Abstract

BackgroundThe aim of this study is to develop a method for optimal in vitro proliferation and differentiation of rat adipocyte. MethodsPreadipocyte were isolated from omentum of Rattus norvegicus Wistar using Collagenase type I dan II (Sigma). Cells were cultured in M199 culture containing either 0%, 8% or 10% Fetal Bovine Serum (FBS). Insulin were added into the media once the cells attached to the culture plate Proliferating and differentiated cells were counted and analysed by using Oil Red O dan Hematoxylen (HE) staining. Results This study demonstrated that adipocyte could be isolated using Collagenase type I but not type II. The addition of 10% FBSsignificantly increased the number of preadipocyte and differentiation of adipocyte more than those of 8% FBS and without FBS. The timing of FBS addition was best performed on day 8 using 10% FBS. Specific adipocyte staining using Oil Red O revealed thatthere were core lipids in mature adipocyte. ConclusionsCollagenase tipe I could be used to isolate preadipocyte cells. Supplementation of culture media with 8-10% FBS could enhance the in vitro proliferation of preadipocyte. Standard media M199 containing 10% FBS and insulin may provide an environment to differentiate preadipocyte specifically into adipocyte in vitro.

Abstract

The effect of Ramadan fasting on some blood parameters i.e. serum glucose, urea, uric acid, lipids and proteins, were investigated on young men in Ar Rohmah Islamic dormitory. Nineteen normal and healthy students aging between 12-25 years, residing in the Islamic dormitory, voluntarily to participated in the study. Blood samples were obtained from the volunteers on the 1st and 26th day of Ramadan and analyzed for the aforementioned biochemical parameters. A non-significant effect of Ramadan fasting was observed on most of the parameters studied. However, serum urea, triglycerides, total cholesterol and LDL-cholesterol were reduced significantly (p 0.05) but remained within the physiological limits. Decrease in blood urea has been attributed to the effect of at least protein and triglycerides intake to increase lipolytic effect. The reduction in serum cholesterol and LDL is a beneficial effect of Ramadan fasting. The results of the study indicated thatRamadan fasting is quite safe for normal healthy adults.

Abstract

Combustion process in industry resulted in harmful free radical compounds such as Dioxin / TCDD (2,3,7,8- tetrachlorodibenzo-p-dioxin). The aim of the this experiment was to determine the therapeutic effect of casein in goat milk yogurt on chickens exposed to dioxins. Parameters used in this experiment were feed consumption and body weight. This research was designed with 18 male COBB strain broiler chickens aged 21 days. They were divided into three groups: (1) the negative group without any treatment, (2) the positive group by giving feed with dioxin 50ng / kg feed in corn oil solvent, (3) ) the therapy group was given feed with dioxin 50ng / kg feed in corn oil solvent in the morning and 750 mg / kg BW / day of yougurt casein goat milk in the afternoon. Both feed and casein were given orally for 21 days with each group having six replications. The samples were measured every day including weight gain and chicken feed consumption. The data were analyzed quantitatively using the One Way Analysis of Variance (ANOVA) test with a confidence level of 95%. The provision of goat milk yogurt casein tablets as a therapy for broiler chickens that were given dioxin-containing feed had a significant increase on chicken body weight, but insignificant on feed consumption.

Abstract

Jatropha or Jatropha curcas L. is a plant of the Euphorbiaceae family with medicinal and non-medical uses, including oral contraceptives. The part of Jatropha that is often used is the seed. However, Jatropha seed also contains saponins, phorbol esters, currants, tannins which can cause organ poisoning. This study was conducted to determine the effect of Jatropha seed extract on liver histopathology and SGPT levels in Rattus Norvegicus rats, using a completely randomized design method. Experimental animals were divided into four groups, and each group consisted of five female rats. The groups were the control group without treatment and those treated with Jatropha seed extract orally at doses of 10, 50, and 100 mg/kg BW. The induction process was carried out every day for 14 days, on the 15th day, a necropsy was performed. Livers were taken for histological analysis, and blood samples were taken to measure SGPT levels. Results showed that the administration of Jatropha seed extract could increase SGPT levels. The administration of Jatropha seed extract at a dose of 100 mg/kg body weight induced the most severe damage of hepatocytes among the treatment groups.

Abstract

The work processes in the furniture industry starts from wood transportation, construction, sanding, assembling, and finishing. One of the potential hazards that always exist in every work process in the furniture industry is ergonomic hazards. The risks of Manual Material Handling (MMH) that includes lifting and lowering, pushing and pulling, rotating, carrying, and holding against an object adversely affect both the company and workers. This study aims to determine potential ergonomic hazards and control efforts to reduce risk of ergonomic hazards and injuries to furniture industry workers by providing training Occupational Safety and Health and proposed work system during Manual Material Handling activities. The method used to identify is the Hazard Identification Risk Assessment Control (HIRAC) technique. HIRAC used to help provide an understanding of the hazards that exist in each work process to minimize the occurrence of adverse Occupational Diseases. This research is qualitative research with observation and interview data collection techniques. Analysis techniques in this research included data reduction, data presentation, conclusions, and verification. Results of the evaluation showed that the identification of ergonomic hazards in the furniture industry was found in all furniture work processes from wood cutting to finishing.

Abstract

Senam pernapasan merupakan olah napas yang bermanfaat untuk meningkatkan kesehatan dengan mengefektifkan semua organ dalam tubuh secara optimal. Pada era pandemi COVID-19 penggunaan masker telah menjadi kebiasaan baru termasuk pada saat berolahraga. Salah satu faktor yang mempengaruhi penggunaan masker yaitu perbedaan gender dimana laki-laki dan perempuan memiliki kapasitas volume paru-paru yang berbeda. Penelitian ini bertujuan untuk mengetahui pengaruh perbedaan gender dan penggunaan masker terhadap tanda-tanda vital, Borg scale dan kadar CO peserta senam pernapasan di RSUD Dr. Saiful Anwar Malang.  Pengamatan dilakukan pada 3 (tiga) sesi yaitu sebelum senam, tahap inti, aerobic, dan setelah tahap pendinginan.  Penelitian menggunakan data sekunder dengan pendekatan metode analitik case control, kemudian dilakukan uji independent t test dan Mann Whitney. Hasil analisis tanda-tanda vital pada setiap sesi menunjukkan bahwa tidak terdapat perbedaan heart rate, respiratory rate, dan kadar CO yang signifikan antara kelompok yang bermasker dan tidak bermasker pada gender yang berbeda (p > 0,05). Sedangkan terdapat perbedaan saturasi oksigen yang nyata (p < 0,05) antara kedua kelompok tersebut pada tahap inti, aerobik, dan setelah pendinginan. Hasil analisis Borg scale menunjukkan berbeda (p < 0,05) pada tahap inti dan aerobik. Penelitian ini menunjukkan bahwa tidak ada perbedaan bermakna penggunaan masker pada senam pernapasan pada gender laki laki maupun perempuan untuk kelompok dewasa muda sehat sehingga penggunaan masker bedah mungkin dapat di pakai ketika melakukan senam pernapasan dalam era pandemi COVID-19.
