Abstract

Striped catfish (Pangasianodon hypophthalmus) spawning success is strongly influenced by environment.Striped catfish spawning generally occurs during the rainy season while in the dry season is difficult to find mature female. This research was conducted to improve reproduction performance of female striped catfish during dry season through combination injection of hormone pregnant mare serum gonadotrophin (PMSG) and human chorionic gonadotrophin (HCG).The treatments were A : control (no hormone injections), B : injection of PMSG 10 IU/kg + HCG 10 IU/kg and C : injection of PMSG 20 IU/kg + HCG 10 IU/kg.Injections were done at interval of two weeks as many as six times. The research was conducted during dry season in June-October 2012 with rainfall 0.20- 14.40 mm/years. The results showed combination injection of PMSG + HCG could improve female reproduction performance. The best treatment was shown on C treatment with values of OSI (ovi somatic index), fecundity, fertilization rate, hatching rate and larva production was 12.18 %, 523 000 eggs/female, 79.65 %, 69.93 % and 290 000 larvae/female respectively. As for control treatment (A) the value of OSI, fecundity, fertilization rate, hatching rate and larval production, was 6.45 %, 197 000 eggs/female, 48.55 %, 35.59 % and 34 000 larvae/female, respectively

Abstract

Vitamin E merupakan salah satu mikronutrien penting yang berpengaruh terhadap performa reproduksi ikan. Penelitian penambahan vitamin E dalam pakan induk ikan nila (Oreochromis niloticus) bertujuan untuk mengetahui pengaruh vitamin E terhadap perkembangan gonad dan performa pemijahan induk ikan nila. Pakan uji yang digunakan adalah pakan formula dengan kadar protein 35% dan lemak 10% ditambahkan vitamin E (tingkat kemurnian 50%) dengan dosis A) 0, B) 75, C) 150, D) 225, E) 300 and F) 375 mg/kg pakan (kontrol). Induk ikan nila umur 6-7 bulan sebanyak 180 ekor dipelihara dalam 6 jaring ukuran 3x3x1,5m yang diletakkan dalam kolam 6000 m2 dengan kedalaman air 80 cm. Masing-masing jaring diisi dengan 20 ekor induk betina dan 10 ekor induk jantan dan dipelihara selama 13 minggu pada bulan September sampai dengan November 2011. Hasil penelitian menunjukkan bahwa penambahan vitamin E pada pakan induk ikan nila memberikan nilai indeks gonad somatik lebih tinggi dibandingkan yang kontrol. Penambahan vitamin E sebanyak 225 mg/kg pakan memberikan nilai persentase induk matang gonad yang tertinggi (80%) dan frekuensi pemijahan yang terbanyak yaitu 22 kali selama masa pemeliharaan 13 minggu. Berdasarkan hasil penelitian yang diperoleh dapat disimpulkan bahwa penambahan vitamin E sebanyak 225 mg/kg pakan dapat meningkatkan perkembangan gonad, jumlah induk betina yang matang gonad, persentase total pemijahan dan frekuensi pemijahan pada induk ikan nila.

Abstract

Pasupati is a pangasiid catfish resulted from hybridization between female striped pangasiid (Pangasianodon hypophthalmus) and male jambal (Pangasius djamba). Pasupati is known as a superior white-fleshy pangasiid catfish promoted for an export commodity. However, its reproductive characteristic in relation to its fertility has not been examined yet. The present study was conducted to investigate the successful of fertilization between female and male pasupati, reciprocal cross between pasupati with striped catfish, and reciprocal cross between pasupati with jambal catfish.Two to three females and males pasupati were used in the present study. All of breeders were hormonally induced for artificial. The result showed that there is no embryogenesis occurred when sperm from pasupati, striped and jambal catfish were used to fertilize eggs of pasupati catfish. On the other side, sperm enabled to fertilize eggs of striped cat fish producing low percentage of larvae with maternaly similar morphological appearances at juvenile stage. This phenomena showed that spontaneous gynogenesis occurred. In conclusion, female and male of pasupati cat fish are sterile.

Abstract

Patin nasutus (Pangasius nasutus Bleeker, 1863) (Pangasiidae; Pisces) is a potential pangasiid catfish species to be cultured in Indonesia.Many efforts has been done to explore the potency of P.nasutus for aquaculture purposes.However, basic knowledge is still very rare such as its embryonic and larval development.The present study aimed to understand the embryonic and larval development of P. nasutus resulted from induced spawning of P.nasutus brooders at Research Institute for Fish Breeding, Sukamandi. Those embryonic and larval development were observed microscopically.The results showed that diameter of the ovulated oocytes ranged 1.40-1.65 mm and fertilized eggs ranged 1.90-2.15 mm.The embryogenesis consisted of several stages as follow: morula, blastula, gastrula, organ developments and newly hatched larvae occured within periods of (minutes) 20-200, 190-220, 210-660, 600-1,140 and 1,800-2,160 after fertilization at water temperature of 28-29oC. Total length of the newly hatched larvae ranged 4.80-5.10 mm with anterior, posterior and dorsal parts of the yolk sac coloured black pigmentation. On caudal peduncle, the black spot existed since 36 hours after hatching was a strong character for P.nasutus larvae.Volume of the yolk sac was 2.65±0.14 mm3, for which 50% absorbed at 36 hours after hatching and relative fully absorbed at 78 hours after hatching. The larvae have complete morphology as adult after seven days. During early developmental stage, larvae showed cannibalism since the third day, corresponding to the first exogenous feeding.

Abstract

Pangasianodon hypophthalmus is an introduced species of fish culture and fisheries resources in Indonesia. Reproductive biology aspects, e.g. maturity, plays an important role on the fisheries management. There was no detail information of oocytes development of P. hypophthalmus. The aim of this study is to find out the detail of its oocytes development through the observation of whole oocytes and ovarian histological slides microscopically. The results showed that the rhythm of oocyte development of P. hypophthalmus was grouped synchronism, assigned by the presence of two oocyte groups in the mature females, i.e. the large and maturing oocytes which would be spawned soon, and the small ones as the stock and still unyolked for the next spawning. The oocyte development could be devided into five stages, i.e. stage 1 (chromatin nucleolar and perinucleolar) with oocytes diameter less than 300 urn, stage 2 (yolk vesicles and cortical alveolar) with oocytes diameter of 230-660 urn, stage 3 (yolk granules) with oocytes diameter of 430-1,100 urn, stage 4 (migratory nucleus and hydrated) with oocytes diameter of 950-1,260 urn, and stage S (atretic) with oocytes diameter less than 800 m

Abstract

The feeding rate and the feeding frequency influencing the growth and survival rate of fish. The objective of this research was to know the proper level of feeding frequency for supporting the growth of patin pasupati (hybride between Pangasius hyphophthalmus and P. djambal).The experiment was conducted in fibre glass tanks located at Research Instalation for Genetic and Freshwater Aquaculture Technology, Sukamandi. Patin pasupati fingerling with average body weight of 2.23 ± 0.48 g were stocked in 9 fibre glass tanks, each filled with 30 litres of water.The experimental design was Completely Randomized Design with 3 treatments and 3 replications. The treatments were feeding frequency as follows 1, 3 and 5 times per day. The feeding rate was 8% of total body weight per day.The parameters were specific growth rate, average body weight gain, feed conversion ratio, protein retention,lipid retention, protein efficiency ratio and survival rate. The results of experiment indicated that there were significant different(P<0.05) among treatments for specific growth rate, individual body weight gain, feed convertion ratio,protein retention, lipid retention and protein efficiency ratio.The highest value for specific growth rate (7.03%) and protein retention (54.59%) were found for feeding frequency 5 times per day.The values of feed conversion ratio and survival rate for feeding frequency 5 times per day were 0.83 and 100%, respectively.

Abstract

Perkasa pangasiid (Pangasianodon hypophthalmus) is a superior catfish selected by the family that has been released by the Minister of Marine and Fisheries as aquaculture fish which has a selection response of 38.86 %. Genetic quality improvement to accelerate growth can be done through selection activities. The performance test of the Siamese catfish from the selection (Perkasa strain) compared to the non-selection strain (local strain) originating from the farmer. The purpose of this study was to compare the productivity of selection and non-selection Siamese catfish cultivation maintained in a net pond. The initial seeds of the test fish were 3.76 g to 3.79 g with a total length of 2 inches to 3 inches kept in a net pond measuring 3 × 5 × 1.25 m. The water level was around 100 cm with a density of 175 fish/net. Maintenance was carried out for six months. Test parameters observed included: weight gain, specific growth rate, biomass harvest, productivity and cost of production. The results showed that Siamese catfish which were selected for six months of maintenance had a final weight of 698.40 ± 164.54 g and non-selection strains of 595.77 ± 103.41 g. As well as having a higher specific growth rate (3.51 %), higher harvest biomass (11.44 %), and higher productivity (18.71 %) than non-selection Siamese catfish. Based on the bioeconomic analysis, the selection of enlarged Siamese catfish cultivation can reduce production costs up to Rp 390 /kg. The use of Perkasa Pangasiid superior seeds can increase harvest biomass higher than the non-selection seeds.

Abstract

Konten DNA inti (KDI) suatu spesies atau populasi terbentuk melalui berbagai mekanisme seperti mutasi kromosom, insersi dan delesi, transposable element, dan duplikasi. Selain itu, hibridisasi dan posisi spesies dalam filogeni juga berperan mempengaruhi keragaman KDI. Penelitian in bertujuan menginvestigasi profil KDI pada empat spesies ikan patin dari genus Pangsius yang meliputi Pangasionodon hypophthalmus, Pangasius djambal, Pangasius nasusutus, Pangasius nieuwenhuisii, hibrida P. hypophthalmus betina dan P. djambal jantan (Hibrida HD), dan hibrida P. hypophthalmus betina and P. nasutus jantan (Hibrida HN). Sel darah merah diambil dari masing-masing spesies dan pengukuran KDI dilakukan menggunakan Attune Acoustics Flow cyometer (ABI), dengan pewarna DAPI dan sel darah merah ayam, Gallus domesticus, sebagai kontrol standar. Hasil penelitian menunjukkan rerata KDI P. hypophthalmus, P. djambal. P. nasusutus, P. nieuwenhuisii, adalah 0.960±0.0254 pg, 1.017±0.0510 pg, 1.000±0.0410 pg, 1.074±0.0231 pg, berada dalam kisaran KDI famili catfish lainnya. KDI Hibrida HD dan Hibrida HN masing-masing 005±0.0358 and 0.956± 0.0089. Diantara species tetua, KDI terendah terdapat pada P. hypophthalmus dan berdeda nyata (P<0.05) dengan KDI P. djambal dan P. nieuwenhuisii tetapi tidak berbeda (P>0.05) dengan P. nasutus. KDI Hibrida HD dan Hibrida HN tidak berbeda dengan masing-masing tetuanya. Namun, pola KDI kedua hibrida berbeda dimana Hibrida HD berada diantara kedua tetuanya sedangkan Hibrida HN berada di bawah kedua tetuanya. Secara filogenetik, keragaman KDI pada genus Pangasius tidak mengikuti pola kekerabatan sesuai hubungan filogenetik berdasarkan marka genetik sitoplasmik dan inti. Kata kunci: Flow cytometry, konten DNA inti, P.hypophthalmus, P. djambal, P. nasutus, P. nieuwenhuisii, hibrida.
