Abstract

ABSTRACTThe diversity of butterflies’ family of Nymphalidae and Pieridae at Cirengganis area and Cikamal Savannah area of Pananjung Sanctuary of PangandaranThe research reported the diversity o.f butterflies of Nymphalidae and Pieridae Families at Cirengganis area and Cikamal Savannah area which located at Pananjung Sanctuary, Pangandaran. The experiment was conducted using visual encounter survey method. The butterflies were caught using sweeping technique. The butterflies were collected and identified. Abiotic factors such light intensity and temperature were measured during observation. The result showed that 13 species of nymphalidae family were identified whilst only 2 species were identified belongs to the family of Pieridae. Butterfly diversity index at Cirengganis and Cikamal Savannah categorized as moderate with value of 2.285647. Vegetation that interacted with the butterflies were Tectona grandis), Psychotria aurantiaca, and Melastoma malabathricum. Furthermore, light intensity and temperature at Cirengganis were detected at 12473.33 lux and 28.6oC, respectively. While at Cikamal Savannah, the light intensity was measured at 26900 lux and temperature reached 33.6oC.Keywords: Visual encounter survey, Abiotic factor, VegetationABSTRAKPenelitian ini melaporkan mengenai keanekaragaman Jenis Kupu-Kupu Famili Nymphalidae dan Pieridae di kawasan Cirengganis dan Padang Rumput Cikamal Cagar Alam Pananjung Pangandaran. Metode yang digunakan adalah metode Visual Encounter Survey (VES). Penangkapan kupu-kupu dilakukan dengan teknik sweeping. Kupu-kupu yang diperoleh dikoleksi dan diidentifikasi. Faktor abiotik seperti intensitas cahaya dan suhu diukur selama pengamatan. Serta diamati pula vegetasi sekitar lokasi pengamatan yang berinteraksi dengan kupu-kupu. Hasil pengamatan diperoleh total kupu-kupu famili Nymphalidae sebanyak 13 spesies dan kupu-kupu famili Pieridae sebanyak 2 spesies. Nilai indeks keanekaragaman kupu-kupu di kawasan Cirengganis dan Padang Rumput Cikamal tergolong kategori sedang yaitu 2,285647. Hasil vegetasi yang diketahui berinteraksi dengan kupu-kupu yaitu jati (Tectona grandis), ki kores (Psychotria aurantiaca), dan harendong (Melastoma malabathricum). Sementara rata-rata intensitas cahaya dan suhu di Cirengganis sebesar 12473,33 lux dan 28,6oC dan di Padang Rumput Cikamal sebesar 26900 lux dan 33,6oC.Kata Kunci: Visual Encounter Survey, Faktor abiotik, Vegetasi

Abstract

Ulat krop (Crocidolomia pavonana) merupakan hama kubis potensial yang diketahui telah resisten terhadap insektisida sintesis. Metabolit aktif tumbuhan memiliki berbagai mekanisme bioaktivitas diantaranya berpotensi mencegah efek resistensi serangga hama. Satu diantaranya bioktivitas antifidan dengan mekanisme menghambat respon dan aktivitas makan serangga hama. Andrografolida merupakan senyawa aktif turunan diterpenoid yang diisolasi dari tumbuhan Sambiloto (Andrographis paniculata), sifat antifidannya telah diteliti terhadap beberapa hama kubis ordo Lepidoptera terkecuali C. pavonana. Berdasarkan hal tersebut, senyawa andrografolida diujikan terhadap larva C. pavonana instar-3 untuk mengetahui efek antifidan dan konsentrasi minimum efektifnya. Eksperimen uji hayati dilakukan menggunakan Rancangan Acak Lengkap dengan 6 taraf konsentrasi (0 (kontrol), 500, 1000, 1600, 2500, 4000, dan 6500 ppm) dan 4 pengulangan. Pengujian menggunakan metode daun celup diuji dengan uji antifidan pilihan (choice) dan tanpa pilihan (no-choice). Parameter ukur adalah rata-rata luas daun yang dimakan larva uji, selanjutnya perbandingan jumlah rata-rata luas daun perlakuan dan kontrol dianalisis dengan uji non parametrik Mann-Whitney U (P<0,05). Hasil uji antifidan menunjukkan andrografolida bersifat antifidan secara nyata yang ditunjukkan oleh rata-rata luas daerah makan pada daun perlakuan yang lebih kecil dibandingkan kontrol ditiap-tiap konsentrasi uji baik melalui uji pilihan maupun tanpa pilihan (P<0,05). Konsentrasi 1000ppm, merupakan konsentrasi minimum efektif pada uji pilihan dan tanpa pilihan yang telah bersifat antifidan terhadap larva instar-3 C. pavonana.

Abstract

The Helicoverpa armigera Nuclear Polyhedrosis Virus (HaNPV1) is a subculture derived from the original HaNPV, and it has been cultivated in Spodoptera litura larvae as an alternative host. HaNPV1 was subsequently formulated using gypsum and talcum as carrier media. Following this formulation, a bacterial contamination test was conducted to assess the quality of the viral formulation.  The experiment was arranged in the randomized factorial block design (RFBD) with 2 replications. The viral formulations was stored for 16 weeks and the samples were taken every two weeks for contamination analysis. The data was then analyzed with the analysis of variance (ANOVA) and a post-hoc using Duncan’s Multiple Range test. The variable observed was the number of the bacterial colonies cultivated on the specific media i.e., Nutrient Agar (NA), Salmonella Shigella Agar (SSA) and Eosin Methilen Blue Agar (EMB). The results showed that the bacterial contaminants was detected from 0 to 12 weeks of storage time. However, the highest contamination was found in viral formulation after 8 weeks of storage time and the highest bacterial contaminations were recorded from all viral formulation tested in NA. The results indicated that the bacterial contamination were found around 1.45 × 109 cfu/gram and 1.97 × 109 cfu/gram in gypsum and talcum formulations, respectively. On SSA and EMB media, the bacteria contaminants from all formulation found in 8 weeks of storage time, but Salmonella, Shigella, or Escherichia coli (aspathogenic bacteria) were not found. After 12 weeks storage time, there was no indication of  contamination found in all media. Furthermore, Bacillus species was found as a most dominant contaminant in all samples. In conclusion, although the viral formulations using gypsum and talc were not contaminated by pathogenic bacteria such Salmonella, Shigella, or E. coli. Nevetherless, the viral formulation was still easily contaminated by other non-pathogenic bacterial species. Thus, a more standardized and stricted strategy needs to be developed for a better viral formulation product.
