Abstract

Ceriops tagal are found at Segara Anakan Cilacap, Baluran National Park and Karimunjawa National Park. Information on the genetic diversity can be used for conservation policy. Genetic variation of C. tagal were analysed by using Random Amplified Polymorphic DNA (RAPD). This study aims to determine the genetic variation in each population and between populations C. tagal based on RAPD markers. The total DNA was extracted with CTAB extraction method. Amplification was performed using 10 primer (OPA-3, OPA-10, OPB-1, OPB-6, OPB-7, OPB-8, OPB-10, OPB-12, OPB-17 and OPB-18).  Amplification includes pradenaturation at 94°C for 5 min, followed by 45 cycles of denaturation at 94°C for 1 min, primer annealing at 37°C for 1 min, and primer elongation on 72°C for 1 min 30 sec, final extension at 72°C for 5 min and ends with holding at 4°C for 1 min. The analysis was done by changing the pattern of fragments to binary data (for bands appear = "1" and for bands do not appear = "0"). It is done using a program GenAlEx 2.1 and NTSYS-PC 6.1. A population that has the highest genetic variation (h) was Segara Anakan (0.308), followed by Baluran (0.295) and Karimunjawa (0.123). Baluran has the highest polymorphism (75.00%), Segara Anakan 69.23%, while Karimunjawa was 30.77%. The results of AMOVA, showed that the percentage of genetic variation within populations is greater (77%) than among populations (23%). The results of PCA analysis showed a similar pattern of genetic distance and UPGMA. Samples of Karimunjawa clustered with samples of the population, SA1 and SA3 likely close to the sample of Karimunjawa and SA2 are likely not one group with another sample of the Segara Anakan. Results of mantel test, the value of r = 0.347 and p = 0.02.

Abstract

This study to determine the effect of the effect of Course Review Horray learningassisted by human apron media on science learning oitcomes research on 5th elementarygrade students in kradenan village. This type of pre-experimental research with the designof one group protest posts.Course Review Horray learning with human aprons media on science learningoutcomes in fiifth grade elementary school students in Kradenan village totaling 18students.The data collection method used in this research is using the pretest and posttesttest methods in the form of multiple choice questions. The sampling technique used totalsampling. Data was collected through wal pretest posttest and than analyzed using unWilcoxon in the SPSS 25.00 program for windows.The results showed that the learning Course Review Horray assisted by human themecermatusia apron media had an effect on the learning outcomes of natural science. Thisis evidenced from the results of the analysis of the Wilcoxon signed rank test test with theAsymp value. Sig. (2-tailed) is 0,000 <0.05 and Z score is -3,728. Based on the result of theanalysis and discussion, there is a difference in the average score pretest and posttest,namely the results pretest 54,97 and posttest 78,96, both of which have a difference of 20%.The results of the study can be concluded that learning Course Review Horray assisted byhuman apron media has an effect on learning outcomes of natural science learning in 5thgrade students of Kradenan Village, Srumbung District, Magelang Regency.

Abstract

Diabetes melitus merupakan masalah kesehatan global dengan prevalensi yang terus meningkat. Salah satu pendekatan alternatif dalam pengelolaan diabetes adalah pemanfaatan tanaman obat, seperti Daun Jeruju (Acanthus ilicifolius) yang diketahui mengandung senyawa bioaktif seperti saponin dan tanin. Penelitian ini bertujuan untuk mengevaluasi efek ekstrak etanol daun Acanthus ilicifolius terhadap penurunan kadar glukosa darah puasa pada tikus jantan galur Wistar (Rattus norvegicus) yang diinduksi streptozotocin, serta menentukan dosis optimal ekstrak. Ekstrak daun diperoleh melalui metode maserasi menggunakan etanol 96%. Identifikasi kandungan saponin dan tanin dilakukan dengan metode KLT. Tikus dibagi menjadi lima kelompok (kontrol negatif, kontrol positif, dan tiga kelompok perlakuan dengan dosis 125, 250, dan 500 mg/kgBB). Kadar glukosa darah diukur pada hari ke-0, ke-3, ke-5, dan ke-7. Analisis data menggunakan uji ANOVA dan uji lanjut Tukey HSD. Uji kualitatif menunjukkan bahwa ekstrak mengandung saponin dan tanin. Pemberian ekstrak pada dosis 125 mg/kgBB dan 250 mg/kgBB secara signifikan menurunkan kadar glukosa darah (p < 0,05), dengan efikasi tertinggi pada dosis 125 mg/kgBB. Dosis 500 mg/kgBB tidak menunjukkan efek yang signifikan dibandingkan kontrol negatif. Ekstrak daun Acanthus ilicifolius memiliki potensi sebagai agen antidiabetes, dengan dosis optimal 125 mg/kgBB. Kandungan tanin dan saponin diduga berperan dalam efek hipoglikemik ini. Temuan ini mendukung pengembangan fitofarmaka untuk terapi pendukung diabetes melitus
