Abstract

Background: Gonad differentiation in some teleostei is a critical phase of gonad development. Gonad differentiation in some fish such as the tilapia group is influenced by environmental factors, namely temperature. The aim of this study was to determine the differentiation process of Nilem fish gonads (Osteochilus vittatus) under the influence of temperature. Methods: One day after fertilization (days postfertilization/dpf) fish were exposed to different medium temperature ranges, namely: 28-29 oC, 30-31 oC, 32-33 oC, and room temperature as a control for 35 days. The percentage of fish survival was calculated on the 35th day by calculating the number of surviving fish divided by the total number of fish in 1 tank times 100%. Thirty fish in each treatment were measured for body length at day 10 dpf and day 35 dpf to determine fish growth. Six fish 35 dpf from each treatment were fixed in 70% alcohol for making histological preparations using the paraffin method, Haematoxylin-Eosin staining. Fish survival data growth data in the form of fish length were analyzed using Anova. Gonadal differentiation data were analyzed descriptively. Results: There was no significant difference in the growth of fish in all treatment and control groups. The average survival rate of fish in all treatment and control groups was 100%. Observation of the gonads showed that the gonads were not differentiated in all treatment and control groups. Conclusion: The room temperature range up to 33 oC supports the growth of Nilem fish with gonad development in the indifferent gonad stage.

Abstract

Pemberdayaan masyarakat tani dengan sasaran petani di Desa Dawuhan, Kecamatan Banyumas, Kabupaten Banyumas merupakan program pengabdian masyarakat yang didanai oleh LPPM Universitas Jenderal Soedirman melalui skema Program Penerapan IPTEKS. Tujuan jangka panjang dari pemberdayaan masyarakat tani ini agar petani dapat mengusahakan budidaya tanaman padi secara organik. Dengan budidaya padi secara organik, maka petani tidak saja diberdayakan untuk mendapatkan hasil produksi yang meningkat, namun aspek keberlanjutan usahatani, kemandirian petani, daya saing produk yang dihasilkan, daya tawar petani dan keramahan lingkungan akan dicapai. Namun dalam rangka mencapai tujuan jangka panjang tersebut, permasalahan awal yang dihadapi oleh petani yakni masih rendahnya tingkat ketrampilan petani (psikomotorik) dalam pembuatan Pupuk Organik Cair (POC) dan Pestisida Nabati (Pesnab). Demi mengatasi permasalahan tersebut, Program Penerapan IPTEKS dilaksanakan dengan metode berupa pelatihan pembuatan pupuk organik cair dan pestisida nabati yang dilaksanakan di Desa Dawuhan, Kecamatan Banyumas, Kabupaten Banyumas. Pupuk organik cair dan pestisida nabati dibuat dengan memanfaatkan sumberdaya alam sekitar, karena bahan baku yang dibutuhkan relatif banyak di lingkungan pedesaan. Dengan adanya kegiatan pemberdayaan masyarakat tani ini, petani di Desa Dawuhan menjadi lebih terampil dan mampu dalam membuat pupuk organik cair dan pestisida nabati, sehingga dapat dimanfaatkan langsung untuk budidaya tanaman padi secara organik

Abstract

Kegiatan Penerapan IPTEKS dengan khalayak sasaran di Kelompok Tani Ngudi Raharjo III Desa Lemberang ini merupakan program pendampingan penguatan kelembagaan kelompok tani, serta adopsi teknologi benih di lahan persawahan milik anggota. Permasalahan yang dihadapi oleh khalayak sasaran dikelompokan menjadi tiga bagian, yakni : (a) Ketidakaktifan petani anggota dalam kelompok tani; (b) Belum berjalannya instrumen kelembagaan kelompok tani yang baik; dan (c) Pasokan air di lahan persawahan yang kurang, serta belum adanya teknologi benih yang cocok. Untuk mengatasi permasalahan tersebut, Program Penerapan IPTEKS dilaksanakan berupa : (1) Penyuluhan dan pemaparan terkait keuntungan berpartisipasi aktif dalam sebuah kelompok tani. Dengan penyuluhan ini diharapkan dapat merubah pemahaman, perasaan dan psikomotorik petani untuk dapat terlibat dalam setiap kegiatan kelompok. (2) Rembug Tani / FGD, dimana kelompok tani melakukan diskusi dan kesepakatan dalam pembuatan susunan pengurus, kegiatan kelompok, jadwal rutinan, pembagian tugas, dan kelengkapan instrumen kelembagaan lainnya. (3) Penyuluhan mengenai keunggulan teknologi benih INPAGO UNSOED-1, Pengelolaan Tanaman Terpadu (PTT) dan penentuan lokasi Demonstration Plot (Demplot). (4) Pelaksanaan demplot penanaman benih INPAGO UNSOED-1 sekaligus monitoring kegiatan usahatani. Dari semua rangkaian kegiatan ini diharapkan dapat mendukung peningkatan produksi pertanian khalayak sasaran.

Abstract

Primordial Germ Cells (PGCs) merupakan prekusor gamet yang penting dalam reproduksi ikan. PGCs dialokasikan di luar gonad dan bermigrasi menuju gonad primordia selama masa perkembangan embrional ikan. Pengetahuan mengenai alokasi dan migrasi PGCs penting dalam pengembangan model penelitian gametogenesis dan peningkatan produksi akuakultur. Penelitian ini bertujuan mengetahui lokasi PGCs pada tahap embrio ikan nilem menggunakan metode hibridisasi in situ dengan penanda vasa germ cell ikan nilem. Embrio ikan nilem umur 30 menit, 60, 180 dan 240 menit paska fertilisasi difiksasi dalam NBF 10% kemudian didechorionasi. Selanjutnya dilakukan hibridisasi in situ dengan penanda vasa germ cell ikan nilem. PGCs pada embrio ikan nilem ditemukan pada daerah di sekitar alur pembelahan blastomere pada embrio umur 30 menit, 60 menit dan terdeteksi pada blastoderma di daerah kutub animalis pada embrio umur 18 menit dan 240 menit yang telah memasuki tahapan blastula dan gastrula.
