Abstract

Everyone needs to know the condition of the body that can restore the body's stamina so that the optimal condition is the need for sleep. Working hours and biological rhythms that do not match can cause stress in the form of hormonal changes that are controlled by the hypothalamus. The hypothalamus will trigger the anterior pituitary to increase cortisol. Cortisol reduces the number of eosinophils, basophils, monocytes, and lymphocytes, interleukins 1 and 6 as well as suppression of T cells in the blood so that the level of immunity against foreign substances entering the body is reduced. The purpose of this study was to determine the effect of sleep quality on the formation of erythrocytes in workers with a shift system in the peripheral circulation. This study uses analytical estimation through a horizontal approach. Based on the analysis, there is a significant relationship between sleep quality and the number of ATLM episodes with the shift work system at BBLK Palembang. Chi Square statistical test results obtained from 86 people there are 35 people with good sleep quality, from 35 respondents with good sleep quality 33 people (94.3%) have a good blood count and 2 respondents (5.7%) others have a low blood cell count. As for ATLM with a shift work system, it is expected to be able to maintain good and sufficient sleep quality, as well as improve good sleep quality.

Abstract

Candida albicans is a fungus that lives as normal flora in certain parts of the human body which can instantly become pathogenic if the patient experiences changes or decreases in body resistance. One of the diagnoses for the Candida albicans fungus is culturing, namely isolating it on gowth media such as Sabouraud Dextrose Agar (SDA), but in some areas, it is difficult to use this medium because the price is quite expensive. Jackfruit seeds contain many nutrients needed by fungi such as protein, carbohydrates, and other minerals so that they can gow the Candida albicans fungus, this study aims to determine the difference in the number of Candida albicans colonies on SDA media and jackfruit seed modification media. The research method used a pure experiment. This research was conducted at the microbiology laboratory of the IKesT Muhammadiyah Palembang with a total of 32 samples. The results showed that the average number of colonies of Candida albicans on SDA was 112 CFU/mL media and jackfruit seed modified media 156 CFU/mL. Wilcoxon test obtained a p-value of 0.05 with a value of 0.000. This study concludes that there were differences between SDA media and jackfruit seed modification media.

Abstract

Introduction: Leukocytes are blood cellscontaining nuclei, orso-called white blood cells. Leukocytes have a role in the cellular and humoral defense of organisms against foreign substances. Some types of leukocytes or white blood cells are present in the blood. Leukocytes are generally divided into granuocytes (which have special granules) andagranulocytes (which do not have special granules). Granulocytes consist of neutrophils, eosinophils, and basophils. Agranulocytes consist of lymphocytes and monocytes. Leukocyte count examination is a routine blood test conducted in the clinical laboratory because of the frequent request for a test to calculate the number of leukocytes. Calculate the number of leukocytes not only determinethe number ofleukocyte cells, but can also be any type of leukocyte cells. Objectives: The purpose of this study is to determine the difference in the number of leucocytes immediately and stored for 6 hours.Methods: This research method usescross sectional, this sample was taken from studentslevel 1 and 4Medical Laboratory Technologyof Health Sciences and Technology Muhammadiyah Palembang. Result: The results of this study obtained an averagecount of the number of leukocytes immediately is 6.3667 cells/μl, while the average count of the number of leukocytes stored for 6 hours is6.4667 cells/μl. Result: The results of T-test statistics showed a significant value (0,477) was higher than the alpha value of 0.05 or ρ>ɑthen Ho, receivedwhich means there is no difference in calculating the number of leukocytes immediately and stored for 6 hours. Conclusion: The conclusion of this study is that there is no difference in calculating the number of leukocytes immediately and disinced for 6 hours.

Abstract

Background : Laboratory tests, especially hematology, are frequently requested by doctors to help make a diagnosis. White blood cells (leukocytes) are an important part of the body’s defense system whose function is to fight infection causing microorganisms, tumor cells, and harmful foreign substances. Counting the number of leukocytes by manual method is done by diluting the blood using Tur ation, but the problem is that sometimes the Turk solution is not available. or is available but has expired, so an alternative is done by modifying key orange. juice (Citrus macrocarpa Bunge). Research purposes : To determine the difference in the number of leukocytes counted using modified key lime juice (Citrus microcarpa Bunge) and Turk solution. Research Methods : Experimental this research was conducted on January 19, 2023 at Bunda Hospital Palembang. and sampling was carried out at the IkeST Muhammadiyah Palembang Hematology Laboratory, totaling 31 EDTA blood samples. Results : The results obiained in this study showed that there was a difference in the leukocyte count with the modification of key orange juice (Citrus macrocarpa Bunge), turk solution, and a hematology analyzer. The results of the research data were carried out using data analysis with the SPSS program with the Shapiro–Wilk normality test and continued with the Kruskal test. Conclusion : There is a difference between the leukocyte count examination using modified key lime juice (Citrus macrocarpa Bunge) and Turk solution.

Abstract

ABSTRAK Pendahuluan : Eritrosit merupakan salah satu komponen darah yang jumlahnya paling banyak dalam susunan komponen darah manusia eritrosit normal selalu berbentuk bikonkaf, tidak memiliki inti, dan mengandung hemoglobin yang merupakan representasi warna merah dalam darah. Pemeriksaan hitung jumlah eritrosit adalah pemeriksaan yang bertujuan untuk menentukan jumlah eritrosit dalam 1 µL darah dan digunakan sebagai tes skrining anemia dan polistemia,Menghitung jumlah eritrosit dapat dilakukan dengan dua metode, yaitu manual dan otomatis larutan isotonic yang kemudian dihitung didalam kamar hitung. Pemeriksaan hitung jumlah eritrosit metode manual menggunakan reagen hayen. Tujuan: untuk mengetahui perbandingan hasil pemeriksaan eritrosit menggunakan garam dapur dan reagen hayem. Metode: Jenis penelitian ini adalah cross sectional komperatif . penelitian ini dilakukan dilaboratorium hematologi IkesT Muhammadiyah palembang pada tanggal 5 januari 2022 dengan jumlah sempel sebanyak 32 sempel. Hasil : hasil penelitian memiliki nilai Rata-rata hitung erirosit dengan garam dapur sebesar 422.812 sel/mm3 dan Rata-rata hitung eritrosit dengan reagen hayem sebesar 2.263.750 sel/mm3. Hasil uji T berpasangan didapatkan nilai p = 0.000 Kesimpulan dari hasil penelitian adalah terdapat bebedaan hasil hitung eritrosit menggunakan garam dapur dan reagen hayem

Abstract

Latar belakang: Diabetes Melitus (DM) merupakan pembunuh nomor empat di dunia. Pemeriksaan HbA1c mencerminkan konsentrasi glukosa darah jangka Panjang. Penggunaan HbA1c sebagai Tindakan diagnostic merupakan standar perawatan oleh American Diabetes Association berdasarkan rekomendasi dari International Expert Committee. Stabilitas penundaan analisis menunjukkan bahwa inflasi nilai HbA1c berhubungan langsung dengan waktu penyimpanan, kondisi penyimpanan sampel harus dipantau dan dikontrol dengan hati-hati. Pemeriksaan HbA1c menggunakan metode Immunoassay dengan alat FinacareTM FIA Meter Plus Wondfo dari sampel pasien diabetes melitus sebanyak 29 sampel. Tujuan penelitian: Untuk mengetahui apakah ada perbandingan kadar HbA1c pada darah EDTA yang diperiksa segera dan yang disimpan 2 jam disuhu ruang 20-25°C. Metode penelitian: menggunakan studi Experimental Laboratory dengan rancangan one group pretest and posttest design. Data yang didapatkan diolah dengan program SPSS dilakukan uji Normalitas dengan uji Shapiro – wilk Bila hasil terdistribusi normal maka dilanjutkan uji hipotesis dengan uji – T berpasangan (Paired sampel T test). Dan jika tidak normal dilanjutkan dengan transformasi data, dan jika tidak normal maka dilakukan dengan uji non parametrik dengan Wilcoxon. Hasil: Hasil data yang dilakukan dengan uji (Paired Sampel T Test) didapatkan signifikan p = 0,959. Yaitu tidak ada perbedaan. Simpulan: Berdasarkan hasil penelitian dapat disimpulkan bahwa antara pemeriksaan kadar HbA1c pada EDTA yang diperiksa segera dan disimpan 2 jam disuhu ruang (20-25℃) tidak terdapat perbandingan yang signifikan, artinya pada penyimpanan 2 jam disuhu ruang (20-25℃) sampel untuk pemeriksaan HbA1c masih memiliki hasil yang baik.

Abstract

Examination of total cholesterol levels in serum samples is more often used as an examination material, but serum samples often encounter difficulties due to insufficient blood volume or the condition of the serum being lysed due to poor sampling. Many studies have been conducted to look at total cholesterol levels using the anticoagulants EDTA, Sodium Citrate, and Sodium Oxalate to look at differences in total cholesterol levels, but not many studies have looked at tests of differences in total cholesterol test results during fasting with serum and EDTA. This study aims to determine differences in total cholesterol levels in the fasting state of EDTA serum and plasma samples using the BA200 biosystem. The type of research used was cross sectional, which was carried out at the Hematology Laboratory of the Palembang Muhammadiyah Institute of Health Sciences and Technology and the Palembang Health Laboratory Center. A sample of 30 people was taken purposively, female, and fasting for 10 hours. Based on the results of examining total cholesterol levels in a fasting state using the Biosystem BA200, the average result for serum samples was 4.8 mmol/L and the average for plasma samples was 4.7 mmol/L. Analysis was carried out using an independent sample T-test and the result was p = 0.00, the hypothesis was accepted. This research can conclude that there is a difference between the results of examining total cholesterol levels in fasting serum and EDTA plasma samples using the BA200 biosystem.

Abstract

Oxidative stress is a condition characterized by an imbalance between the production of free radicals and the body's ability to neutralize them with antioxidants. In young adults, oxidative stress is a significant risk factor for chronic diseases such as diabetes, hypertension, and heart disease. This study aims to analyze the effect of physical activity on oxidative stress biomarkers, specifically malondialdehyde (MDA) and glutathione peroxidase (GPX), in young adults. A cross-sectional study was conducted involving 50 participants aged 20–30 years, categorized into low, moderate, and high physical activity groups. Physical activity levels were assessed using a questionnaire, while MDA and GPX levels were measured through blood sample analysis using the spectrophotometric method. The data were analyzed using correlation and ANOVA tests to evaluate the relationships and differences in biomarker levels among the physical activity groups. The results showed significant differences in oxidative stress biomarkers and antioxidant capacity between physical activity groups (p < 0.05). Higher physical activity levels were associated with lower MDA levels, indicating reduced lipid peroxidation and oxidative stress. These findings support the promotion of regular, moderate-to-vigorous physical activity among young adults as an effective strategy for enhancing antioxidant defense, reducing oxidative stress, and potentially preventing chronic diseases linked to oxidative damage.
