Abstract

Pengendalian mutu internal laboratorium, dapat digunakan bahan kontrol untuk memantau ketepatan dan mengawasi kualitas hasil pemeriksaan. Bahan kontrol dapat dibuat sendiri menggunakan sisa serum pasien yang dikumpulkan atau disebut pooled sera. Untuk menjaga kestabilan pooled sera, dibutuhkan pengawet. WHO menyarankan untuk menggunakan pengawet etilen glikol, selain itu natrium azida sering dijadikan pengawet dalam serum. Adapun parameter analit yang dipakai dalam penelitian ini adalah glukosa darah sehingga tujuan dari penelitian ini adalah unuk mengetahui perbandingan kadar glukosa darah pada pooled sera dengan konsentrasi etilen glikol 7,5% dan natrium azida 1%. Hasil penelitian secara statistik maupun secara klinis diperoleh bahwa pooled sera dengan konsentrasi etilen glikol 7,5% masih stabil hingga 30 hari dan pooled sera yang ditambah natrium azida 1% dapat stabil hingga 8 hari. Secara klinis, pada pooled sera yang ditambah natrium azida stabil hingga 25 hari. Berdasarkan hasil tersebut, diketahui bahwa terdapat perbedaan stabilitas antara kedua pengawet pada pemeriksaan glukosa darah.

Abstract

Pemakaian setengah volume reagen dan sampel pada pemeriksaan glukosa darah metode GOD-PAP diketahui tidak memiliki perbedaan terhadap nilai simpangan baku dan koefisien korelasi dibandingkan dengan metode standarnya. Metode GOD-PAP setengah volume reagen dan sampel ini lebih efisien dalam segi ekonomi namun belum tervalidasi. Penelitian ini bertujuan untuk mengetahui kinerja metode GOD-PAP setengah volume reagen dan sampel terhadap pemeriksaan glukosa darah. Jenis penelitian ini adalah deskriptif analitik dengan melakukan pengumpulan dan analisis data dari hasil pengujian terhadap delapan parameter validasi yaitu presisi, akurasi, linearitas, reportable range, LoD, LoQ, recovery, dan interferen. Sampel yang digunakan berupa serum kontrol komersial normal (Level 1) dan patologis (Level 2). Data yang diperoleh diolah dengan uji statistik deskriptif menggunakan Microsoft Excel. Hasil penelitian menunjukkan nilai presisi berupa CV Level 1= 1,66%, Level 2= 1,37% (CV<0,33 TEa); akurasi berupa TE Level 1= 3,84%, Level 2= 3,76% (TE<TEa); linearitas dengan nilai r=0,997 (r>0,990); reportable range dengan rentang nilai ≥ rentang nilai normal parameter; LoD = 7,51 mg/dL; LoQ = 20,54 mg/dL (lebih kecil dari batas nilai normal terendah); %Recovery = 99% (berada dalam rentang 95-105%); dan uji interferen menunjukkan hemoglobin mulai mengganggu pemeriksaan dengan %selisih>TEa pada konsentrasi 600 mg/dL. Berdasarkan hasil tersebut dapat disimpulkan bahwa semua parameter uji validasi memenuhi kriteria penerimaan sehingga metode GOD-PAP dengan pemakaian setengah volume reagen dan sampel diterima kinerjanya sebagai prosedur tervalidasi pada pemeriksaan glukosa darah.

Abstract

Liver disease can be caused by infectious substances, pollutants, free radicals, toxic foods, drugs and chemical compounds. Research has been carried out on the effect of green tea leaf ethanol extract in protecting the liver of Balb/C strain mice from CCl4 induction, with the result that green tea leaf extract can protect liver cells from damage. Green tea leaves contain compounds that function as antioxidants, anti-bacterial and hepatoprotectors so that they can be useful in maintaining liver health. This community service aims to provide education and socialization about the benefits of green tea leaves in protecting liver health to residents. Education includes explanations about liver disease, its causes and modes of transmission and how to prevent it. Education and socialization about the benefits of green tea leaves and how to consume them. Evaluation is done by checking blood pressure and questionnaires on the knowledge that has been given. A total of 40 residents have participated in community service, with the following results, the highest age is 41-50 years (42.5%), a decrease in blood pressure of 11% from 150/91 to 133/88, an increase in the percentage of knowledge about green tea after education and socialization from 55% to 69% and increased knowledge of liver and liver disease after education and socialization from 45% to 71%. The conclusion of this community service is that there are 40 people who participate in the community, aged around 20-83 years, a decrease in the blood pressure of residents, after consuming steeped green tea leaves, there is an increase in people's knowledge after socialization and education about green tea and how to consume the leaves have been carried out. green tea and increasing public knowledge after socialization and education about liver and liver disease were carried out

Abstract

Cholinesterase examination related to liver function and carried out by kinetic photometric method with two type reagent addition techniques, namely sample start and substrat start. Both techniques are related to incubation time during examination which affects waiting time for laboratory service result. This study aims to determine comparison of serum cholinesterase activity measured by sample start and substrat start techniques. The research was conducted in May 2024 at Clinical Chemistry Laboratory of Bandung Polytechnic Health. The research using Paired Sample T-test of 30 serum samples measured cholinesterase activity at 405 nm wavelength photometer using sample start and substrat start techniques so that 60 research data were obtained. The result of Paired Sample T-test showed that serum cholinesterase activity with sample start technique had lower trend value than serum cholinesterase activity with substrat start technique and there was no clinical effect with an analytical difference in cholinesterase activity measured by both techniques indicated by the Sig value 0.000 and TE% < TEa% that 4.72% < 8.7%. Therefore, the sample start and substrat start techniques in photometric method of serum cholinesterase activity can be applied for daily testing because the difference in value of two techniques has no clinical effect.
