Abstract

Kerapu sunu merupakan salah satu kerapu yang mempunyai nilai ekonomi tinggi. Ketersediaan pakan buatan merupakan salah satu kendala pada pengembangan budidaya kerapu di samping ketersediaan benih. Informasi kebutuhan protein dan lemak optimum, energi, asam lemak, dan vitamin telah dimanfaatkan sebagai dasar untuk pengembangan pakan buatan untuk kerapu. Namun demikian, informasi kebutuhan asam amino untuk kerapu masih terbatas sekali. Untuk itu, telah dilakukan riset untuk mengetahui kandungan asam amino lisin optimal dalam pakan untuk pertumbuhan kerapu sunu. Percobaan dilakukan dalam 18 buah bak polikarbonat volume 30 L yang dilengkapi dengan sistem air mengalir. Setiap bak diisi 12 ekor benih kerapu sunu yang berasal dari hatcheri dengan bobot awal 17,6 ± 3,1g. Ikan diberi pakan percobaan dua kali sehari pada level satiasi selama 84 hari. Enam pakan percobaan dibuat berupa pelet kering dengan kandungan protein 45% dan mempunyai kandungan lisin berbeda, yaitu 1,71%; 2,21%; 2,71%; 3,21%; 3,71% dan 4,21%. Pakan percobaan mempunyai komposisi asam amino yang sama kecuali kandungan lisinnya. Percobaan dirancang menggunakan rancangan acak lengkap dengan 6 perlakuan beda level lisin dan tiap perlakuan terdiri atas 3 ulangan. Hasil percobaan menunjukkan bahwa perbedaan level lisin dalam pakan berpengaruh nyata terhadap pertumbuhan dan sintasan kerapu sunu. Berdasarkan data pertambahan bobot ikan diperoleh bahwa kandungan lisin optimal dalam pakan untuk pertumbuhan kerapu sunu adalah 2,84%.Coral trout grouper is one among the most expensive species of grouper. The availability of a practical diet for grouper is still a major constraint to grow-out production besides of good quality of seed. Information regarding the optimum dietary protein and lipid levels, energy, essential fatty acids and vitamin requirements has been used to develop diet for grouper. However, very limited information is available on amino acid requirement for grouper. The objective of the study was to find out optimum lysine content in diet for growth of juvenile of coral trout grouper. A 84-day feeding experiment was conducted in 18 polycarbonate tanks of 30 L volume. Each tank was equipped with flow-through water system. Twelve hatchery_produced juveniles of coral trout grouper (17.6 ± 3.1 g BW) were stocked in each tank. Fish fed experimental diets twice everyday at satiation level. Six experimental diets with different lysine level of 1.71%, 2.21%, 2.71%, 3.21%, 3.71%, and 4.21% were prepared in form of dry pellet. All diets have the same protein level of 45% and the same amino acid composition to the amino acid composition of whole body protein of coral trout grouper, except for its lysine content. The experiment was designed based on completely random design with 6 treatments and 3 replications for each treatment. Result of the experiment showed that dietary lysine levels had significant effect on growth and survival of juvenile of coral trout grouper. Optimum dietary lysine requirement was calculated using orthogonal polynomial regression analysis. Based on weight gain data of indicates that the optimum lysine level in diet for growth of juvenile of coral trout is 2.84%.

Abstract

Percobaan penentuan kebutuhan asam amino lisin untuk benih ikan kerapu bebek (Cromileptes altivelis) telah dilakukan dengan metode pemeliharaan ikan menggunakan pakan yang mempunyai kandungan lisin berbeda. Ikan dengan bobot rata-rata 4,4 ± 0,3 gram dipelihara dalam bak polikarbonat volume 100 L dengan kepadatan 12 ekor ikan per bak. Setiap bak dilengkapi dengan sistem air mengalir dan aerasi. Pakan percobaan dibuat dalam bentuk pelet kering dengan kandungan protein 49% yang bersumber dari kasein dan tepung ikan serta campuran asam amino murni sehingga menyerupai komposisi asam amino tubuh ikan kerapu bebek, kecuali untuk lisin. Pakan percobaan mempunyai kandungan lisin berbeda, yaitu 1,77%; 2,27%; 2,77%; 3,27%; 3,77%; dan 4,27%. Ikan diberi pakan percobaan 2 kali sehari pada level satiasi selama 63 hari. Hasil percobaan menunjukkan bahwa bobot ikan akhir, persen pertambahan bobot, laju pertumbuhan spesifik, efisiensi pakan, retensi protein, dan kandungan protein tubuh ikan dipengaruhi oleh kandungan lisin pakan. Kadar optimum lisin dalam pakan untuk benih ikan kerapu bebek dihitung dengan regresi broken line berdasarkan data persen pertambahan bobot. Dari perhitungan ini diperoleh bahwa kebutuhan lisin untuk benih ikan kerapu bebek mencapai 2,77% dalam pakan atau setara dengan 5,63% dari protein pakan.The experiment to find out an amino acid lysine requirement for growth of  humpback grouper  juvenile has been conducted in 18 polycarbonate tanks, 100 liters volume. Each tank is equipped with flow-through water system. Twelve juveniles of humpback grouper (4.4 ± 0.3 gram in body weight), which were produced in hatchery, were randomly selected, and stock in each tank. Fish fed with experimental diets twice everyday at satiation level for 63 days. Experimental diets were prepared as dry pellet with casein and fish meal as the intact protein sources, and the mixture of crystalline amino acids to reach protein level of 49% and the composition of amino acid similar to the amino acid composition of whole body protein of humpback grouper, except for lysine content. Basal diet (diet-1) contains 1.77% lysine that was supplied from casein and fish meal. Graded level (0.5%) of crystalline lysine was added to the basal diet to get the final lysine level in each diet of 1.77%, 2.27%, 2.77%, 3.27%, 3.77%, and 4.27%. The experiment was designed according to completely random design (CRD) with 6 treatments (lysine levels) and three replicates for each treatment. Result of the experiment showed that dietary lysine content influenced final weight, weight gain, specific growth rate, feed efficiency, protein retention, and body protein content of  humpback grouper juvenile. Optimum dietary lysine for  humpback grouper juvenile was calculated using broken line regression analysis. Optimum dietary lysine requirement for growth of juvenile humpback grouper is 2.77% or equal to 5.63% of dietary protein.

Abstract

Penelitian untuk mengetahui kebutuhan vitamin C untuk pertumbuhan dan ketahanan benih ikan kerapu macan (Epinephelus fuscoguttatus) telah dilakukan, menggunakan 18 buah bak polikarbonat volume 100 liter yang dilengkapi dengan sistem air mengalir dan aerasi. Benih kerapu macan dengan bobot rata-rata 23,0 ± 0,1 gram ditebar dalam bak penelitian dengan kepadatan 12 ekor per bak. Ikan diberi pakan penelitian 2 kali sehari pada level satiasi selama 13 minggu. Pakan penelitian diformulasi dengan kandungan nutrien yang sama kecuali kandungan vitamin C. Pada formula pakan ditambahkan vitamin C (L-ascorbyl-2-phosphate Magnesium, APM) dengan dosis berbeda, yaitu 0, 15, 30, 60, 120, dan 250 mg/kg pakan. Pakan dibuat dalam bentuk pelet dan dikeringkan menggunakan freeze dryer. Penelitian dirancang menggunakan rancangan acak lengkap (RAL) dengan 6 perlakuan beda kandungan vitamin C dalam pakan dan setiap perlakuan terdiri atas 3 ulangan. Hasil penelitian menunjukkan bahwa ikan yang diberi pakan tanpa penambahan vitamin C pertumbuhannya, kadar hemoglobin dan hematokrit darah, aktivitas fagositik, dan indeks fagositiknya adalah paling rendah dibandingkan dengan kelompok ikan lainnya. Dari hasil ini disimpulkan bahwa diperlukan penambahan vitamin C sebanyak 30 mg/kg pakan untuk pertumbuhan ikan yang baik dan penambahan vitamin C sebanyak 60—120 mg/kg pakan untuk kesehatan ikan kerapu macan.The experiment to determine dietary vitamin C requirement for good growth of tiger grouper has been conducted in 18 polycarbonate tanks, 100 liters volume. Each tank is equipped with flow-through water system. Twelve juvenile of tiger grouper (23.0 ± 0.1 gram in body weight), which were produced in hatchery, were randomly selected and stocked in each tank. Fish fed by experimental diets twice everyday at satiation level for 13 weeks. Experimental diets were formulated to contain the same level of nutrients (protein, lipid, and energy), except for vitamin C. Graded level of vitamin C (L-ascorbyl-2-phosphate Magnesium, APM) were supplemented to experimental diet at the level of 0, 15, 30, 60, 120, and 250 mg/kg diet. Diets were prepared as pellet and dried using freeze dryer. The experiment was designed according to complete random design (CRD) with 6 treatments (vitamin C levels) and three replicates for each treatment. Result of the experiment showed that growth, blood hemoglobin and hematocrite, phagocytic activity, and phagocytic index of fish fed diet without supplemental vitamin C were the lowest. This result indicates that vitamin C is essential for tiger grouper. Tiger grouper juvenile requires supplemental 30 mg vitamin C (APM)/kg diet for good growth or 60—120 mg vitamin C for good health condition.

Abstract

The experiment was aimed to know dietary protein requirement and the effect of dietary protein levels on juvenile red snapper (Lutjanus argentimaculatus). The experiment was conducted in 18 polycarbonate tanks, with each 30 liter volume. All tanks were equipped with aeration and flow through water system to maintain good water quality. Hatchery produced juvenile red snapper of 9.4 ± 0.1 g average body weight and total length of 7.7 ± 0.4 cm was randomly selected and 10 fish was stocked in each tank. Fish were fed with experimental diets contained different levels of protein, i.e., 32, 36, 40, 44, 48, and 52%.The experiment used completely random design (CRD) in triplicates. Results showed that dietary protein levels influenced growth, feed efficiency, feed intake, body protein content, and protein retention of juvenile red snapper (p<0.05). The highest weight gain (263.7%) was found on fish fed diet with 40% protein. Based on the growth and feed efficiency data, it was concluded that dietary protein requirement for juvenile red snapper was 40%.

Abstract

ω-3 HUFA are essential fatty acids for marine fish. Deficiency of these essential fatty acids is responsible for high mortality, low growth rate, incomplete formation and disfunction of swim bladder. This experiment was conducted to know dietary ω-3 HUFA requirement for growth of humpback  grouper juveniles.  Juveniles (9.8-10.3  g) were reared in 100 l of polycarbonate  tank with density of 12 juveniles/tank for 8 weeks with flow through system. The levels of ω-3 HUFA (EPA +DHA) supplementation in the test diet  were 0.0, 0.5, 1.0, 1.5, 2.0  and 3.0%. The results showed that the levels of ω-3 HUFA affected the growth and feed efficiency of humpback grouper juveniles (P<0.05). The growth of juveniles fed diet with 0.0-3.0% ω-3 HUFA were  significantly different with the growth of juveniles fed diet with 0.0 and 0.5% ω-3 HUFA. The growth of juveniles fed diet with supplementation levels of ω-3 HUFA at 1.0,  1.5,  2.0,  and 3.0% were 182.2, 168.5, 183.4, and 189.4%, respectively. Whereas supplementation of  ω-3 HUFA at 0.0 and 0.5% resulted the growth of  114.9 and 139.6%, respectively. These results indicated that the requirement of ω-3 HUFA for growth of humpback grouper should be around 1.0% in diet.ω
