Abstract

Background: A good collection of fungi culture can be maintained with good preservation methods. With appropriate methods, preservation can be performed in low-budget laboratories, such as with periodic subculture and preservation in either sterile water or in paraffin oil. The aim of this study was to assess the viability, morphology, contamination, and antifungal susceptibility profile of fungi preserved in sterile water or paraffin oil compared to a periodic subculture.Methods: This study was an experimental study using Aspergillus sp., Trichophyton mentagrophytes and Candida albicans. The fungi were preserved by a periodic subculture or in sterile water or in paraffin oil for six months. A comparison of viability, morphology, contamination, and antifungal susceptibility profile between methods were made before and after six months of study.Results: The viability of all the fungi was maintained in both periodic subculture and preservation with paraffin oil. However, the T. mentagrophytes was not viable in preservation using sterile water. All methods could maintain the morphology without contamination. There were reductions in the inhibition zone against antifungal drug in preservation with sterile water and paraffin oil.Conclusion: Preservation in paraffin oil is superior to preservation in sterile water in terms of maintaining fungal viability. This method is suitable for the use in a simple laboratory, albeit considerations should be given to the reduction of inhibition zone.

Abstract

BACKGROUND Women with HIV/AIDS (WLWHA) have a complex psychosocial burden and a tendency to negative self-esteem, possibly resulting in mental and emotional problems. They need family support to deal with the HIV/AIDS infection and its psychosocial burden. The purpose of this study was to determine chacteristics of family support, self-esteem, and depression of WLWHA and the relationship between family support and self-esteem and depression. METHOD This was a cross-sectional study of 99 WLWHA infected through their husbands/partners, with no history of drug abuse. The data was taken by a consecutive sampling of two proportions test at Dharmais Cancer Hospital from November 2013 – January 2014. The instruments comprised a demographic questionnaire, the Rosenberg Self-Esteem questionnaire, the Hamilton Depression Rating Scale (HDRS), and a family support questionnaire. The data was analyzed by binary logistic regression. RESULTS There were 99 respondents with mean age of 36 years, of whom 44.4% were high school graduates, 54.5% unemployed, and 91.9% had HIV/ AIDS for more than a year. Binary logistic regression analysis showed no significant relationship between family support and self-esteem (p=0.700) and depression (p=0.396). Good family support has a protective effect of 1.3 times (OR=0.772; 95%CI: 0.138-3.770) towards increasing self-esteem, whereas poor family support increases the risk of depression 1.5 times (OR=1.477; 95%CI: 0.598-3.645) in WLWHA infected with HIV/AIDS from their husband/partner. CONCLUSIONS Good family support tend to have a protective effect towards increasing self-esteem, whereas poor family support increases the risk of depression in WLWHA infected with HIV/AIDS from their husband/partner.

Abstract

Latar belakang: Tuberkulosis (TB) merupakan salah satu masalah kesehatan masyarakat yang penting di dunia, sepertiga penduduk dunia telah terinfeksi kuman TB. Dalam upaya menanggulangi penyakit TB, dibutuhkan diagnosis yang dapat ditegakkan melalui pemeriksaan bakteriologi, radiologi, dan pemeriksaan penunjang lain, seperti pemeriksaan nilai laju endap darah (LED). Infeksi TB berpengaruh terhadap peningkatan nilai LED dan pembentukan lesi radiologi pada TB Paru. Tujuan: untuk mengetahui korelasi nilai LED dengan gambaran luas lesi radiologi toraks pada pasien TB Paru di RS Atma Jaya Jakarta. Metode: Desain dalam penelitian ini adalah korelatif analitik dengan pendekatan cross-sectional. Sampel penelitian ini yaitu pasien TB paru dewasa baru aktif yang melakukan pengobatan di RS Atma Jaya sebanyak 68 pasien yang diambil dengan teknik consecutive sampling. Pengolahan data menggunakan metode korelasi Pearson dengan interval kepercayaan 95%, dilanjutkan dengan metode analisis regresi linear. Hasil: Penelitian menunjukan hubungan yang bermakna antara nilai laju endap darah (LED) dengan luas lesi radiologi toraks dengan koefisien korelasi 0,452  (p = 0,0001), artinya mempunyai kekuatan korelasi sedang. Hasil dari faktor perancu usia (p=0,906), jenis kelamin (p=0,887), dan gula darah sewaktu (p=0,056) tidak bermakna terhadap hubungan nilai LED dengan luas lesi radiologi toraks. Kesimpulan: Penelitian ini menunjukkan adanya hubungan bermakna antara nilai LED dengan luas lesi radiologi toraks pada pasien TB paru di RS Atma Jaya.
