Abstract

'Jayanti Agrihorti' is a superior chrysanthemum therefore rooted cuttings quality is required. In vitro propagation increases the rate of propagation and produces true-to-type plants. The research to obtain the best explants and culture medium composition that is capable of producing high-quality plants.  The research was arranged in a randomized complete block design consisting of two factors. The first factor was the explant, including the apical shoot of 0.5 cm, 1.0 cm, and the nodal segment. The second factor was in vitro culture medium composition (CP), consisting of  CP1 (Murashige and Skoog (MS) medium for initiation, followed by MS + 2.5 mg.L⁻¹ gibberellic acid (GA₃) for subculture), CP2 (MS + 0.25 mg.L⁻¹ benzyl amino purine (BAP) for initiation, followed by MS for subculture), CP3 (MS + 0.25 mg.L⁻¹ BAP for initiation, followed by MS + 2.5 mg.L⁻¹ GA₃ for subculture), and CP4 (MS + 0.5 mg.L⁻¹ BAP for initiation, followed by MS + 0.25 mg.L⁻¹ BAP for subculture). Acclimatization was performed after third subculture. The apical shoot size of 0.5−1.0 cm is optimum for producing chrysanthemum-rooted cuttings. Meanwhile, MS medium for initiation stage, followed by MS + 2.5 mg.L⁻¹ GA₃ for subculture is the best culture medium composition for in vitro propagation. This explant and culture medium composition produced higher chlorophyll a, b, and a+b content, thereby resulting in higher plant, more leaves, larger stem diameter, and longer root length. This are recommended for chrysanthemum propagation, particularly in 'Jayanti Agrihorti'.

Abstract

Impatiens hawkeri x platypetala rooted cuttings are typically cultivated using rice husks as a substrate. However, due to the poor water-holding capacity of this medium, Impatiens grows slowly in this environment. A viable alternative is the cocopeat perlite mixture which has a better water-holding capacity compared to the rice husk. Currently, there is no sufficient information available regarding the best growth medium to improve the quality of I. hawkeri x platypetala rooted cuttings in Indonesia. Therefore, this study aimed to obtain the best medium for quality rooted cuttings I. hawkeri x platypetala. The method employed was a randomized complete block design with two factors. The first factor was the Impatiens varieties consisting of Impala Agrihorti, Imadata Agrihorti, and Lavender, while the second was the growth medium, comprised of four levels namely rice husk (control), as well as a mixture of cocopeat with 3%, 5% and 8% perlite. The result showed that there was no interaction between varieties and growth medium. The mixture of 3% perlite in cocopeat exhibited better outcomes compared to other treatments. This medium increased the number and length of roots, stem diameter, plant height, leaves thickness, leaves number and survival rate. Therefore, the addition of 3% perlite in cocopeat could replace the rice husk as a rooted cutting growth medium for I. hawkeri x platypetala.

Abstract

The drought-tolerant Impatiens genotypes are known for their resistance to limited or stressed water. The study aimed to identify drought-tolerance of Impatiens clones. The experiment used a split-plot design replicated three times, with the water stress treatment as the main plot and Impatiens clones as subplots. The main plot consists of 100% and 60% of field capacity. The subplots consist of five Impatiens clones, 17.12; 12; 33.3; 40 B and Impatiens cv of Impala Agrihorti as a control. The results showed that 60% field capacity decreased morphological and physiological traits. Still, the drought-tolerant clones were less affected by the stress and produced more flowers than the others. The most drought-tolerant Impatiens was clone 12. The mechanism of drought tolerance Impatiens was by stomatal closure when the humidity in the growing medium was decreasing. The stomata closure did not significantly reduce the fresh and dry weight in drought-tolerant plants, but it affected the delay in flower initiation. The plant accumulated assimilate for plant height and diameter growth but is not sufficient for generative initiation. They assimilate in the vegetative phase and can be used as sources for flower formation, which show no significant decrease in the number of flowers. The study implies that the drought-tolerant Impatiens clones can be used as genotype sources for drought-tolerant or can be released as new varieties of Impatiens for landscape plants with the superiority in having drought tolerant.
