Abstract

Aim of this study was to examine the characteristics cytology of mucous cell-,temperature- and pH vagina during estrous cycle in local sheep.  31local sheep were synchronized with vaginal sponge consist of 20  mg  progesterone hormone before carried out observations of cytology of cells from the vaginal mucose through vaginal swabs, temperature and pH of the vagina.  Vaginal swabs were collected daily at 7 am for a weeks.Vaginal temperature and pH measurement is carried out twice a day, at 07.00 am and 15.00 pm for a weeks after vaginal swabs. Smears of the swab were then prepared on glass slide and they were stained with Giemsa.  Vaginal epithelial cells; Parabasal, intermediate and superficial cells were counted and their percentages during pro-estrous, estrous and di-estrous were determined. Di-estrous was characterized by the absent of superficial cells in the epithelial vagina. Pro-estrous was characterized by the increasing progressively of intermediate/superficial cells in epithelial vagina, whereas estrous was characterized by the presence of superficial/cornification cells in most epithelial vagina. Based on the dominance of superficial cell, the number of sheep identified as estrous is highest on third day, with 52%.  Observation on vaginal temperature also resulting that the highest temperature values obtained on the third day of 39,08±0.28°C.  It could be effected of the vaginal pH during the observation. Underthe influence ofestrogen, the epithelial vaginalcellssynthesizeand accumulateglycogenin large quantitiesdepositedin the lumen ofvagina. Vaginal bacteriametabolizethe glycogenformlactic acid, which causesvaginal pHis low.The pH conditions prevent from pathogenic microorganisms and fungi. Increased estrogenal so cause cell proliferation through the thickening of the epithelium lining of the vagina so that the cells differentiate.Increasing of glycogenin the superficial cells, and  ceratin cells found in the cytoplasm of cells, as well as acidic pH in the lumen vagina causing changes in the chemical composition of each cell (Zaid, 2011).It can be concluded that during estrous phase, cytology of  vaginal mucose cell is dominated by superficial cell and vaginal temperature is increasing as well as pH of vagina.

Abstract

The objective of the research was to determine the estrous profile of Etawah Crossbred goats after estrous synchronization with different methods. Eighteen does aged 12-24 month old were divided in three groups to receive estrous synchronization treatments (T1 = 14 days intravaginal implant of 60 mg of progesterone (MPA), T2 = two times injection of 5 mg PGF2α (lutalyse) in 11 days interval, and T3 = 10 days of intravaginal implant of 60 mg of progesterone (MPA) + injection of 5 mg PGF2α 48 hours before removal) with six replications. The parameters consisted of estrous behaviour, changes in size and colour of vulva, and duration of estrus when the number of superficial and keratin cells were dominating in the vaginal mucus cell. Data from estrous behaviour and score of vulvar colour was analyzed using Kurkal Wallis test, while onset of estrus, size of vulva slit and estrous duration was analyzed using ANOVA and Duncan test. The result showed that estrous behaviour and changes in color and size of vulva were not significantly different, but estrous duration was significantly different. Estrous duration in T1 (31.30 hour) and T2 (31.10 hour) was significantly longer than that of T3 (11.36 hour). It is concluded that different methods of estrus synchronization affected estrous quality equally but it affected the estrous duration differently based on vaginal mucus cells. Treatment implant vaginal sponge content progesterone for 14 days and double injection of PGF2α with 11-day interval given longest estrous duration.

Abstract

The research was conducted to evaluate the effect of incubation time on viability, plasma membrane integrity, abnormality, and DNA integrity of sexed Pasundan’s bulls sperm. The sperm sexing used 5% and 10% concentrations of Bovine Serum Albumin (BSA).  A completely randomized design with three treatments and six replications was used in this study. The data were analyzed using variance analysis followed by Duncan’s multiple distance test.  Parameter evaluated were sperm longevity, plasma membrane integrity (PMI), abnormality, and DNA integrity of sexed Pasundan bulls sperm. Results showed that incubation time gave significant effect (P<0.05) on the longevity of sperm, but not on the PMI of Pasundan bulls sexed sperm.  The incubation time of 45 minutes gave the highest value of longevity sperm on the upper layer (4.33 days) and the lower layer (4.17 days). Furthermore, the abnormality of sperm X in the upper layer was 4.00%-4.20% and the lower layer was 4.10%- 4.40%.  Meanwhile, the DNA integrity of an upper layer was 98.16%-98.66%, and the lower layer was 97.83%-98.58%.  It is concluded that 45 minutes of incubation time significantly affected the longevity of sperm, but not plasma membrane integrity, abnormality, and DNA integrity of Pasundan bulls sexed sperm.

Abstract

ABSTRAK Glutathione merupakan antioksidan yang berfungsi melindungi sel dari kerusakan akibat oksigen reaktif (ROS) sehingga dapat mencegah reaksi peroksidasi lipid. Penggunaan glutathione dalam pengencer harus sesuai supaya tidak menimbulkan efek negatif. Penelitian ini bertujuan untuk mengetahui pengaruh level glutathione terhadap kualitas semen kambing Peranakan Etawah post thawing dan mengetahui level glutathione yang menghasilkan kualitas semen yang terbaik. Metode penelitian yang digunakan adalah percobaan laboratorium. Penelitian ini mengunakan rancangan acak kelompok dengan lima kelompok kambing dan lima level glutathione (0, 4, 6, 8 dan 10 mM), setiap perlakuan diulang dua kali. Semen ditampung dengan vagina buatan dan dievaluasi secara makroskopis dan mikroskopis, selanjutnya dilakukan pengolahan menjadi semen beku Data dianalisis dengan analisis ragam dan perbedaan antar perlakuan diuji dengan uji lanjut Duncan. Parameter yang diamati adalah kualitas semen post-thawing meliputi motilitas, abnormalitas, membran plasma utuh dan tudung akrosom utuh (TAU). Hasil penelitian menunjukkan bahwa perlakuan level glutathione berpengaruh nyata (p<0,05) terhadap kualitas semen post-thawing. Hasil uji Duncan menunjukkan bahwa motilitas, TAU dari perlakuan level glutathione 6 mM dan 8 mM tidak berbeda nyata, namun MPU hasil 8 mM nyata lebih tinggi dari level lainnya. Disimpulkan bahwa level glutathione berpengaruh terhadap kualitas semen post-thawing, dan level glutathione 8 mM menghasilkan kualitas semen yang terbaik.Kata kunci: glutathione, kualitas semen, post-thawing, kambing peranakan etawahABSTRACT Glutathione is an antioxidant that functions to protect cells from damage caused by reactive oxygen (ROS) so that it can prevent lipid peroxidation reactions. The use of glutathione in diluents must be suitable so as not to cause negative effects The aim of the research was to knew the effect of glutathione level on semen quality of Etawah Crossbreed goat and to know the level of glutathione that produce the best post-thawed semen quality. This research used grouped randomized design with five groups of goat and five glutathione levels (0, 4, 6, 8, and 10 mM), every treatment was repeated twice. Semen was collected with an artificial vagina and was evaluated as macroscopic and microscopic, they were being processed as frozen semen. Data were analyzed with analysis of variance and differences between treatments were analyzed using the Duncan test. Parameter was observed is pot-thawed semen quality consist of motility, abnormality, intact plasma membrane (IPM) and intact acrosome cup (IAC). The result showed that treatment of glutathione level significantly (p<0.05) affect on post-thawed semen quality. Duncan test showed that motility and IAC from treatment glutathione of 6 mM and 8mM were not significant, nevertheless IPM from treatment 8 mM glutathione significantly higher than other levels. It is concluded that the glutathione level significantly affect on post-thawed semen quality, and 8 mM glutathione level resulted in the best quality.Keywords:  glutathione, semen quality, post-thawed, etawah crossbreed goat

Abstract

Aim of the research is to find out recovery rate and percentage spermatozoa X and Y after sexing with gradientt Percoll in Etawah Crossbreed Goat. Extender was used TRIS Egg Yolk. The semen sample from five male Etawah Crossbreed Goat were used in this research. Gradientt percoll performed 10 fraction layer with centrifugation time 10 minute at 2500 rpm. The data were analyzed descriptively. Result of the research shows that average recovery rate of spermatozoa X is 50.96±26.07 % and spermatozoa Y is 44.13±6.11 %. Average percentage of X sperm is 78.00±7.06 % and Y sperm is 76.10±5.95 % .

Abstract

Parity gives an indication of physical maturity of cows. Physical maturity state of primiparous was 82 – 90% while that for pluriparous was 100%. In postpartum period, cows have a negative energy balance. Cows experiencing this condition will use the deposit glycogen in their liver and lipids in adipose tissue to be metabolized by liver become glucose. This experiment aimed at studying blood glucose at different parities of post-partum Fries Holland dairy cows. One-way classification design was use to observe 90 heads of post-partum dairy cows (30 heads of cows each in the first, the second and the third parity state. Data were analyze with General Linear Model (GLM) and the significant difference data among blood glucoses of the cows further analyzed with Duncan Multiple Range Test (DMRT). The results of the study showed that there was a significant (P<0.05) difference between blood glucose of the first parity state of cows (51. 57±5. 56 mg/dL) compared to that of the second (45. 57±8.01 mg/dL ) and the third (46.7±8.62 mg/dL ) cows. This was due to cows in the first parity is in growing state than in producing milk and reproduction function. Cows in the second and the third parities are in opposite function. Blood glucose profile at different parities of post-partum Fries Holland dairy cows was in the normal range (35-55 mg/dL). Blood glucose of the first parity of cows was higher (51.57±5.56 mg/dL) compared to that in the second (45.57±8.01 mg/dL) and the third cows (46.7±8.62 mg/dL). Abstract

This study was conducted to determine the effectiveness of Glycerol as a cryoprotectant with various concentrations on the quality of Pasundan cattle semen.  Semen was collected from seven bulls of Pasundan Cow using an artificial vagina twice a week for three months.  The semen sample was added with a TRIS-Egg Yolk Extender containing 20% (v/v) egg yolk and treated with the addition of Glycerol with five different concentrations (G5=5%, G6=6%, G7=7%, G8= 8%, and G9=9%) were then performed with cryopreservation. A Completely Randomized Design (CRD) was used to examine the effect of five different concentrations of Glycerol on motility, intact plasma membrane (IPM), the integrity of acrosome cap (IAC), abnormalities, and recovery rate (RR) of spermatozoa after cryopreservation (post-thawing).  The results of diluted Pasundan cattle semen evaluation showed that the addition of 7% Glycerol (G7) resulted in the best motility and IAC values (83.68% and 72.84%), the addition of 7% and 8% Glycerol (G7 and G8) resulted in the best IPM values (85.00% and 84.50%).  The addition of 6%, 7%, 8%, and 9% Glycerol (G6, G7, G8, and G9) resulted in the lowest abnormality values (1%).  On the post-thawing Pasundan cattle semen evaluation, the addition of 7% Glycerol (G7) resulted in the best motility, IAC, IPM, and RR values (54.49%, 38.57%, 54.29%, and 72.28%).  Meanwhile, adding various Glycerol concentrations did not significantly affect the abnormality value of post-thawing spermatozoa.  Generally, the addition of 7% Glycerol in semen extenders shows optimal results as a cryoprotectant.

Abstract

Entog (Cairina moschata) is a very potential waterfowl. Attempts to increase the population by diluting semen with Tris diluent. The addition of egg yolks is necessary to avoid cold shock. The purpose of this study was to determine the effect of egg yolk in Tris diluent on abnormalities and integrity of the plasma membrane of sperm. This research was conducted in January – February 2022 at the Faculty of Animal Husbandry, Padjadjaran University. This study used a completely randomized design (CRD) method with 4 treatments and 5 replications. The treatments used were egg yolk level 5% (T1), egg yolk level 10% (T2), egg yolk level 15% (T3), and egg yolk level 20% (T4) in Tris diluent. Data were analyzed by Duncan Multiple Range Test. Based on the results it can be concluded that the addition of egg yolk levels into the Tris diluent affects the abnormality and plasma membrane integrity of sperm. The addition of egg yolk 20% in Tris diluent is the best level to preserve abnormality and plasma membrane integrity of sperm.Keywords: Tris diluent, egg yolk, muscovy duck Sperma’s, Abnormality, Plasma membrane Integrity

Abstract

(Nuclear maturity of bovine oocyte after 24 hours ovary preservation)ABSTRACT. The objective of the research was to investigate their meiotic competence or nuclear maturity of bovine oocytes maturity in vitro after 24 hour preservation on 5C. Oocytes were collected by slicing the ovaries in modified phosphate buffer saline (m-PBS). Selected cumulus-oocyte complexes (COCs) homogenous ooplasm were cultured in maturity medium at 38C in humidified atmosphere of 5% CO2 incubator. After 24 hours, oocytes stained for nuclear maturitys evaluation. The proportion of oocytes at metaphase II (MII) was significantly difference (P 0.05) on oocytes that 24 hours preservated (44.21 3.04%) vs oocytes from fresh ovary (73.97 9.32%) (P0.05). These results indicated that 24 hours preservation bovines ovary on 50C cause decreases of nuclear oocyte maturity.

Abstract

Microsatellite analysis in biotechnology of animal reproduction (A Review)ABSTRACT. In year 1970 was found nucleotide sequence which have repeated sequence of nucleotide. with high polymorh and using PCR could be amplified. That sequenz of nucleotide called Microsatelite. Microsatelite consist of 1 6 repeated nucleotide, which is CA repeated as mostly a repeated DNA in the animal (Tautz and Renz, 1984). Based on difference of long and amount of repeated nucleotide, there are three kind of DNA satelite, midi-, mini- and microsatelite (Matiat and Vergmaud, 1982). Microsatelite analysis was used to analyze of paternity and identity of animal, which was done as a conventional analysis with blood group analysis. The advantage of microsatelite analysis compare to blood group system are the exclution probability was high (EXP 99.9%), needs small sampel (tissues, sperm or follicel of hair), could be use for all animal without special age and possible for died animal.
