Abstract

This study aimes of determine the effect of mutagen Ethyl Methane Sulfonate (EMS) on upland local rice (Oryza sativa L.) varieties at generation M-1. This research has been carried out at the Department of Agrotechnology Greenhouse, Faculty of Agriculture, University of Riau, in November to December 2013. The research used factorial Completely Randomized Design consisting of two factors, namely two local rice varieties and five concentrations of mutagens EMS and four replications. The mean deviation between treatments tested by Duncan New Multiple Range Test (DNMRT) at the level of 5%. Parameters observed were time of first tiller appeared, lamina length, midrib length, seedling height, number of leaves, number of tillers, number of life seedling, the number of variegate seedling. Results indicated that the mutagen EMS at a concentration of 1%, reduced the number of first seedling appear, decreasing plant height, number of tillers and total life seeds.   Keywords: Upland rice, Ethyl Methane Sulfonate.

Abstract

Two cultivars of local rice, namely Mentik Wangi and Mentik Susu, have been grown around west to south flank of Merapi volcano. They are highly valued for their good taste and cooking characteristics. An attempt to introduce drought-tolerant rice varieties has been conducted by crossing them with two sources, namely Kasalath and Bluebonnet. Molecular-assisted selection using a set of SSR markers was applied to check the F1 and F2 generation trueness and segregation. This research was conducted in the facilities of Department of Agronomy, Faculty of Agriculture, Universitas Gadjah Mada, Yogyakarta. The research was a field experiment arranged in a completely randomized design, consisting of Mentik Wangi, Mentik Susu, Bluebonnet, and Kasalath, as well as their crossings’ F1 and F2 plants as treatments. The SSR markers used are RM72, RM228, RM518, and RM20(A). Polymorphism test of the parents showed that polymorphism exists between local and donor parents, thus these markers were considered eligible for the F1 and F2 tests. The heterozygous individuals of F1 were 75% for Bluebonnet × Mentik Wangi, 44.4% for Bluebonnet × Mentik Susu, and 46.7% for Mentik Wangi × Bluebonnet; all were consistent in every primer used. We could not confirm that the F2 populations showed segregation pattern that followed Mendelian segregation in some crosses due to too small size of the sample. Heterozygous individuals in F2 showed the differences pattern for each marker, indicating that the location of the SSR markers were far from each other in the genome.

Abstract

Poliembrioni merupakan pembentukan lebih dari satu embrio yang terjadi pada biji yang dikecambahkan. Terjadinya poliembrioni akibat zigot yang terpecah, perkembangan satu atau lebih sinergit, dan terdapat satu atau lebih kantung embrio per inti sel, dan variasi benuk adventif dari embrio. Tujuan penelitian untuk mengkaji pembentukan benih poliembrioni pada buah jeruk peras dan mempelajari pertumbuhan bibit dari benih poliembrioni. Berdasarkan pengamatan terhadap beberapa buah jeruk peras didapat bahwa rerata presentrase pembentukan benih poliembrioni pada setiap buah jeruk peras yaitu 47,92%. Adapun pertumbuhan bibit jeruk asal benih poliembrioni saat berumur 4 minggu yaitu tinggi tanaman rata-rata sebesar 3,075 cm, diameter biji 0,111 cm, jumlah daun 2 helai, luas daun 2,21 cm2, berat kering bibit 0,34 g, dan vigor hipotetis sebesar 7,76. Bibit jeruk yang tumbuh dari embriozigotik umumnya memiliki tampilan yang berbeda dari induknya karena gen yang berasa dari dua tetua. Namun bibit yang tumbuh dari embrio nuselar memiliki tampilan yang sama dengan induk betina akibat diferensiasi sel somatik.

Abstract

The use of liquid silica nutrients under salt stress can increase plant resistance through cell wall thickening which will also affect to increase productivity. The study aims to examine the effect of liquid silica nutrients to plant growth analyze and rice productivity affected by salt stress (NaCl). The experiment used a factorial completely randomized design. The first factor was NaCl concentrations consisted of 0 dS m-1, 4 dS m-1, 8 dS m-1, and 12 dS m-1. The second factor was application of liquid silica nutrients consisted of 0.2 mL L-1, 0.4 mL L-1, and 0.6 mL L-1. The treatments were repeated three times. Increasing the NaCl concentration to 12 dS m-1 when the plants were 4 weeks after planting caused a decrease in leaf area index. The addition of liquid silica nutrients with concentration up to 0.6 mL L-1 under salted conditions led to an increase in root shoot ratio, and the addition of liquid silica nutrients with concentration of 0.6 mL L-1 in salted conditions of 4 dS m-1 caused an increase in plant growth rate. The results showed that the leaf area index, leaf area ratio, root shoot ratio, and plant growth rate were positively correlated with plant biomass. Furthermore, the root shoot ratio had a positively correlated with productivity, the harvested index had a positively correlated with the number of productive tillers, percentage of filled grains, and rice productivity. However, the net assimilation rate and harvested index were negatively correlated with plant biomass.

Abstract

Local rice is a very important germplasm in the plant breeding program of superior rice. Jambi is one of the provinces in Indonesia which has a high diversity of local rice. This high diversity includes the diversity of physical characters of the shape and size of grain. This study aims to look at the physical diversity of lokal Jambi rice grains found in Ture Village. Research on the diversity of physical characters of Jambi local rice was conducted in Ture Village, Pemayung District, Batanghari Regency, Jambi Province. The local rice observed were Putih, Layap, Sereh wangi, Sekempol, Putih Kuning, Jarum Mas, Nak Daro and Ketan Hitam. The physical characters of grain that were observed were grain length, grain width, grain shape, rice color. The results showed that Sekepol was a short type of rice, Ketan Hitam and Jarum  Mas were a long rice and the other cultivars were a medium type. Based on the shape of the rice, the Sereh wangi, Jarum  Mas and  Ketan hitam  have a slender shape while the other cultivars are oval. The color of Ketan hitam is red and the other cultivars are white. Keywords : germplasm; local rice; physical characters; grain shape

Abstract

Cekaman garam merupakan cekaman abiotik yang mampu menghambat pertumbuhan dan produksi padi melalui mekanisme metabolisme. Usaha yang dapat dilakukan untuk membudidayakan tanaman padi dalam kondisi cekaman garam yaitu mengetahui karakteristik suatu varietas padi agar dapat tumbuh dan berkembang optimal. Tujuan penelitian untuk mengkaji karakteristik pertumbuhan dan hasil dua varietas padi tercekam garam NaCl. Rancangan Acak Lengkap faktorial digunakan dalam penelitian ini. Faktor pertama konsentrasi NaCl yang terdiri atas dua taraf yaitu 0 dS m-1 dan 4 dS m-1. Faktor kedua varietas padi yang terdiri atas dua taraf yaitu varietas Dendang dan varietas IPB 4S. Berdasarkan hasil penelitian menunjukkan bahwa padi varietas Dendang memiliki jumlah anakan (JA), panjang akar (PA), jumlah malai per rumpun (JMpR), persentase gabah isi (GI), dan bobot kering tajuk (BKT) yang lebih besar dibandingkan dengan padi varietas IPB 4S. Namun, padi IPB 4S memiliki karakteristik tanaman yang lebih tinggi dibandingkan padi varietas Dendang. Pemberian konsentrasi NaCl 0 dS m-1 menghasilkan jumlah anakan dan akar lebih panjang dibandingkan pemberian konsentrasi NaCl 4 dS m-1. Hasil analisis korelasi menunjukkan bahwa beberapa parameter berkorelasi positif, yakni panjang akar terhadap jumlah anakan (R= 0,89), bobot kering tajuk terhadap jumlah anakan dan panjang akar (R= 0,89; R= 0,91), dan persentase gabah isi terhadap bobot kering tajuk, panjang akar, jumlah malai per rumpun, dan jumlah anakan (R= 0,87; R= 0,76; R= 0,83; R= 0,89). Penggunaan varietas padi berpengaruh nyata terhadap karakteristik pertumbuhan dan hasil yang terlihat pada parameter JA, PA, JMpR, GI, dan BKT. Sedangkan konsentrasi NaCl berpengaruh nyata terhadap karakter tinggi tanaman.

Abstract

Pengabdian kepada masyarakat ini difokuskan pada pendampingan dan pelatihan peningkatan keterampilan sambung pucuk dalam pembuatan bibit tanaman alpukat oleh santri Pondok Pesantren Darul   Ulum II. Diseminasi ini dilatar belakangi oleh pemanfaatkan waktu luang yang masih kurang optimal oleh santri dan penyediaan bibit alpukat unggul untuk ditanam di lahan pondok yang belum dimanfaatkan secara optimal. Tujuan yang ingin dicapai pada kegiatan ini adalah  menambah  pengetahuan  dan  keterampilan  santri  mengenai  pembuatan  bibit  unggul dengan teknik sambung pucuk, serta menyediakan bibit alpukat unggul untuk optimalisasi penggunaan lahan di pondok pesantren. Metode yang digunakan adalah ceramah dan pelatihan. Materi-materi terkait pelatihan disampaikan melalui ceramah dan diskusi. Kemudian dilanjutkan dengan praktik secara langsung. Hasil dari diseminasi ini adalah santri dan masyarakat pondok pesantren memiliki pengetahuan dan keterampilan dalam pembuatan bibit alpukat dengan teknik sambung pucuk dan tersedianya bibit alpukat unggul untuk ditanam oleh pondok pesantren. Kesimpulan  dari  kegiatan  ini  adalah  kegiatan  diseminasi  pembuatan  bibit  alpukat  unggul  ini menjadi wawasan dan keterampilan baru bagi santri dan masyarakat pondok pesantren untuk dapat membuat bibit tanaman buah unggul. Abstract
