Abstract

Periodontitis is an inflammatory disease affects teeth supporting tissues which causing loss of periodontal attachment and damaging alveolar bone. Periodontitis trigger host to produce monocytes, polymorphonuclears (PMN), macrophages and others to release inflammatory mediators such as IL-1, TNF-α, PGE2. In periodontal disease, PGE2 role in breakdown of alveolar bone. The biomarker used PGE2 that contained in human saliva. This research aims to prove that PGE2 in saliva could be a new method to measure severity and early diagnosis of periodontitis. This research used Post Test Only Controlled Group Design with human saliva as the samples. The samples collected from 20 patients divided into two groups which are: control group (patient without periodontitis) and experimental group (patient with periodontitis). Saliva collecting method which used was Passive Drool with 2 ml for each. Afterwards, the samples isolated with a kit to make sure the PGE2 in saliva active and then the ELISA test did. Data analysis used Independent T Test with p<0,05 to know comparison the level of PGE2 in both groups. Thereafter, a ROC curve analysis with p<0,05 used to find out sensitivity and specifity of PGE2 in saliva towards periodontitis. The amount of PGE2 in saliva were significantly increased in periodontitis patients compared with healthy patients, also PGE2 in saliva specific enough and sensitive to periodontitis so its could be used as a valid diagnostic tool in measuring the severity of periodontitis. Keywords:  Prostaglandin E2, saliva, periodontitis, early diagnose, severity

Abstract

Abstract

Dental and oral health has an important role in supporting the health of the human body. Diseases and disorders of the teeth in school-age children between 7-15 years are disturbances in growth and development. However, many factors still influence the occurrence of dental and oral problems. One of them is the lack of attention and knowledge. At the age of children, parents and teachers have an important role in guiding, providing understanding, reminding and providing facilities to maintain healthy teeth and mouth. Therefore, all three of them need to know to underlie the formation of behaviors that support children adopting a healthy lifestyle. Based on these conditions, we conducted community service to increase this knowledge through the School Health Program, which was implemented in 7 elementary schools in Malang City. In this activity, education was carried out for students, parents and teachers with a total of 325 participants. The results of this program show an increase in knowledge by students, teachers and parents. The knowledge conveyed included basic dental care recommendations such as brushing teeth twice a day with fluoride toothpaste, managing good eating habits, staying away from bad habits that damage teeth and preventing dental caries as early as possible. After completing this activity, participants can adopt a healthy lifestyle to prevent problems with their teeth and mouth.

Abstract

Introduction: Periodontal disease, including periodontitis, ranks as the 6th complication for Diabetes Mellitus. Research has found an increase of inflammation and higher alveolar bone loss in the periodontal of diabetes mellitus patients. One of the components which exhibited multiple bio-activities like anti oxidative, anti-inflammatory, antidiabetic antimicrobial cardioprotective, neuroprotective, immunomodulatory, and anti-obesity activities is procyanidin. Procyanidin can be found in various natural ingredients and one of the most abundant compositions is found in red grape seeds. Methods: The method used in this study is a literature review from several academic databases including : Google Scholar, PubMed, Mendeley, Wiley, and the university's online library with boolean searching method. Results: The several articles used in this literature review show that the protective effect of grape seed procyanidin plays a role in attenuating the increase of MFG-E8, suppressing the increase of IL-1beta protein level, improving HOMA-IR and insulin levels in diabetes, increasing IL-10 and TGF-beta accumulation in the gingival epithelium of periodontitis condition, and MMP- and HIF-1alpha immunohistochemistry showed an increase in its levels for periodontitis with diabetes rats, but decreased as grape seed procyanidin extract (GSPE) was administered. Conclusion: The present study suggested that procyanidin in grape seed extract played a role in treating periodontitis in diabetes patients through its anti- inflammatory role and its ability to decrease alveolar bone loss. There are still very few studies regarding this topic, therefore

Abstract

Background: Dental Pulp Stem Cell (DPSC) is one of the ideal mesenchymal stem cells (MSCs) for the repair of damaged cells, as it has great proliferative potential and the ability to regenerate pulp-like complex dentins. However, DPSC has limitations in providing stem cell sources due to its small number. Therefore, it is induced with umbilical cord cell isolate (hUC-MSC) which is currently widely developed because of its advantages that are easy to obtain, have proliferative potential, and have anti-inflammatory effects. However, its influence and effectiveness on Dental Pulp Stem Cells is not widely known. Objective: To prove the effect of umbilical cord cell isolate on the expression of Stro-1 and CD 34 in Dental Pulp Stem Cell (DPSC) culture. Methods: This study is experimental laboratory with simple random sampling which is divided into 4 groups, namely 2 control groups and 2 groups of umbilical cord isolate administration which are observed for 12 hours and 24 hours. Observation of expression by immunohistochemical staining was then calculated using Qupath software in 5 fields of view. Results: The treatment group that was given umbilical cord cell isolate showed the highest increase in Stro-1 and CD 34 expression at 24-hour observation compared to the untreated group. Conclusion: There was an increase in the expression of Stro-1 and CD 34 in Dental Pulp Stem Cell cultures after being given umbilical cord cell isolate.
