Abstract

ABSTRACT Enzymes were protein molecules that synthesized cells to accelerate biochemical reactions. One of the microorganisms that produce a protease was Bacillus subtilis. B. subtilis used in this research consisted of isolate 1 and isolate 2. Methods used include thequalitative assay of protease activity through theclear zone on skim milk agar and calculation of protease activity on production media such as NB (Nutrient Broth) and TSB (Tryptic Soy Broth). Based on the results revealed that the clear zonediameter isolate 2 better than the isolate 1 after 24hours and 48 hours incubation periods. It showed both isolates having the possibility in different strains but within a species. The highest protease activity calculation obtained from TSB production media respectively  0,14 Unit /ml unit and 0.12 Unit/ml. Keywords:B. subtilis, clear zone, protease, TSB

Abstract

Blood Type is an important thing to every person to understand and can identify using a specific methods, blood typing. Blood typing refers to a specific pattern of reaction to testing antisera within a given system. Blood typing have several methods, the common method that usually used is Direct Slide Test. Over a period of time, our understanding on blood groups has evolved to encompass not only transfusion-related problems but also specific disease association with RBC surface antigens. This community service activity aimed to provide blood typing examination for students who unknown their blood type and strenghten the understanding about the important of Blood Type. This activity was held on December 20, 2021 in Stikes Rajekwesi Bojonegoro. This Community service applied an examination test to several students who still unknown their blood type. Examination of blood group used the A-B-O system and rhesus with the slide test method. The results of the blood group examination of 16 students showed that the highest results were blood group B as many as 6 people (37.5%) and the least blood group being O and AB with 3 people each (18.75). Rhesus examination showed that all students were rhesus positive (100%). Students who examinated during the programmes doesn’t have undestanding of the important of knowing their blood type. Students of Stikes Rajekwesi Bojonegoro get sufficient understanding about the important of knowing their blood type and the blood typing showed 37,5% of student are B type, 18,75% are O and AB type each, while 100% rhesus positive.

Abstract

Background: Squamous cell carcinoma (SCC) is an invasive skin cancer that is found in the human skin. SCC growth from the precancerous lesion is known as a wart or papilloma. Ki67 is a protein expressed by cells during proliferation. Cells with higher Ki67 expression showed abnormal regulation of apoptosis. 5-FU is an antimetabolite compound that works as a DNA/RNA pyrimidine antagonist molecule that can induce cell apoptosis. The main objective of this research is to evaluate the inhibition proliferation of precancerous squamous cell in skin lesion induced by DMBA/croton oil treated by using 5-FU cream topically compared to 5% imiquimod cream. Methods: This research assesses three different concentrations of 5-FU cream namely 1%, 2%, and 5% topically on 24 wild type mice divided into 6 groups of 4 each including positive control (with carcinogenesis but without treatment), negative control (without treatment ; normal), carcinogenesis with treatment 5-FU cream (1%, 2%, and 5%) or 5% imiquimod cream. The expression of Ki67 was analyzed using immunohistochemistry. Statistical analysis was performed by one-way ANOVA using SPSS version 23. Results: Based on this research, 5-FU cream treatment during 4 weeks (week 10-week 14) showed the decrease of cumulative number of papilloma (p<0.05) and immunohistochemistry analysis resulted in the lowest Ki67 expression in 5-FU 2% and 5% groups compared to 5% imiquimod cream (p<0.05). Abstract

The third peak of the Coronavirus Disease 2019 (COVID-19) in Indonesia which was recorded on February-March 2022 had an impact on voluntary blood donations at the Indonesian Red Cross (IRCS) Bojonegoro. The objective of this research was to analyze the voluntary blood donors’ characteristics during the third COVID-19 peak in Bojonegoro Regency. This descriptive, analytical study was conducted using secondary blood donor data from February 22, 2022 to March 19, 2022 at IRCS Bojonegoro. The data were collected in the blood donor selection process in accordance with the Indonesian and World Health Organization (WHO) blood donor selection guidelines. The average number of voluntary blood donors per day was tested using the Mann-Whitney-U test with a significance ofp<0.05. The results showed 219 people (80.51%) between 22-28 February and 332 people (83.63%) between 1-19 March 2022 who were approved to donate at IRCS Bojonegoro. There was a significant decrease in the average of male voluntary blood donors per day (p=0.01),average age group: 17-24 years (p=0.013), 35-44 years (p=0.002), and 45-54 years (p=0.030), hemoglobin (Hbl) levels on 16.1-17.0 g/dL (p=0.000), systolic blood pressure (SBP) <120 (p=0.03) and 130-139 (p= 0.02) and diastolic blood pressure (DBP) <80 (p=0.001), pulse rate (PR) 65-74 beats/minute (p=0.016), and blood type O+ (p=0.008). The characteristics of voluntary blood donors during the third peak of COVID-19 in Bojonegoro Regency indicated there was a significantly decreased number of male voluntary blood donors per day around ages 17-24 and 35-54 years, with Hbl between 16.1-17.0 g/dL, SBP <120 and 130-139 mmHg, DBP <80 mmHg, PR 65-74 beats/minute, body weight 45-54 and 65-74 kg, and blood type O+.

Abstract

Penelitian ini bertujuan untuk meningkatkan hasil prestasi belajar siswa terhadap mata pelajara Bahasa Jawa materi tembang macapat dhandhanggula dengan menggunakan model pembelajaran direct learning pada siswa kelas VIII-A Semester 2 SMP Negeri 1 Karangrejo Tulungagung Tahun Pelajaran 2018/2019. Hal ini didasari oleh masih rendahnya nilai ketuntasan siswa akan materi tersebut. Diharapkan dengan menggunakan model pembelajaran direct learning maka nilai siswa dapat meningkat. Metode yang digunakan yakni penelitian penelitian tindakan kelas (PTK). Siklus yang digunakan sebanyak 2 dan masing – masing siklus memiliki tahapan yang terdiri dari perencanaan, tindakan, observasi dan refleksi. Hasil tindakan dengan model pembelajaran direct learning menunjukkan hasil jika dengan penerapan metode ini dapat meningkatkan hasil prestasi belajar siswa. Pada siklus I nilai ketntasan siswa yakni 61,4% dan pada siklus II mengalami kenaikan menjadi 93,2%.

Abstract

Penelitian yang sudah dilaksankan ini mempunyai tujuan untuk meningkatkan nilai hasil belajar siswa kelas VIII-B semester 1 SMP Negeri 1 Karangrejo Tulungagung tahun pelajaran 2019/2020. Mata pelajaran yang diteliti yakni Bahasa Jawa dengan materi wawancara. Penelitian menggunakan perbaikan pembelajaran dengan menerapkan Cooperative Learning Tipe Group Investigation. Hal yang mendasari penelitian ini yakni rendahnya nilai siswa pada mata pelajaran Bahasa Jawa. Data yang didapatkan dari hasil observasi, wawancara dna juga tes. Hasil penelitian menunjukkan jika penerapan model pembelajaran Cooperative Learning Tipe Group Investigation dapat meningkatkan hasil belajar Bahasa Jawa yang memuat wawancara pada siswa kelas VIII-B Semester 1 SMP Negeri 1 Karangrejo Tulungagung tahun pelajaran 2019/2020. Siswa pada siklus I mendapatkan ketuntasan sebesar 64,3% sedangkan pada siklus II siswa mendapatkan ketuntasan sebesar 85,7%.

Abstract

Skin cancer is a disease that develops in the epidermis of the skin and can be invasive, such as squamous cell carcinoma (SCC). Early detection of squamous cell precancerous can prevent these lesions from progressing to invasive SCC and increase the effectiveness of therapy. 5-fluorouracil (5-FU) is an antimetabolite compound as a pyrimidine DNA/RNA antagonist molecule that induces cell apoptosis. The main objective of this study was to evaluate the effectiveness of the topical 5-FU cream (Dharmais NCH) compared to imiquimod 5% on apoptosis through the expression of caspase-3 in precancerous squamous cells of mouse skin induced by 7,12-dimethylbenzen[a]-anthracene (DMBA)/croton oil treatment. This research assess three differences concentration of 5-FU include 1%, 2%, and 5% on 24 wild type mouse divided into 6 groups including positive control (with carcinogenesis but without treatment), negative control (without treatment; normal), carcinogenesis with treatment 5-FU cream (1%, 2%, and 5%) or 5% imiquimod cream. Two-stages carcinogenesis induced by DMBA and followed by croton oil. The expression of caspase-3 was analyzed using immunohistochemistry. Statistical analysis was performed by one-way ANOVA using SPSS version 23. The induction of two-stages of carcinogenesis (weeks 1 to 10) caused papilloma lesions on the skin of mouse. Furthermore, 5-FU treatment for 4 weeks (weeks 11 to 14) showed a decrease in the cumulative number of papillomas (p<0.05) and immunohistochemical analysis showed caspase-3 expression on 5-FU treatments (1%, 2%, and 5%) was not significantly different from the imiquimod treatment (p>0.05). The apoptotic effect of 5-FU treatment on precancerous skin squamous cell lesions in mouse was not significantly different from the standard treatment using imiquimod. This suggests that 5-FU treatment has potential as a future therapy in squamous cell precancerous skin lesions.Keywords: 5-fluorouracil, caspase-3, squamous cell precancerous, skin, topical treatment.
