produced by monolayer cell culture of bali cattle is endometrial and seminal vesicle epithelialcells more over they have similar ability with dinoprost in luteolytic ability.

Abstract

Studies on the role of zona pellucida glycoprotein-3 (ZP3) in immunocontraception have been conductedin many species including goats (gZP3). Our previous study indicated that gZP3 effective in preventingpregnancy in mice. The aim of this study was to prove the evidence of cross reaction in gZP3 to preventfertilization in cow in vitro. Antibody of gZP3 was produced in mice. Immunized mice serum was analyzedusing ELISA technique and dot blot method. Sperm from frozen semen were processed then incubated incapacitation media supplemented with the gZP3, whilst the cow oocyte was incubated in maturationmedia supplemented with antibody of gZP3. Following this, the normal  in vitro fertilization (withoutincubation neither in gZP3 nor in gZP3 antibody) was performed, respectively. The results showed thatantibody titer of immunized mice serum was higher (p<0.05) than the control group. Dot blotting analysisshowed that the antibody of immunized mice were able to recognize gZP3 protein. The serum of immunizedmice which was supplemented in the maturation media of cow oocyte were able to decrease the cleavagerate (p<0.05). Protein gZP3 which was supplemented in the capacitation media of the sperm also coulddecrease the cleavage rate (p<0.05). It is concluded that goat-ZP3 protein may produce cross reaction incow.

Abstract

The study was carried out to determine the response of PGF2 ? result extraction of Bali cattle seminal vesicle fluid on the level of progesterone. The seminal vesicle fluid was aspirated and then were. extracted with methanol. This reseach was conducted by devided the mare in the luteal phase into two groups. The first group were treated with PGF2 ? result extraction of Bali cattle seminal vesicle fluid and the second group were treated with dinoprost as a patent product of PGF2 ? were administered intra uterine. The level of progesterone was measured before (0 hours) treatment and at 24, 48, 72 hours after treatment. The level of Progesterone were determined by radioimmunoassay (RIA) technique. The result showed that PGF2 ? result extraction of Bali cattle seminal vesicle liquid decreased the progesterone level 73.03% at 24 hours and 92.79% at 48 Hours. However, there was no significant different between PGF2 ? result extraction of Bali cattle seminal vesicle liquid with PGF2 ? of paten product on the progesterone level decreased. In conclusion, PGF2 ? extraction of Bali cattle seminal vesicle liquid can decreas the level of progesterone in the mare with luteal phase.
