Abstract

Rennet, a milk coagulant exhibiting proteolytic activity, is a crucial component in cheese industries. Its price and availability have discouraged the growth of some small scale cheese industries. Therefore, an alternative for rennet will be beneficial for the industries. Among other sources, plant proteases offer some advantages as rennet alternatives. This study aimed to investigate the potential of plant proteases obtained from the latex as potential rennet alternatives. A total of six plants from the genus Ficus and Artocarpus were screened for their proteolytic activity and milk coagulating ability. The screening indicated that all six tested plants displayed proteolytic activity at various levels, but only Ficus benjamina and Artocarpus heterophyllus produced a firm milk curd. Hence, both F. benjamina and A. heterophyllus were determined to be the most potential. Further characterizations suggested that F. benjamina and A. heterophyllus protease were optimum at pH 7.0 also at 50°C and 40°C, respectively. At their optimum conditions, both proteases exhibited a lower MCA/PA ratio than that of the rennet. This study contributed to scientific knowledge development by becoming the first to characterize the optimum conditions of F. benjamina and A. heterophyllus’ proteases, investigate their MCA/PA ratio, and compare their activity against commercial rennet. The examination of their potentials as rennet alternatives could benefit small cheese industries and the communities.

Abstract

The medicinal plant-derived bioactive compounds have a potential for many biological activities, including antimicrobial activity. Artemisia cina is a medicinal plant from the Compositae family with the potential of having antitumor, antifungal, and antibacterial activity. This study aimed to determine the antibacterial activity and the flavonoid content of A. Cina’s ethyl acetate extract. Plants samples were extracted by ethyl acetate maceration method. Antibacterial activity was tested against Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus) by a disk diffusion method using 25, 50, and 100 mg/l extract concentrations. The flavonoid contents (quercetin and kaempferol) were measured using High-Performance Liquid Chromatography. The extracts of diploid and polyploid A. cina displayed some antibacterial activity, with the Gram-negative bacteria being more resistant than the Gram-positive counterpart. However, no significant difference was observed between the diploid and polyploid extracts. As for the flavonoid content, the highest quercetin content (0.5501 mg/ml) was found in the polyploid A. cina (J), while the highest kaempferol content (0.5818 mg/ml) was observed in the diploid A. cina (KJT). Although A. cina is widely grown in Indonesia, compared to other Artemisia species, A. cina has not been widely studied, especially its antibacterial  potential and in related to its flavonoid content and the use of ethyl acetate as the extraction solvent.  This study reveals the potential of A. cina as a natural antibacterial agent.

Abstract

Gedong Songo is a National Tourism Development Area in Bandungan region with an increasing number of visitors. Previous studies found that littering and water pollution were some of the prominent problems in Gedong Songo. The lack of knowledge and awareness towards environmental management was identified as the center of the issues. A conservation effort in the form of an early environmental education targeted elementary school students was suggested to address this problem. The environmental education was performed in State Elementary School Candi 1 and Candi 3 Bandungan and was designed as a series of activities, including lectures, discussions, observations, as well as experimental activities. As a result, many students were found to be actively engaged throughout the activities and showed an improved attitude. They realized the importance of washing hands, keeping environmental hygiene, and problem-solving. Further evaluation indicated that the students had developed a better understanding of general environmental management. Therefore, it can be concluded that early environmental education can potentially transform students' knowledge and awareness about the importance of environmental conservation. Through these activities, it is expected that the obtained experiences will support the Gedong Songo environmental conservation in the future.

Abstract

Saat ini, gerakan kampus hijau dan berkelanjutan mulai menjadi tren baru di berbagai universitas dalam dan luar negeri. Penelitian ini bertujuan untuk mendapatkan gambaran kondisi paduk berdasarkan kriteria limbah (WS) dari UI GreenMetric di Universitas Kristen Satya Wacana (UKSW). Penelitian ini menggunakan kuesioner UI GreenMetric dan wawancara terhadap kepala-kepala direktorat dan laboratorium yang bertanggungjawab terhadap pengelolaan limbah di UKSW, termasuk pihak ketiga yang ditugasi untuk mengelola limbah. Hasil studi menunjukkan bahwa UKSW memiliki program kampus hijau untuk mewujudkan kampus yang ramah lingkungan, UKSW telah mengimplementasikan 3R yang terintegrasi dengan pengelolaan lingkungan kampus dalam rangka mewujudkan lingkungan kampus yang lebih hijau dan berkelanjutan. UKSW direkomendasikan agar ke depan dapat mengimplementasikan strategi-strategi ekonomi sirkuler dalam pengelolaan limbah sehingga dapat mencapai tujuan pengelolaan dan fungsi lingkungan kampus berkelanjutan.

Abstract

Tempe merupakan produk pangan berbahan kedelai (Glycine max) yang difermentasi dengan melibatkan mikroba dari genus Rhizopus. Tempe memiliki berbagai kandungan gizi dan senyawa, salah satunya adalah antioksidan. Untuk meningkatkan kandungan antioksidan dalam tempe dapat dilakukan dengan penambahan bahan alami. Biji chia (Salvia hispanica) dipilih karena termasuk bahan pangan fungsional dan memiliki kandungan antioksidan. Tujuan penelitian ini untuk mengetahui efektivitas penambahan biji chia pada fermentasi tempe kedelai dalam peningkatan antioksidan dan nilai kesukaan. Penelitian dilakukan menggunakan RAL dengan satu faktor dan empat taraf perlakuan, yaitu konsentrasi penambahan biji chia 0%,  0,5%, 1% dan 1,5% (b/b). Penentuan aktivitas antioksidan dilakukan dengan metode DPPH, sedangkan uji hedonik dilakukan menggunakan kuesioner pada 15 orang panelis. Hasil pengukuran antioksidan menunjukkan penambahan biji chia pada tempe dapat meningkatkan aktivitas antioksidan yang dapat dilihat pada persen inhibisi. Persen inhibisi paling tinggi yaitu pada konsentrasi biji chia 1% yaitu sebesar 69,83%. Pada uji hedonik dengan uji Univariate dan uji lanjutan Duncan, perlakuan tempe tanpa pemberian bumbu menunjukkan tidak berbeda nyata antar konsentrasi perlakuan pada parameter aroma dan tekstur. Sedangkan perlakuan tempe dengan penambahan bumbu menunjukkan tidak berbeda nyata antar konsentrasi perlakuan pada parameter tekstur dan rasa.

Abstract

Keluarga merupakan penyangga masyarakat termasuk negara. Kesehatan dan ketahanan pangan keluarga menjadi faktor penting terbentuknya masyarakat yang sehat. Pembangunan di berbagai aspek kehidupan yang meliputi aspek sosial, politik, ekonomi dan juga kesehatan akan dapat terwujud apabila dilaksanakan melalui pemberdayaan masyarakat. Ibu-ibu PKK Harmony Residence mengalami kendala dala. upaya menerapakan pengadaan tanaman sayur (TS) dan tanaman obat keluarga (TOGA) di lingkungan rumah karena kurangnya pengetahuan tentang jenis dan manfaat tanaman serta cara menanam yang baik.Fakultas Biologi UKSW mengadakan kegiatan PkM dengan mitra PKK Harmony Residence, Sendangmulyo, Semarang. Kegiatan bertujuan untuk memberdayakan masyarakat mitra melalui pendampingan pembuatan pojok TS-TOGA sebagai upaya kesadaran kesehatan dan ketahanan pangan masyarakat. Kegiatan terdiri atas observasi dan identifikasi potensi serta permasalahan, persiapan dan sosialisasi kegiatan, sarasehan dan edukasi tentang kesehatan dan pojok TS -TOGA, pelatihan dan praktik pembuatan PTS-TOGA, pemantauan indikator ketercapaian program. Lebih dari 80% ibu-ibu PKK mengalami peningkatan wawasan mengenai jenis dan cara budidaya TS-TOGA, memotivasi untuk menerapkan gaya hidup sehat dan membuat TS-TOGA di lingkungan rumah masing-masing, Pojok TS-TOGA yang dibuat bermanfaat nyata untuk  mencukupi kebutuhan sayur dan obat alami. 75% ibu-ibu PKK menyatakan akan melanjutkan menjalankan program secara mandiri setelah kegiatan berakhir.

Abstract

The quality of a yogurt highly depends on the milk’s quality, the culture, and the incubation temperature. However, many home yogurt makers do not have access to fresh milk and incubator which may lead to subpar-quality yogurt. This research explored the potential of using Saccharomyces cerevisiae to improve yogurt quality when fermentation conditions are suboptimal. The experiment was conducted by inoculating ultra-high temperature (UHT)--sterilized milk with a 10% yogurt starter containing Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus, also various S. cerevisiae concentrations (0; 0.156; 0.625; 2.5; and 10%). The inoculated milk was fermented at 30 °C for 24 hours. Their sensory qualities were examined by the panelists. Their acidity, pH, and microorganism count were examined before and after incubation. S. cerevisiae addition at 2.5% displayed better taste and texture without discernable unpleasant aroma. These improvements might be due to the ethanol production by S. cerevisiae. S. cerevisiae addition was also found to slightly inhibit the growth of L. delbrueckii subsp. bulgaricus and S. thermophilus. However, the combined number of these probiotic microorganisms (2.7 × 108 CFU/mL) was still by the national standard.  S. cerevisiae addition potentially improves yogurt quality made from UHT milk incubated at lower temperatures.

Abstract

This activity aims to explore and improve the collaborative skills of students at SMA Kristen 1, Salatiga with class XII IPA students as participants. The data collection technique used a collaborative skills observation sheet with indicators 1) working productively; 2) being responsible; 3) respecting other students and; 4) compromising. The data analysis technique used the percentage of collaborative skills. In photosynthesis learning, students with collaborative skills in the exemplary category reached 14 people (19.4%), proficient 47 people (65.3%), basic 9 people (12.5%), and novice only 2 people (2.8%). In mitosis learning, students who had collaborative skills in the exemplary category were only 9 people (12.5%), proficient 52 people (72.2%), basic 8 people (11.1%), and novice only 3 people (4.2%). In the aspect of working productively in photosynthesis learning, the novice category reached 8 people (11.1%), while in mitosis learning the novice category was only 2 people (2.8%). In the aspect of being responsible for photosynthesis, the novice category reached 9 people (12.5%), while in mitosis learning, the novice category was only 4 people (5.6%). Furthermore, in photosynthesis learning in the aspect of flexibility and compromise, the novice category reached 8 people (11.1%), while in mitosis learning the novice category was only 7 people (9.7%). Based on these findings, it can be concluded that there has been an increase in the number of students who are increasingly skilled in collaborating with students.
