Abstract

Penelitian ini bertujuan untuk mengisolasi dan menghitung kelimpahan koloni bakteri Vibrio sp. dari air tambak udang dengan 5 lokasi yang berbeda di Kecamatan Simpang Kabupaten Pidie, Aceh. Penelitian ini menggunakan metode eksploratif dan pengambilan sampel menggunakan purposive sampling. Isolasi bakteri menggunakan media selektif TCBSA. Perhitungan kelimpahan koloni Vibrio sp. menggunakan metode Total Plate Count. Hasil penelitian menunjukkan bahwa kelimpahan bakteri Vibrio sp. di Kecamatan Kembang Tanjong untuk kelima kategori lokasi tambak tergolong aman ( 104 cfu/ml) yaitu 6,2 x 102 – 2,6 x 103 cfu/ml. Berbeda halnya dengan tambak di Simpang Tiga yang masih dapat dikatakan aman hanya tambak K2T2 (2,9 x 103 cfu/ml), K4T2 (2,2 x 104 cfu/ml, K2T1; K5T; K5T2 (2,3 x 104 cfu/ml) karena kelimpahannya 8,34 x 104 cfu/ml, sedangkan bakteri Vibrio lainnya sudah bersifat patogen (105 cfu/ml). Hasil pengukuran kualitas air tambak menunjukkan rata-rata nilai salinitas 10,67-30,33 ppt, suhu 27,9-33,8 oC, pH air 7,65-8,45, DO 5,35-8,42 dan kecerahan 20-55 cm. Tidak adanya hubungan korelasi yang signifikan dari kualitas air tambak terhadap kelimpahan koloni bakteri Vibrio sp.

Abstract

Cilembu Sweet Potato is one type of sweet potato that is typical in the West Java area precisely in the Cilembu village or Sumedang. Cilembu sweet potato has a typical characteristic that is taste like honey if it fermentation and then roasted. Rizospheric bacteria with the ability of phosphate solvents are one of the factors supporting growth in plants. The research aims to identify Rhizosphere bacteria in Cilembu sweet potato roots which have the ability as phosphate solvents at two locations during the growth phase. The study was conducted with qualitative by testing the biochemical characteristics and ability of phosphate solvents. The result of this research show that Rhizosphere bacteria with the ability of phosphate solvents at the location of Cilembu (LCI) and Jatinangor (LJA) are not significantly different. In LCI, 12 isolates were obtained and in LJA 7 isolates were obtained. So that it can be concluded that the diversity of rhizosphere bacteria can be influenced by the characteristics of the soil and the growth phase.Keywords: Cilembu yam, Rizosphere bacteria, phosphate solvent

Abstract

Mangrove forest is one of the natural resources that has the potential for protection and conservation of nature. Conservation can be defined as a system of management and sustainable use of the environment and natural resources. Tanjung Rejo Village, Percut Sei Tuan sub-district is one of the coastal areas with mangrove forest conditions that have been damaged. This study aims to identify the conservation efforts carried out by the people of Tanjung Rejo Village, precisely in the tourist area of the Mekar Bahari Mangrove Forest. The research was conducted by means of surveys, observations and interviews with the management community. The results show that the conservation efforts carried out by protecting and culturing mangrove plants and making mangrove forests as ecotourism and education of mangrove forests have not getting reached for maximum, if viewed from the presence of mangrove plants and the surrounding environment. Based on surveys and interviews about community perceptions of eco-edutourism development, the community agrees that in the near of future the mangrove forest in Tanjung Sari Village will be used as an eco-edutourism area. It can be said that conservation efforts do not reach the standard, so it is important to develop eco-tourism areas in mangrove forests by increasing public awareness of   mangrove preservation. Keywords: Conservation, eco-edutourism, Mangrove Forest

Abstract

 Simbiosis mutualisme spons dengan bakteri mampu menghasilkan senyawa bioaktif salah satunya ialah sebagai antibakteri. Tujuan dari penelitian ini adalah mengisolasi dan melakukan pencarian bakteri yang bersimbiosis dengan spons di Pulau Bakar yang mempunyai aktivitas antibakteri terhadap Escherichia coli. Tahapan penelitian ini yaitu isolasi dan identifikasi bakteri simbions spons selanjutnya pengujian aktivitas antibakteri dengan metode difusi cakram. Pada hasil penelitian ini ditemukan 16 jenis bakteri yang bersimbiosis dengan spons. Isolat terbaik yang mampu menghambat bakteri Escherichia coli ialah SPS4(2)B dengan diameter zona hambat yang ditimbulkan ialah  12,01 mm dan tergolong ke dalam kategori kuat. Dapat disimpulkan bahwa bakteri yang bersimbiosis dengan spons di perairan Pulau Bakaran mampu menghambat pertumbuhan bakteri Escherichia coli Kata Kunci: antibakteri, bakteri bersimbiosis,  spons, Escherichia coli

Abstract

Persoalan sampah merupakan persoalan yang sangat penting bagi masyarakat khususnya masyarakat yang tinggal di Pedesaan. Hal ini menjadi penting untuk dibahas karena berkaitan dengan sistem kehidupan di masyarakat yang berkaitan juga dengan nilai-nilai sosial dan budaya. Secara garis besar lingkungan yang sehat menjadi pemicu berkembangnya interaksi sosial dan kebudayaan yang positif bagi masyarakat. Desa Sukatendel Kecamatan Tiganderket Kabupaten Karo merupakan salah satu desa yang juga dihadapkan dengan persoalan sampah. Mengingat begitu banyaknya dampak negatif yang ditimbulkan oleh sampah terhadap kesehatan lingkungan yang juga merusak nilai sosial dan budaya, dilakukan edukasi juga upaya pengelolaan dan penanggulangan sampah sebagai solusi dari permasalahan sampah yang ada di Desa Sukatendel Kecamatan Tiganderket Kabupaten Karo. Penelitian ini menggunakan penelitian kualitatif deskriptif dengan teknik pengumpulan data melalui analisa dan verifikasi terhadap program yang sudah dilakukan oleh Tim KKN 185 di Desa Sukatendel sehingga peneliti dapat menarik kesimpulan bahwa dalam melakukan edukasi kepada masyarakat untuk pengelolaan dan penanggulangan sampah di Desa Sukatendel adalah dengan memberikan edukasi kepada masyarakat mengenai optimalisasi pengelolaan sampah dan dampak pada Desa Sukatendel melalui kegiatan seminar, pemberdayaan sampah menjadi lahan ekonomi baru bagi masyarakat Desa Sukatendel melalui pembuatan ekonomi kreatif dari sampah kulit jagung, dan juga pembuatan tempat sampah permanen sebagai solusi dari penanggulangan sampah di Desa Sukatendel.

Abstract

Salak is a fruit that contains various health benefits, including the flesh of the fruit which contains bioactive compounds. Taking bioactive compounds directly from the flesh of the salak fruit will require quite a lot of biomass. Obtaining these bioactive compounds can be streamlined by using endophytic bacteria. Endophytic bacteria able to produce compounds of  antibacterial. The aimed of this study to examine the antibacterial potential of  endophytic bacteria derived from the flesh of the Pondoh and Padangsidempuan salak fruit against pathogenic bacteria, namely Escherichia coli. Method this studies is a laboratory experiment. A total of 10 isolates consisting of 6 isolates of Pondoh salak and 4 isolates of Padangsidempuan salak used were the results of isolation from salak fruit flesh in previous studies. Based on the results of the study, ten isolates indicate of antimicrobial activity to Escherichia coli. The inhibition zone diameter of highest value was owned by SP4 isolate, which was 1.1 mm. These results indicate that endophytic bacteria from the pulp of Pondoh and Padangsidempuan salak possess the potential to produce antibacterial compounds against gram-negative pathogenic bacteria, namely Escherichia coli.

Abstract

The aims of this study were to determine whether Clostridium botulinum bacteria were present in used cooking oil, and to look at the characteristics of Clostridium botulinum bacteria in used cooking oil. In this study, isolation of bacteria from used cooking oil using blood agar media, identification of bacteria using gram staining and biochemical tests such as gelatin test, triple sugar iron agar (TSIA) test, simon citrate test, and motility test were carried out. The results of the study obtained 2 out of 3 samples containing Clostridium botulinum bacteria. These bacteria are more common in used cooking oil, which has a more blackish brown color, lots of crumbs left over from food, and temperatures around 30-55℃. In conclusion, there is Clostridium botulinum bacteria found in used cooking oil. Clostridium botulinum colonies are grey, alpha hemolytic, circular to irregular in shape with scalloped edges or rhizoids, and flat to raised surfaces. Abstract

This study aims to determine whether there are Coliform bacteria contained in residents' well water in Tanjung Selamat Village, Medan Tuntungan District. In this study, physical tests were carried out on samples using sensory devices, chemical tests using pH paper, and bacterial identification using the MPN test which included a prediction test using Lactose Borth media, a confirmation test using Brilliant Green Lactose Borth (BGLB) media, and a certainty test. using eosin methylene blue (EMB) media. The results of the physical test obtained 1 sample and the chemical test obtained 3 samples that met the requirements for clean water health. Meanwhile, from the MPN test, the MPN value was obtained for sample 1: 43/100 ml of water, sample 2: >2400/100 ml of water, sample 3: 60.06/100 ml of water. The difference in the number of MPN values found in the samples indicates that the samples have different levels of contamination and pollution levels. Based on the research that has been done, it can be concluded that of the three water samples taken from residents' well water in Tanjung Selamat Village, Medan District, Tuntungan was positive for Coliform bacteria.
