Abstract

Polyhydroxyalkanoates (PHAs) are intracellular material accumulated by several bacteria. Commercial production of PHAs faces the issue of high production cost especially substrate cost and recovery/separation cost. An alternative to reduce the production cost is to use enzyme and or chemical to recover PHAs from bacterial cells. Recovery of PHAs from bacterial cells was done using enzyme, chemical, and a mixture of enzyme and chemical. Enzyme (s) and or chemical(s) were added into culture broth to disrupt cells after adjusting pH and temperature of the culture broth. Treatment by adding enzyme or chemical only into culture broth showed a low level of PHAs recovered from bacterial cells. Treatment by adding a mixture of enzymes and chemicals showed the best result among 22 examined combinations, i.e. a mixture of EDTA, lisozyme, papain enzyme, and SDS. This combination gave a PHA recovery of 65 % w/w.

Abstract

ABSTRAKPengobatan regeneratif menggunakan Media Terkondisi Sel Punca Mesenkimal (MTSPM) merupakan salah satu metode yang dikembangkan untuk pengobatan Diabetes Melitus (DM). Pengembangan metode produksi MTSPM perlu dilakukan untuk meningkatkan efikasi, salah satunya dengan menginduksi sekresi dari sel punca mesenkimal (SPM) menggunakan senyawa penanda dari penyakit DM yaitu Interleukin-6 (IL-6). Penelitian ini bertujuan untuk menguji efek dari penambahan IL-6 pada media kultur terhadap kultur SPM dan konsentrasi protein pada MTSPM yang dihasilkan. Kultur SPM dilakukan pada temperatur 37˚C dan 5% CO2. Kultur SPM dibagi menjadi tiga kelompok yang diberi media tumbuh berbeda, kelompok I dikultur pada media basal, kelompok II dengan media basal + 1pg/mL IL-6, dan kelompok III dengan media basal +10pg/mL IL-6. Pengujian efek IL-6 dilakukan dengan melakukan penghitungan jumlah SPM pada kultur dan pengukuran total protein pada MTSPM menggunakan metode Qubit Fluorometer. Hasil pengukuran dianalisa secara statistik dengan uji One Way Anova, Kruskall Wallis dan Mann Whitney. Hasil pengujian menunjukkan pemberian IL-6 mampu meningkatkan jumlah sel pada kultur SPM. Total protein pada MTSPM mengalami peningkatan hingga 65% dan 262% untuk 24 jam pertama dan kedua ketika diberi penambahan IL-6 dengan konsentrasi 10pg/mL. Hal ini menunjukkan terdapat lebih banyak senyawa sitokin dan faktor tumbuh dalam MTSPM yang dihasilkan sehingga terdapat kemungkinan terjadinya peningkatan efikasi dari MTSPM tersebut saat dipergunakan dalam terapi DM. Tetapi hal tersebut perlu diteliti lebih lanjut dengan melihat profil sitokin dan faktor tumbuh dalam MTSPM dan juga dilanjutkan dengan uji in vivo pada hewan model DM. Kata Kunci: Sel Punca Mesenkimal, Interleukin-6, Media Terkondisi. ABSTRACTRegenerative medicine using Conditioned Medium-Mesenchymal Stem Cell (CMMSCs) is one of the developed methods for the treatment of Diabetes Mellitus (DM). However, the method to produce CMMSCs still needs to be developed to increase its efficacy, one of which is by inducing secretion from mesenchymal stem cells (MSCs) using marker compounds in DM, such as Interleukin-6 (IL-6). This study aims to examine the effect of Interleukin-6 (IL-6) in the growth medium on the culture of MSCs and the concentration of protein on CMMSCs. The MSCs were cultured at 37oC and 5% CO2. Culture of MSCs grouped into three which were given different growth mediums, group I cultured in basal medium, group II cultured in basal medium + 1pg/mL IL-6, and group III cultured in basal medium +10pg/mL IL-6. Tests were carried out by calculating the amount of MSCs in culture and measuring the total protein on CMMSCs using the Qubit Fluorometer method. Results were analyzed statistically using the One Way Anova test, Kruskall Wallis, and Mann Whitney test. The results showed that the addition of IL-6 was able to increase the number of cells in the MSCs culture. Total protein in CMMSCs increased up to 65% and 262% for the first and second 24 hours when given the addition of IL-6 at a concentration of 10pg/mL. This shows that there are more cytokine compounds and growth factors in the resulting CMMSC, therefore there is a possibility of an increase in efficacy from the CMMSC when used to treat DM. However, it needs further research to evaluate the cytokine and growth factor profile of the CMMSC and followed up with an in vivo trial using the DM animal model. Keywords: Mesenchymal Stem Cells, Interleukin-6, Conditioned Medium.

Abstract

This study was conducted to find out the usability of books acquired by the Library of Sepuluh Nopember Institute Surabaya in 2010/2011. The study analysed data of the books usability, the novelty of the books for citations, and the most frequent publishers. The study used both quantitative and qualitative analysis. The results showed that the usability of the books (both for onsite reading and loan) was 4.04 times/book/5 years. Therefore it concludes that the books were used frequently. Books that were mostly used were the most current collections in three years under the subject of engineering published by Wiley, Cambridge and Pearson.

Abstract

Pergeseran pola hidup di dalam masyarakat yang saat ini cenderung back to nature menyebabkan semakin meningkatnya permintaan obat herbal organik. Namun demikian, budidaya tanaman obat secara organik dan pengelolaannya secara professional masih jarang dilakukan. Kota Batu, Malang Jawa Timur terkenal sebagai agropolitan. Pengabdian masyarakat ini bertujuan untuk mengembangkan kebun bibit herbal organik yang berfungsi sebagai area pembibitan tanaman herbal dan agrowidyawisata di Desa Oro-Oro Ombo Kota Batu, Malang. Pengabdian masyarakat dilakukan bersama mitra Kelompok Tani Hutan (KTH) Panderman. Adapun metode yang dilakukan berupa penggalian informasi lokal dan pendampingan pengembangan kebun bibit herbal. Hasil pengabdian menunjukkan bahwa Desa Oro-Oro Ombo, Kota Batu Malang merupakan kawasan potensial untuk budidaya tanaman herbal. Diharapkan dengan adanya kegiatan ini, partisipasi masyarakat desa terhadap pengembangan budidaya herbal organik semakin meningkat. Sehingga hal ini dapat berdampak positif pada peningkatan ekonomi masyarakat desa.

Abstract

This study aims to explore challenges and learning behavior of children at Desa Balerejo, Kecamatan Pangungrejo, Blitar, in welcoming ICT introduction program. Education is the heart of community development. The creation of learning community and its’ development is sustained by community education, including its’ young generation education. Children are the future of the community. They will be the next leaders and their community developers. Thus, providing sufficient education is vital for assuring certain community existence and development sustainability. However, in reality, not all children can pursue equal education. Education in some remote areas is still left behind. Consequently, children in rural areas, are frequently suffering from educational disparities. The data are collected through direct observation during the community service program, through our roles as community service program providers as well as the instructors. The study is the result of our reflection, what happens in the field and what can be improved for future empowerment programs. The study indicates that there are several substantial factors determine and pose challenges for the effectiveness of ICT education for children at rural area. The first factor is culture and social context. Submissive and collectivism cultural orientation may affect the children’s behavior. The second aspect is the teaching approaches and learning psychology of children. Teaching method and materials should be adapted to the children, what they like and how they may learn best. The last factor is relating the nature of the empowerment program, which should be executed collaboratively and simultaneously with other programs.
