Abstract

Decolorization of Sumifix Blue and Reactive Red 2 Textile Dyes by Microbes Isolated from Textile Waste WaterAzo dyes represent the most commonly used group of dyes in textile industry and discharged into industrial effluents worldwide. Aims of this study are to isolate microbe from textile waste water and to determine their ability to decolorize Sumifix Blue and Reactive Red 2 textile dyes. Microbe was isolated from textile effluent of PT Timatex, Salatiga. The activity for decolorization was assayed by inoculating microbial isolates into dye containing medium. Living and nonliving cell were incubated in dye containing medium in order to determine if microbial cells involved in decolorizing dye. Five different microbial isolates have been isolated from textile waste water.  Isolates IBLTT_1 and IBLTT_5 showed the highest activity to decolorize Sumifix Blue, and only isolate IBLTT_1 showed the highest capability in decolorizing Reactive Red 2. Both isolates indicated positive potential towards biotreatment of textile waste water. Further results confirmed that decolorization was due to biodegradation, rather than physical adsorption by inactive cells. Keywords: decolorization, microbial isolation, Reactive Red 2, Sumifix Blue, textile effluent ABSTRAKPewarna azo mewakili kelompok pewarna yang umum digunakan pada industri tekstil dan banyak dijumpai di buangan limbah industri tekstil.

Abstract

Although herbicide and insecticide contamination of surface waters normally occurs in the form of mixtures, the toxicity interactions displayed by such mixtures have only rarely been characterized. This study evaluated the acute effects of single pesticides (paraquat dichloride and deltamethrin, tested in their commercial formulations Gramoxone 276 SL and Decis 25EC) and their equitoxic mixtures on the survival of adult male guppy fish (Poecilia reticulata). Mixture toxicity was evaluated against the reference models of Concentration Addition (CA) and Independent Action (IA). In the single treatments, the 72h LC10 and LC50 values were 1.5 and 6.0 mg a.s. L-1 and 0.53 and 3.6 µg a.s. L-1 for paraquat dichloride and deltamethrin, respectively. The equitoxic mixtures showed significant paraquat dichloride-deltamethrin antagonism, both based on the CA and the IA model, without significant dose-level dependent deviations.

Abstract

Service-learning has gained substantial recognition as an effective type of pedagogy and has enhanced civic education across the disciplines, however remains a lack of understanding of this type of learning in Indonesia. The goals of the study were (1) to explore the forms of local knowledge systems practiced in Semarang Regency and how they are used in resources conservation using service-learning method, (2) to foster student engagement with the community, and (3) to promote student awareness of community resources that are directly relevant to local knowledge system issues. The success of the service-learning projects in meeting these goals was assessed via qualitative analysis of student reflective papers and classroom presentations. The results indicated there were local knowledges that were still successfully in use, but often only very locally. Furthermore, the results also indicated that the service-learning projects promoted students’ valuable academic skills, including communication, team-building, and critical thinking, built their self-esteem, their awareness of community needs and resources, and demonstrated the relevance of course content to real life.

Abstract

Pesticides pose a risk to public health and environmental in agricultural areas. This study described pesticide use among farmers in Oebobo, Batnun, Tupan and Pollo villages, South Central Timor, Indonesia. Some 400 farmers were interviewed about pesticide use, knowledge and attitude, symptoms of intoxication, personal protective equipment and hygiene. The farmers had adequate knowledge about pesticide toxicity, but farmer’s awareness did not necessarily translate into action. Majority did not use appropiate personal protective equipment nor good hygiene when handling pesticides. Farmers in South Central Timor did not use the most hazardous pesticides (class Ia and Ib). However, use of class II pesticides and those of lower toxicity in combination with inadequate practice among farmers was found in the study areas. Expensive protective equipment and behavioral patterns, such as spraying without protective clothing and mixing pesticides with bare hands combine to produce several well-documented health problems. Therefore, alternative crop management systems which could help preventing the negative effects of pesticides on farmer health and the environment are needed. Training farmers in integrated pest management methods, use of proper hygiene and personal protective equipment when handling pesticides should be promoted.

Abstract

Gedong Songo is a National Tourism Development Area in Bandungan region with an increasing number of visitors. Previous studies found that littering and water pollution were some of the prominent problems in Gedong Songo. The lack of knowledge and awareness towards environmental management was identified as the center of the issues. A conservation effort in the form of an early environmental education targeted elementary school students was suggested to address this problem. The environmental education was performed in State Elementary School Candi 1 and Candi 3 Bandungan and was designed as a series of activities, including lectures, discussions, observations, as well as experimental activities. As a result, many students were found to be actively engaged throughout the activities and showed an improved attitude. They realized the importance of washing hands, keeping environmental hygiene, and problem-solving. Further evaluation indicated that the students had developed a better understanding of general environmental management. Therefore, it can be concluded that early environmental education can potentially transform students' knowledge and awareness about the importance of environmental conservation. Through these activities, it is expected that the obtained experiences will support the Gedong Songo environmental conservation in the future.

Abstract

Phosphorus is the most important key element in the nutrition of plants. Although P is abundant in soils, it is a major limiting factor for plant growth as it is in an unavailable form for roots uptake. Phosphate solubilizing bacteria (PSB) has ability to convert insoluble form of P to an available form. This study was aimed at screening and characterizing phosphate-solubilizing bacteria from manure and different rhizosphere and to ascertain a potential benefit to use mixed cultures to improve P solubilization. A total of 12 PSB colonies were isolated on Pikovskaya’s agar medium containing tricalcium phosphate. Out of 12 bacterial isolates, 2 isolates showed high phosphate solubilization index (2.17 and 1.83, respectively) were selected for further study. Based on the 16S rRNA gene sequence analysis, PSB3 was closely related to Burkholderia contaminans (99%), and PSB11 was closely related to Acinetobacter baumannii (99%). The mean P dissolved in liquid cultures of PSB3 and PSB11 in a 14-day incubation were 96.7 and 39.3 mg l-1, respectively. Mixed inoculation of B. contaminans PSB3 and A. baumannii PSB11 could not increase the solubilization activity significantly, suggesting there is antagonistic behavior of one isolate towards another. As the interaction of these two isolates may be antagonistic, co-inoculation of these bacteria for P solubilization is not recommended. However, further study is needed to confirm these results.

Abstract

The pathogenic fungi, such as Fusarium in the rhizosphere of tomato (Solanum lycopersicum) negatively affects the yield and quality of the plant. A number of biological control agents have been used for protecting tomato plants against wilt diseases including various fungal species. The objective of this study was to evaluate the antagonism effects of Trichoderma atroviride and T. harzianum against the pathogen Fusarium sp. associated with tomato wilt. In this study, the antagonism of these Trichoderma spp. against the Fusarium sp. was tested in vitro by the dual culture technique, and the percentage inhibition of radial growth (PIRG) and the antagonism reaction (scale 1-5) were evaluated. The results showed that T. atroviride and T. harzianum led to 70.8% PIRG and scale 1 antagonism reaction, and 40.6% PIRG and scale 3 antagonism reaction against Fusarium sp. associated with tomato wilt after 7 days of incubation, respectively. These results indicate that application of T. atroviride and T. harzianum may be promising approach for biological control of Fusarium wilt of tomato and may play an important role in sustainable agriculture.
