Abstract

ABSTRACT The effect of mahkota dewa polyphenol in the cell proliferation and apoptosis in BaP induced Balb/c ratsBackground: The polyphenol of mahkota dewa (Phaleria macrocarpa [Scheff.] Boerl) has the potency as antioxidant and anticancer which can handle free radicals, but there has not been extensive research on this. This research is aimed to prove the role of mahkota dewa polyphenol in the cell proliferation inhibition and induct lung carcinogenesis apoptosis in strain Balb/c mice which inducted with benzo(a)pyrene (BaP).Methods: Posttest control group design was carried out among 40 strain Balb/c mice sample, aged 1-2 weeks, weighed 20-30 grams, healthy mice condition. All mice were inducted with BaP and then randomized into 2 groups, as control group and the treatment group. The development of the lung tumor was observed by tissue surgery in the 8th, 17th and 26th week. The data collected were AgNORs, and IHC-TUNEL-apoptosis index dying. The data analysis was conducted using Kruskal-Wallis, Mann-Whitney, One-way ANOVA, and Post hoc test LSD with significance degree of p<α (0.05).Results: The oral administration of mahkota dewa polyphenol showed significantly decreasing cell proliferation, increasing apoptosis index in treatment group in week 8, 17 and 26 (p=0.000). Carcinogenesis incidence for the treatment group week 8 and 26 were 2.32±0.26 and 3.93±0.46, while for the treatment group were 1.88±0.38 and 0.88±0.22 (p=0.000). The cell proliferations for control group week 8 and 26 were 1.57±0.12 and 2.29±0.15, while for the treatment group were 1.53±0.11 and 1.60±0.04 (p=0.000). Apoptosis index for the control group for week 8 was 0.00±0.00 and 0.92±0.22 in week 26, while the treatment group was 1.12±0.71 and 2.02±1.05 (p=0.000).Conclusion: The administration of mahkota dewa polyphenol effectively inhibited the cell proliferation activity and increased apoptosis measured by apoptosis index. Therefore polyphenol has anticancer and antioxidant activities which can inhibit lung carcinogenesis in Balb/c mice.Keywords: Polyphenol, mahkota dewa (Phaleria macrocarpa [Scheff.] Boerl), apoptosis index, strain Balb/c mice, benzo(a)pyrene (BaP).ABSTRAKLatar belakang: Polifenol mahkota dewa (Phaleria macrocarpa [Scheff.] Boerl) berpotensi sebagai antioksidan dan antikanker yang mampu menangkap radikal bebas, namun belum banyak diteliti. Penelitian ini bertujuan membuktikan peran polifenol mahkota dewa dalam menginhibisi proliferasi sel dan menginduksi apoptosis pada mencit strain Balb/c hasil induksi Benzo(a)pyrene (BaP).Metode: Posttest control group design dengan sampel 40 mencit strain Balb/c, umur 1-2 minggu, berat 20-30 g, kondisi mencit sehat. Semua mencit diinduksi BaP kemudian hewan dirandomisasi menjadi 2 kelompok yaitu kelompok kontrol dan kelompok perlakuan polifenol 50 mg. Perkembangan tumor paru diamati dengan pembedahan jaringan pada minggu ke-8, 17 dan 26. Data dikumpulkan meliputi AgNORs, pengecatan IHC-TUNEL-indeks apoptosis. Analisis data dengan Kruskal-Wallis, Mann-Whitney, One-way ANOVA, post hoc test LSD dengan derajat kemaknaan p<α (0,05).Hasil: Pemberian oral polifenol mahkota dewa sebesar 50 mg secara bermakna memperlihatkan penurunan insidens karsinogenesis paru, proliferasi sel, protein Bax dan peningkatan indeks apoptosis, protein p53, Bcl-2, ekspresi caspase 3, 8, 9 pada kelompok perlakuan pada minggu ke-8, 17 dan 26 (p=0,000). Insidens karsinogenesis untuk kelompok kontrol minggu ke-8, dan 26 sebesar 2,32±0,26 dan 3,93±0,46, sedangkan kelompok perlakuan sebesar 1,88±0,38 dan 0,88±0,22 (p=0,000). Proliferasi sel untuk kelompok kontrol minggu ke-8 dan 26 sebesar 1,57±0,12 dan 2,29±0,15, sedangkan kelompok perlakuan sebesar 1,53±0,11 dan 1,60±0,04 (p=0,000). Indeks apoptosis pada kelompok kontrol minggu ke-8 sebesar 0,00±0,00 dan 0,92±0,22 minggu ke-26, sedangkan kelompok perlakuan sebesar 1,12±0,71 dan 2,02±1,05 (p=0,000).Simpulan: Pemberian polifenol mahkota dewa efektif meng-inhibisi proliferasi sel dan menginduksi apoptosis melalui peningkatan indeks apoptosis. Jadi polifenol mahkota dewa memiliki aktivitas antikanker dan antioksidan mampu menghambat karsinogenesis paru mencit Balb/c.

Abstract

Background: Clove is a spice native plants Maluku Islands, and has been traded and cultivated for generations in the form of people's plantation. Separation of chemical constituents of clove showed that clove leaves contain saponins, alkaloids, flavonoid glycosides and tannins. Flavonoids are one of the types of compounds that are toxic / residues, the sugar bound compound from the flavonoids. Flavonoids have distinctive properties that is a very sharp odor, bitter taste, soluble in water and organic solvents, and easily decompose at high temperatures. Methods: This study was conducted in February-April of 2015 at the Laboratory and Greenhouse biology education Pattimura University. This research is a lab experiment. Clove leaf extract obtained by polar solvent is macerated with ethanol 96% and then applied to the growth of weeds sedges. Clove leaf extract concentration used a 5%, 10%, 20%, 50% and control. Results: There is a natural herbicide effectiveness of clove leaf extract at a concentration of 50%, which can be used as an alternative to inhibit the growth of tall weeds sedges. Conclusion: The higher the clove leaf extract given against weeds sedges will increasingly serve to inhibit weed sedges as measured by plant height, phytotoxicity, fresh weight and dry weight of weeds sedges.

Abstract

Background: Mangrove forests are one of the unique natural ecosystems with high ecological value. Mangrove ecosystems are one of the areas with high productivity because there are litter and decomposition of litter and detritus occurs. This study was conducted to determine the association between gastropods and mangrove plants. Method: Research on gastropod association with mangrove plants in the coastal waters of Tiouw Village and Haria Village was conducted in January 2018. Results: This study found from gastropods 6 species namely variabilis strombus, microurceus strombus, Nassarius luridus, Strombus urceus, Cypraea annulus, Strombus, Strombus, Nerita squamulata. From the mangrove plants, 10 species were found: Avicennia eucalyptifolia, Avicennia lanata, Bruguiera sexangula, Rhizophora apiculata, Bruguiera hainessi, Aegiceras floridum R, Aegiceras curniculatum L, Bruguiera palviflora, Bruguiera exaristafa, Ceriops tagal. Conclusion: Association values ​​ranged from 0.741-0.9773 with positive and negative types. This means that both types of seagrass are often found together or not found together in the observation location

Abstract

Background: Analysis of the level of storage of Carbon compounds (C-Stock) on mangrove forest vegetation based on differences in substrate on Saparua Island, Central Maluku Regency. This study aims to determine the amount of carbon content and the difference in carbon content stored in mangrove forests. Method: This study used a survey method through direct observation in the field and analysis of sample weight in the laboratory. The data obtained were analyzed using the biomass estimation formula and carbon content and statistical equation to determine the difference in carbon content at each station. Results: From the results of the study obtained total carbon in station I was 3.498351 tons / ha, station II was 0.180750 tons / ha and station III was 16.727136 tons / ha. Conclusion: With the difference of each station based on the BNT Test with a confidence level of 99% showing the value of the difference of 20.58, while the BNT test of the confidence level of 95% shows the difference in value of 13.59.

Abstract

Madarasah Aliyah BPD Iha-Kulur is one of the schools at the high school level under the Ministry of Religion but the pattern of development refers to the Ministry of National Education. therefore these schools are required to comply with various regulations regulated by the Ministry of National Education. The purpose of this study was to determine the effect of the availability of biology learning facilities on students' motivation and learning achievement. This study uses a type of quantitative research. Located at Madrasah Aliyah BPD Iha Kulur. On december 2020 - February 2021. Population in this study were students of class XII IPA totaling 110 and only taking a sample of 21% of the total population. Sampling technique using probability sampling technique. The instrument used in this study was a questionnaire, while the data analysis used multiple regression analysis. The show that the availability of biology learning facilities has a positive and significant influence on students' motivation and learning outcomes. The availability of biology learning facilities greatly determines student learning outcomes.

Abstract

The main objective of this study is to analyze students' scientific creativity in each domain with an essay test referring to the Torrance Tests of Creative Thinking. This research was conducted on 50 teachers and 500 students in 10 elementary schools in Cluster 2, Nusaniwe District, Ambon City. The results of this study report descriptive demographic results of respondents and the level of creativity, namely fluency, flexibility, originality and elaboration among students is very good. The results of the study as a whole show that most of the students are at a good level after being taught with the PjBL model. Students increase their creativity with the teacher's instructions and directions. Initially in detail, many students lack the dimensions of fluency, flexibility, originality and elaboration so they cannot provide more ideas, and cannot provide answers. So that the originality becomes less and they have no ideas to elaborate further than the answers given. However, based on the findings from several student meetings, there has been a significant increase. This is because it applies a problem-oriented learning model that enhances each realm of creativity in science subjects. In addition, the researcher also recommends conducting more in-depth research on creative thinking processes among students based on the local advantages of each school based on geographical location which has its own unique value. Therefore, teachers are expected to be able to better understand cognitively, affectively and psychomotor in students' minds and possibly improve students' creative thinking skills on each dimension of creativity

Abstract

One of the roles of the teacher in implementing an education is by using a learning starts with a question method. The learning starts with a question method is a method in which students are directed to learn independently by making questions based on the reading provided by the teacher. This study aims to see the effect of applying learning starts with a question on photosynthesis material on students' cognitive learning outcomes, critical thinking, and scientific work in class VIII SMP Negeri 15, SMP Negeri 7, SMP Kristen Kusu-Kusu, SMP Negeri 11, and SMP Negeri 4. Research data was analyzed quantitatively using descriptive and inferential statistics in the form of anacova analysis to see the effect of applying learning starts with a question on students' cognitive learning outcomes, critical thinking, and scientific work. The results of this study indicate that there is an effect of the application of the learning starts with photosynthesis material on students' cognitive learning outcomes, critical thinking, and scientific work.
