Abstract

Forty healthy and non-pregnant Fries Holland dairy cows which had received oestrus synchronization were divided randomly into four experimental groups. Artificial inseminations (AI) were implemented 8-10 hours post estrous using frozen semen from the same breed. The first three groups were injected with 100, 150 and 200 mg medroxy progesterone acetate (MPA) i.m., respectively, three days post AI, while the other group was injected with placebo as control group. Madura cattle embryos were transfered contralaterally seven days post AI. The percentage of pregnancy in groups which had received 100 and 150 mg MPA was 37.5%r, and in group received 200 mg MPA and control was 55.6%, and 60%, (P>0.05), respectively. Calving percentages of the 100, 150, 200 mg MPA and the control groups were 25,37.5,55.6 and 60% respectively (P>0.05). All birth gave single calve.

Abstract

Cellular transplantation using stem cells provides very promising solutions in the regeneration and repairment of cells that have experienced degeneration where recovery through medical or surgical intervension is impossible. However, the very low viability of transplanted stem cells limits the transplantation efficacy. The aim of this research was to obtain viable bone marrow derived stem cells by an adaptation treatment in a low oxygen tensioned in vitro culture. Low oxygen tension adaptation was adjusted to the niche of the stem cells in vivo. In this study, in vitro culture of stem cells in 1% oxygen was compared to those of the conventional culture in 21 % oxygen.Results showed that under 1% oxygentension cell proliferation was slower with larger or rounded triangle shaped cells, and senescence or dead cells was low. Meanwhile under 21 % oxygen tension cell proliferation was two fold faster with flattened and slender cells, and senescence or dead cells was higher. In conclusion, conventional in vitro culture under 21% oxygen caused cell aging (senescence) and rapid cell death, therefore the transplanted cells were not viable.

Abstract

Infectious disease is a major problem for animal husbandry. One of infectious disease is caused by Ascaridia galli (A. galli). Ascaridiasis occurs in either broiler or layer that caused decrease of meat and egg productions. Garlic (Allium sativum) contain alicin  that have the ability to kill parasites.This research was conducted to determine the effect of garlic on time of death of A. galli. In this research used A. galli with length 7-11 cm regardless the sex. The treatment used local (Indonesian garlic), kating and single garlic with concentration of 39.50%, 62.40% and 98.60%. The negative control used NaCl physiology (0.9 %). The result showed that all the treatment group can shortened the time of death of A. galli compared to control. The time of death of A. galli on local garlic infusion with 39.50 % concentration was 6.24 hours, on 62.40 % concentration was 4.94 hours and on 98.60 % concentration was 3.97 hours, kating garlic infusion on 39.50 % concentration was 5.08 hours on 62.40 % concentration was 3.81 hours, on 98.60 % concentration was 2.96 hours, single garlic infusion on 39.50 % concentration was 5.4 hours, on 62.40 % concentration  was 3.86 hours, on 98.60 % concentration was 3.83 hours. The best treatment was kating garlic on 98.60% concentration because give shortest time of death.

Abstract

The aim of this research was to identify worm eggs species that infected Egrets in Surabaya. A total of 40 samples were taken and faecal examination was conducted in the Laboratory by means of native, sedimentation, and floating methods, In floating method saturated brown sugar solution was used as floating media because brown sugar solution was faster in floating worm eggs. Then further examination of the worm eggs species were matched by microscopic taxonomy and textbook comparison observation. Eggs of three classes of helmints, Trematode, Cestode, and Nematode were found in faeces of Egrets in Surabaya. Species of the Nematode eggs found were Ascaris sp., Ascaridia galli, , Capillaria sp., Toxocara cati. Species of Cestode class egg found were Raillietina sp, from Trematode class were Echinostoma revolutum.

Abstract

This study aims to determine the prevalence and species of endoparasite that infect the digestive tract of goats in Ujungpangkah Sub-District, Gresik District. This study was conducted in February-March 2021 with 100 samples of goat’s stool. Sample examination was conducted in the laboratory of the Division of Veterinary Parasitology, Faculty of Veterinary Medicine, Universitas Airlangga. In fecal examination found four types of endoparasites, which were: Eimeria sp. 62%, Blastocystis sp. 5%, Haemonchus sp. 4%, Strongyloides sp. 2% and mixed infection prevalence was 3%. This study showed a prevalence of 76% digestive tract endoparasite. The Chi-Square test showed significant differences (p <0.05) between groups of goats aged under one year (57%) and over one year (43%).

Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis. A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL. The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies. Positive results showed a brownish color on the oocyte and its cumulus. The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased. It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

This study aimed to evaluate the profile of progesterone in dairy cattle with ovarian hypofunction. A total of 10 cows in this study were evaluated three times in the collection phase, i.e. (F1) when the cow was diagnosed with ovarian hypofunction, (F2) when the cow was in heat and (F3) 21 days after artificial insemination to detect pregnancy. Samples in the form of blood serum were then analyzed using ELISA. As a result, the average time of heat in ovarian hypofunction cows was 7,4 days. Progesterone levels in F1 were 1,027 ng/ml, 2,774 ng/ml, 1,476 ng/ml, 2,256 ng/ml, 1,258 ng/ml, 1,758 ng/ml, 2,393 ng/ml, 0,592 ng/ml, 0,755 ng /ml, 1,876 ng/ml, respectively. Progesterone levels in F2 were 0,671 ng/ml, 0,517 ng/ml, 0,763 ng/ml, 0,598 ng/ml, 0,615 ng/ml, 0,537 ng/ml, 0,726 ng/ml, 0,643 ng/ml, 0,593 ng/ml, 0,975ng/ml, respectively. Progesterone levels in F3 were 15,642 ng/ml, 4,215 ng/ml, 17,327 ng/ml, 20,721 ng/ml, 5,796 ng/ml, 17,214 ng/ml, 15,815 ng/ml, 16,745 ng/ml, 4,632 ng /ml, 18,281 ng/ml, respectively. The pregnancy rate in hypofunctional cows treated with PG-600 in this study was 70%.

Abstract

The Leptin gene is the gene that produces the leptin hormone, which is released from adipose tissue and can increase the productivity of animals. This study aimed to identify polymorphic nucleotides, changes in amino acid components, and species of goats based on GenBank Leptin DNA sequence data. A total of five goat leptin DNA sequences were extracted from NCBI GenBank data. The leptin DNA sequence was aligned with Bioedit to locate SNPs and amino acid changes. The tree produces cultivars grown using Clustal Omega Ver. 1.2.4. Based on the DNA sequencing results of leptin genes in five goats, five SNPs were located in the coding sequence (CDS), SNPs g.17T/A, g.43T/A, g.74G/A, g.93C/A. and d. 386A/G. SNP was a missense mutation and a silent mutation. The analysis of phylogenetic trees of Leptin showed that there were three breeds of goats in one branch and two breeds of goats in different branches. These results provided the first report for further studies on the genetic diversity of leptin genes in different local goat breeds.

Abstract

The aim of this research was to know histopathological changes of kidney of broiler chicken exposed to chronic heat stress. Twenty broilers were divided randomly into 2 groups, 10 broilers each are being exposed to chronic heat stress for 21 days. The first 21 days were the adaptation period in a chamber with temperature 24-28o C and humidity 40-55%. After 21 days, continue with exposure to heat stress in a chamber with temperature 36-40o C and humidity 50-65% in 8 hours per day. After exposed to chronic heat stress, kidney tissues were processed, and kidney tissue histopathological changes were evaluated by using the Klopfleisch modified scoring method. The data was analyzed by Mann – Whitney Test. The result of this research showed that chronic heat stress exposure causing the presence of degeneration of tubular epithelial cell, necrosis of tubular epithelial cell, necrosis of glomerular and interstitial infiltration. From the analysis data the overviewof multiparametric showed that when control group compared with the treatment group showed significantly difference (p<0.05).

Abstract

The aim of this research was to determine the level of stability of Progesterone-retention time on Eluen Phase Mobile (EPM) solution in storage period for six, eight and 12 hours using High Performance Liquid Chromatography (HPLC). The research method used posttest-only control group design by using three treatments and six repetitions. The three repetitions consist into six hours, eight hours and 12 hours. The data were obtained analyzed by Summery Independent T-Test with SPSS 24 for windows. The results showed that the progesterone treatment in EPM solvents with six-hour storage (P1) and eight hours (P2) retention time progesterone are stable but the progesterone treatment in EPM solvent with 12-hour storage (P3) showed an unstable due to storage time. Besides that there are irrelevant peak which show that many pure molecular fractions are separated in the stationary phase so that the molecular structure of the substance begins to break down. Based on these results, it can be concluded that the storage time of Progesterone in Eluen Phase Mobile began to be unstable at 12 hours of storage.
