Abstract

ABSTRAK Daun Jawer kotok (Plectranthus scutellaroides), diketahui banyak mempunyai manfaat dalam pengobatan tradisional, salah satunya  sebagai antiinflamasi yang menjadi langkah awal pencarian anti kanker dari tumbuhan. Kanker adalah suatu penyakit yang diakibatkan terjadinya mutasi gen yang mengakibatkan kelainan pada kromosom. Penelitian ini bertujuan untuk mengetahui aktivitas pro-apoptosis fraksi polar daun jawer kotok (Plectranthus scutellaroides) terhadap ekor larva katak Rana catesbeiana saat stadium metamorfosis klimaks. Fraksi polar daun jawer kotok diperoleh melalui fraksinasi ekstrak etanol yang diperoleh dengan cara maserasi  dalam pelarut non polar, semi polar, dan pelarut polar. Metode pengujian aktivitas pro-apoptosis adalah dengan menghitung frekuensi sel apoptosis diantara sel nekrosis dan sel normal per seribu sel per ekor larva katak di bawah mikroskop. Jenis kematian sel ditentukan dengan pewarnaan Giemsa. Hasil pengujian menunjukkan bahwa fraksi polar daun jawer kotok dengan beberapa variasi   dosis mempengaruhi  frekuensi sel apoptosis jika dibandingkan dengan kontrol (a = 0,05). Frekuensi sel apoptosis tertinggi ditunjukkan oleh dosis 500 mg/kg BB yaitu sebesar 79,51%. Dari hasil penelitian dapat disimpulkan bahwa fraksi polar daun jawer kotok  menunjukkan  aktivitas pro-apoptosis pada ekor larva katak Rana catesbeiana stadium metamorfosis klimaks.Kata kunci :   Plectratus scutellaroides, apoptosis, Rana catesbeianaABSTRACTLeaves of jawer kotok (Plectranthus scutellaroides) has many benefits in tusing of traditional medicine, one among  them is antiinflammation which can be starting point of seeking and discovering of anticancer from plants. Cancer is one of disease due to gen mutation that effect anomaly of chromosom. The purpose of this experiment was to identify the pro-apoptotic activity of polar fraction of jawer kotok leaves on Rana catesbeiana tadpole tail in climax metamorphosis phase. Polar fraction was obtained through fractination of ethanolic extract which are made by maceration method in non polar, semi polar, and polar solvents.  The method use in this experiment was calculating the apoptotic cell frequency between necrotic cells and normal cells which were counted per one thousand of tadpole tails cells under microscope. The modes of cell death determined by Giemsa staining. The experiment showed that variation of dose of polar fraction of jawer kotok leaves is affecting  the frequency of apoptotic cell death compared with control (a = 0.05). Dose of 500 mg/kg body weight of water fraction of jawer kotok leaves gave the highest apoptotic cell frequency (79.51 %) between normal and necrotic cell. It can be concluded that polar fraction of jawer kotok leaves has pro-apoptotic activity on Rana catesbeiana tadpole tail in climax metamorphosis phase.Keywords: Plectranthus scutellaroides, apoptosis, Rana catesbeiana

Abstract

Plectranthus  scutellarioides  (L.)  R.Br.,  or  jawer  kotok,  Family  Lamiaceae, grows  widely  in  Indonesia,  and  has  a  long  history  of  therapeutic  usage  in Indonesian traditional jamuto cure various diseases. The brownish purple leaves of  Plecranthus  contain  alkaloids,  saponin,  flavonoids,  tannin,  volatile  oils,  and quercetin  which  has  been  proven  to  exert  antiinflammatory  activity.  In  this research,  a  determination  of  quercetin  in  Plecranthus  leaves  extract  was performed and followed by a study of its interaction with histamine H4 receptor to  understand its  anti-inflammatory  activity.  The  dry  leaves  were  macerated by using  a  mixture  of  methanol  and  water  (1:1)  for  48  hours  and  the  solvent  was evaporated  at  low  temperature  (40-50oC).  Analysis  of  quercetin  in  the  extract was performed by using reversed-phase HPLC method LC-10AT VP (Shimadzu), Atlantis  Hilicsilica  C18  (Waters®)  150  mm  x  4.6  mm,  5  µm  as  stationary  phase and  a  mixture  of  acetonitrile,  phosphoric  acid,  and methanol  (40:50:10),  flow rate 0.8 mL/minute.  In silicostudy of quercetin with histamine H4 receptor was performed by using AutoDock Tools 3.0.5. Histamine H4 receptor (H4R) belongs to  G  protein-coupled  receptors  which  is  involved  in arthritis,  asthma,  and inflammations.  The  3D  structure  model  of  H4R  was  built  by  using  MODELLER 9v7.  Quercetin  contained  in  Plecranthus  leaves  extract  was  0.05  %.  This compound interacted with H4R viahydrogen bond formation with Lys158 (2.006 Å)  and  Glu182  (2.048  Å),  and  van  der  Waals  interaction  with  Trp90,  Leu91, Asp94, Tyr95, Phe168, Thr178, Ser179, Tyr319, Phe344, and Tyr340, therefore Plecranthus  leaves  extract  might  have  a  chance  to  be  used  as  histamine  H4 receptor inhibitor.Key  words  :   histamine  H4  receptor,  in  silico  study,  Plecranthus  leaves,  Plectranthus scutellarioides, quercetin

Abstract

Drugs consultation by pharmacist is an important service to improve the quality of medication use in patients with type 2 diabetes mellitus (T2DM). Perception of patients regarding this service is very useful to boost pharmacists confidence to conduct such service. The purpose of this study is to provide an overview regarding interests and categories of drugs consultation needed by T2DM patients and to determine the effect of quality of drugs consultation on patients interest to participate in such program. Survey and interviews were conducted with T2DM patients in outpatient clinic, Dr. M. Salamun Air Force Hospital, Bandung during April-June 2013. One hundred patients were recruited for this study. Most of the patients consumed combination of oral antidiabetic and non-antidiabetic medication (71 %) and were given polypharmacy (45 %). The results showed that the information regarding drugs interactions, adverse drugs reaction (ADR), and drugs dosage were among the aspects that should be included in drugs consultation service by pharmacists. The quality of drug information service was associated with increased interest to participate in such program. These aspects should be considered in the implementation of drugs consultation program by pharmacists.Keywords: drugs consultation, T2DM, pharmacists

Abstract

 Crotalaria pallida Aiton (C. pallida Aiton) is empirically used as dietary supplement to treat cancer by the people of North Sulawesi. However, its scientific pharmacology activity has not been explored yet. Therefore, this study was conducted to evaluate anti-proliferative activity of C. pallida Aiton on MCF-7 breast cancer cells. The extraction of leaves and seeds were performed using ethanol, ethyl acetate, n-hexane, and water. Phytochemical screening was then performed to identify secondary metabolites in this extract. Anti-proliferative activity was evaluated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The results indicated that ethyl acetate fraction of C. pallida Aiton has the lowest IC50 (29,67). In conclusion, ethyl acetate fraction of C. pallida Aiton is potential to be developed as anti cancer agent.Keywords: Crotalaria pallida Aiton, WST assay, MCF-7 cell line

Abstract

Virus influenza A (H5N1) merupakan wabah utama yang berpotensi menyebabkan epidemik di seluruh dunia. Telah ditemukan banyaknya kasus resistensi terhadap obat antivirus influenza A sehingga penggunaannya mulai dibatasi. Oleh karena itu, perlu dilakukan penelitian untuk melakukan pengembangan obat baru yang lebih efektif dan aman untuk melawan virus influenza A. Tanaman herbal menjadi salah satu sumber pengembangan obat antivirus baru. Dalam penelitian ini, dilakukan pengujian aktivitas antivirus dari ekstrak dan fraksi ekstrak daun miana (Plectranthus scutellarioides (L.) R. Br) melalui penentuan IC50 terhadap enzim neuraminidase menggunakan metode MUNANA Assay. Konsentrasi optimal enzim neuraminidase yang digunakan adalah 1,25 U/mL. Validasi metode dilakukan melalui penentuan IC50 dari inhibitor DANA terhadap enzim neuraminidase yaitu sebesar 15,72 µg/mL. Hasil pengujian aktivitas inhibisi enzim neuraminidase oleh ekstrak etanol, fraksi etil asetat, fraksi n-heksan dan fraksi air daun miana secara berturut-turut adalah 339,1 µg/mL; 125,6 µg/mL, 355,4 µg/mL  dan 288,5 µg/mL. Dari seluruh sampel yang diuji, fraksi etil asetat memiliki aktivitas potensial penghambatan yang paling baik.

Abstract

Various ethnics of the world have a long history on using their surrounding as the source of foods. Foods which are act as product of traditional ethnic knowledge known as traditional and ethnic foods (TEF). Vegetables are one of the main ingredients of TEF. As sources of many combination of phytonutrients, vegetables offer various health benefits. As a country with long history of agricultural practice, Indonesia also has abundant knowledge of how Indonesian ethnics process and consume vegetables.Vegetables, in Sundanese tradition, usually consumed as “lalap” or “lalapan”. Some lalapan are cultivated vegetables and some others are come from wild sources. This article discuss the health benefits of wild source lalapan, which can be found in natural habitat.
