Abstract

Osphronemus gouramy is a potential Indonesian fish that needs to be supported by good nutritional feed and water quality in a culture system. The research on the effect of combining Chlorella vulgaris (C. vulgaris) supplementation with culture systems of biofloc and non-biofloc in gourami fish on amylase activity and blood urea level has not been conducted. Therefore, this study aims to determine the effect of C. vulgaris supplementation with different culture systems on the amylase activity of various digestive organs and blood urea levels. It obtains the best C. vulgaris supplementation feed combination to increase the amylase activity of various digestive organs and reduce blood urea levels. This research was carried out experimentally with a Factorial Completely Randomized Design consisting of two factors. Furthermore, gourami was fed with C. vulgaris levels of 0, 2, 3, 4, and 5 g kg-1 for 28 days and divided into biofloc and non-biofloc groups of culture systems. The amylase activity was measured at three pH buffers, and blood urea levels were analyzed using a urea kit. The results showed that amylase activity increased when supplemented with C. vulgaris 5 g kg-1 feed using the biofloc system, while blood urea levels decreased. C. vulgaris supplementation and biofloc system are alternative solutions to improving nutrition and water quality.

Abstract

Primordial Germ Cells (PGCs) merupakan prekusor gamet yang penting dalam reproduksi ikan. PGCs dialokasikan di luar gonad dan bermigrasi menuju gonad primordia selama masa perkembangan embrional ikan. Pengetahuan mengenai alokasi dan migrasi PGCs penting dalam pengembangan model penelitian gametogenesis dan peningkatan produksi akuakultur. Penelitian ini bertujuan mengetahui lokasi PGCs pada tahap embrio ikan nilem menggunakan metode hibridisasi in situ dengan penanda vasa germ cell ikan nilem. Embrio ikan nilem umur 30 menit, 60, 180 dan 240 menit paska fertilisasi difiksasi dalam NBF 10% kemudian didechorionasi. Selanjutnya dilakukan hibridisasi in situ dengan penanda vasa germ cell ikan nilem. PGCs pada embrio ikan nilem ditemukan pada daerah di sekitar alur pembelahan blastomere pada embrio umur 30 menit, 60 menit dan terdeteksi pada blastoderma di daerah kutub animalis pada embrio umur 18 menit dan 240 menit yang telah memasuki tahapan blastula dan gastrula.

Abstract

merupakan kelompok yang membudidayakan pembesaran ikan gurami dan ikan lele. Kelompok ini memiliki masalah dalam penyedian pakan ikan. Pengabdian masyarakat ini bertujuan memberikan ketrampilan dalam budidaya larva Hermentia illucens sebagai pakan alternatif untuk ikan. Metode yang digunakan dalam pengabdian kepada masyarakat ini adalah sosialisasi program, paparan materi, pembutan demplot budidaya larva Hermentia illucens, pendampingan, dan evaluasi program. Evaluasi diberikan dengan memberikan pre-test dan post-test. Melalui transfer pengetahun dan praktek langsung dalam bentuk demplot, POKDAKAN memiliki pengetahuan dan ketrampilan dalam cara penetasan telur black soldier fly (BSF), manajemen pemeliharaan larva usia 1-7 hari, larva sebelum fase prepupa dan pupa, menetaskan pupa menjadi BSF, manajemen pemeliharaan BSF, menstimulasi BSF bertelur. Selain itu mitra memiliki ketrampilan dalam pengembangan rak inkubasi telur, modifikasi pembuatan biopond, modifikasi tempat bertelur, dan mampu memilih media yang tepat untuk pertumbuhan larva. Dengan demikian dapat disimpulkan bahwa POKDAKAN sudah memiliki pemahaman dan keterampilan dalam budidaya larva  Hermentia illucens. Guna untuk memanfaatkan larva Hermentia illucens sebagai pakan ikan maka POKDAKAN berkomitmen akan menambahkan siklus budidaya larva teserbut agar mempoleh induk yang cukup digunakan menghasilkan telur.  Kata Kunci: BSF, Budidaya Hermentia illucens, POKDAKAN, Pemberdayaan

Abstract

Potassium dichromate, K2Cr2O7, is a well-known heavy metal, commonly used as an oxidizing and tanning agent in industrial applications. Prolonged or repeated exposure of hexavalent chromium is deemed very toxic for aquatic biota, with long lasting effect. This substance induce damage to DNA and tissue structures, as well as disruption of survival and growth rate. The present research exposed Osteochilus vittatus in larval and juvenile stages, to varying concentration of K2Cr2O7. This experiment was aimed to evaluate the effect of chromium on primordial germ cells (PGCs) and subsequently, to the subject gonadal development. The evaluation was based on paraffin-embedded section, stained with Haematoxylin-Eosin. K2Cr2O7 of 2.5 and 5 ppm were applied to four crucial developmental stages; post-hatching larvae, 1-month, 2-months, and 3-months juvenile, for 30 days. There was a consistent pattern in all test subject, in which higher concentration of K2Cr2O7 resulted in lower PGCs number and delayed gonadal appearance. Our results suggested that sublethal Cr exposure to larval stage potentially decrease PGCs and thus, hinder the formation of gonad. Regulation of Cr-containing waste disposal should be issued in near future, to prevent further damage on local freshwater fish.Keywords: Chromium, Cyprinids, Juvenile, Larvae, PGC

Abstract

An integral aspect of anticancer experimentation involves delineating the optimal dosage of the test compound to ascertain its efficacy in targeting malignant cells. Numerous variables may influence a compound's cytotoxicity, among which is the choice of cell culture medium. Within in vitro settings, supplementary mediums are employed to foster cellular proliferation. Platelet lysate (PL) serves as a growth supplement, presenting an alternative to fetal bovine serum (FBS), primarily due to its incorporation of growth factors such as platelet-derived growth factor (PDGF), a component absents in FBS. The integrity of PL may be subject to various factors, including the age of the donor. This study sought to evaluate the impact of donor age on PL quality. Furthermore, it aimed to discern whether PL derived from platelet concentrate (PC) blood components of different age cohorts influences the IC50 value in anticancer compound assessment. Expired PCs were utilized, subsequently classified into age categories: ≤30 years, >30 years, and a combination of ages. PL analysis encompassed parameters such as pH, blood profile, protein, glucose, and cholesterol levels. The investigation scrutinized the influence of PL quality, as a cellular growth supplement, on the anticancer compound cisplatin's activity against HeLa cells. Findings indicate that donor age influenced the IC50 value of cisplatin on HeLa cells. Notably, elevated cholesterol levels and decreased pH in PL from donor ages >30 years were associated with reduced cisplatin toxicity.Keywords: Cisplatin, Donor Age, HeLa, IC50, Platelet Lysate.
