Abstract

Difficulties in finding anti-diabetics which are effective, tolerable by the patients, safe, cause people to try to find other alternative, such as taking traditional medicine like “mengkudu” [Morinda citrifolia L.]. Mengkudu fruit had been proven to be able to increase the number of protein GLUT4 at type 2 diabetic male white rat’s sceletal muscle. Fruit of mengkudu has similar chemical ingredient with its leaves. So far, however, mengkudu leaves had not been studied to increase the number of protein GLUT4 at sceletal muscle. This study use male white rats Wistar strain. Total number of white rats involved in this study were 40 rats which treated : 10 rats as negative control group [no treatment], 10 rats as positive control group and they are treated with exogen insulin 0,35 U per body weight three times daily for 10 days. The last 20 rats is divided into 2 groups with 10 rat for each group as first trial group and second trial group. The trial group are treated with exogen insulin and first trial group were given water extract of mengkudu leaves 7,5 g per body weight and second trial group were given alcohol extract of mengkudu leaves 1,8 g per body weight twice daily which were administered starting from the seventh to tenth day of exogen insulin administration. After ten days of administration, blood glucose level of all rats was measured and all rats were killed afterwards. The next step is sceletal muscle tissue processing to be painted immunohistochemistry to GLUT4. Conclusion of the result is water extract and alcohol extract of mengkudu leaves  could not increase the number of protein GLUT4 at type 2 diabetic male white rats. ABSTRAK Kesulitan mendapatkan obat antidiabetes yang efektif dan aman menyebabkan pasien mencoba obat alternatif, misalnya dengan menggunakan obat tradisional “mengkudu” (Morinda citrifolia L.). Buah mengkudu telah terbukti dapat meningkatkan jumlah protein Glut4 pada otot rangka tikus putih jantan diabetik. Buah mengkudu memiliki kandungan kimia yang hampir sama dengan daunnya. Oleh sebab itu dalam penelitian ini dilakukan penelitian untuk mengetahui apakah daun mengkudu juga dapat meningkatkan jumlah protein Glut4 seperti buahnya. Dalam penelitian ini digunakan 40 tikus putih jantan strain Wistar yang dibagi menjadi 4 kelompok, yaitu kelompok kontrol negatif, kelompok kontrol positif yang diberi insulin eksogen 0,35 U per berat badan tiga kali sehari selama 10 hari, kelompok perlakuan pertama yang diberi ekstrak air daun mengkudu 7,5 g per kg berat badan, dan kelompok perlakuan kedua yang diberi ekstrak alkohol daun mengkudu 1,8 g per kg berat badan dua kali sehari mulai hari ke-7 sampai hari ke-10. Kadar gula darah diukur setelah hari ke-10. Jaringan otot rangka diambil dan dilakukan pewarnaan untuk melihat protein Glut4. Dari hasil penelitian yang diperoleh disimpulkan bahwa ekstrak air dan alkohol daun mengkudu tidak dapat meningkatkan jumlah protein Glut4 pada tikus putih jantan diabetik tipe 2.

Abstract

Canned sardines are hermetically sealed and processed to ensure sterility and provide practical convenience in processing fish. Contact between food and canned causes the migration of metal elements into them. The high content of metals, lead (Pb) and cadmium (Cd) in canned food has a negative impact on health. This study aims to determine both metal levels in canned sardines and to identify the fulfillment of several validation parameters against Atomic Absorption Spectrophotometry (AAS). Sardines were weighed, digested with concentrated HNO3:30% H2O2 and analyzed using AAS. Pb and Cd levels in canned sardines were not detected by AAS. This method meets the requirements of validation parameters, Pb and Cd, selective on specific wavelengths 283.3 nm and 228.3 nm, the standard curve is linear with a relative process standard deviation (Vx0) value < 5% and the Xp value is smaller than the lowest concentration in the standard curve, 0.69 mg/l and 2.31 mg/l are the results of the Limit of Detection (LOD) and Limit of Quantitation (LOQ) values of Pb while Cd 0.20 mg/l and 0.67 mg/l, 80-120% is %recovery of accurate parameters and a relative standard deviation (RSD) value < 2% is precision parameters. The SSA method can be used to quantitatively determine Pb and Cd levels in canned sardines.

Abstract

Amonia (NH3) adalah senyawa anorganik yang berasal dari senyawa nitrogen-hidrogen yang bersumber dari alam. Kadar amonia yang berlebihan mengakibatkan kematian pada organisme yang hidup dilingkungan tersebut berdasarkan hal tersebut penelitian ini bertujuan untuk mengetahuinilai keberterimaan, dan kadar amonia dalam air sungai yang berlokasi di daerah sekitar industri SIER menurut persyaratan peraturan menteri lingkungan hidup No.03 tahun 2010. Metode yang digunakan pada penelitian ini adalah metode fenat dengan menggunakan alat spektrofotometer visible. Hasil dari penelitian ini kandungan kadar amonia pada sampel air sungai di daerah industri SIER dengan rata-rata dari ketiga titik pengambilan sampel sebesar 2,24 mg/L, hasil tersebut menunjukan bahwa kadar amonia dalam air sungai di daerah SIER memenuhi persyaratan peraturan menteri lingkungan hidup No.03 tahun 2010 (standar baku mutu air limbah bagi kawasan industri) dengan kadar maksimum untuk amonia sebesar 20 mg/L.

Abstract

Apple vinegar is commonly made from fermented apple juice, which involves two consecutive stages of fermentation: alcoholic and acetic acid fermentation. Both Saccharomyces cerevisiae and Acetobacter acetii are involved in fermentation. Apple vinegar has various benefits, such as stabilizing blood pressure, treating rheumatism, and detoxifying toxins. Excessive consumption of this product may cause damage to the esophagus, low blood potassium levels or hypokalemia, diarrhoea, and ulcers due to the high acetic acid content. Moreover, the ethanol content also needs to be determined. It should be lower than 0,5% to fulfil the halal requirements for certification. The local brands of apple vinegar in Indonesia are generally produced by small-medium enterprises (SMEs), especially in Malang, Indonesia. Commercial brands of various apple vinegar, XCJO, YSW, THT, NTF, and BST, were sampled for their ethanol and acetic acid content in one method of analysis: gas chromatography, utilizing the high volatility properties of ethanol and acetic acid. The samples contain 17.73%, 11.45%, 6.43%, 2.67%, and 1.49% v/v of acetic acid, respectively, for XCJO, YSW, THT, NTF, and BST. At the same time, the ethanolic content is 0.11 and 0.02% v/v for XCJO and YSW, respectively, which are lower than the label value informed. Meanwhile, the rest of the brands' content is 4.00%, 2.39%, and 3.21% ethanol for THT, NTF, and BST, respectively. In addition, the acetic acid content of XCJO is high. A consumer should be aware of dilution before consuming the XCJO apple vinegar. It is unnecessary in the case of YSW, THT, NTF, and BST. Abstract

Wine is an alcoholic drink that results from the fermentation of grape juice with the help of microbes. One of the variants of wine circulating in Indonesia is wine Kolesom. Kolesom, also known as ginseng wine. Gingseng/kolesom/temulawak is classified as grapes wine with other food ingredients as an addition. It contains EtOH in between 7-24 %v/v according to the quality standard of Kolesom in The Regulation of the National Agency of Drug and Food Control of Republic Indonesia (BPOM) No. 5/2021. Kolesom extracts, considered nutritious for health, are added to grape juice, though it is Khmer and haram. In traditional herbal medicine, kolesom usually use as an addition, giving the body a strong taste, warm effect, and vitality. Adding kolesom in Jamu is a choice depending on the customer's favor. Kolesom wine is classified as B-class liquor, containing 5-20% v/v ethanol. Considering that most Indonesian citizens are Muslim, it is necessary to analyze and cross-check information to the customer about the level content of the alcohol in Javanese wine, kolesom. Ensuring the correct information to the customer and halal lifestyle. Gas chromatography equipped with a flame ionized detector (GC-FID) is proposed as a method for analyzing the ethanol (EtOH) content in kolosem, Javanese wine, as well as the traditional wine of Bali. The results showed that the proposed GC-FID method was well-validated.

Abstract

Introduction: The utilisation of traditional medicine to address health concerns is on the rise. One such traditional medicine product is antihypertensive herbal medicine. The Indonesian Food and Drug Authority (BPOM) has identified the presence of medicinal chemicals in traditional medicinal products, which has the potential to pose a significant risk to consumers. Captopril is one of the antihypertensive drugs that may be added as BKO in herbal medicine. Aims: The objective of this study was to develop a method for the analysis of the chemical content of captopril in antihypertensive herbs sold online under the following brands: A, B, C, and D. Method: The analytical method used was TLC-densitometry, utilising a mobile phase of chloroform: methanol (7:3). Result: The results show that the method used has fulfilled the requisite validation criteria for linearity, limit of detection (LOD), limit of quantitation (LOQ), accuracy, and precision parameters. The results of the analysis demonstrated that the sample did not contain any detectable quantities of captopril. Conclusion: The developed method has been proven to be effective for the analysis of captopril in antihypertensive herbal products and it can be confirmed that there is no presence of captopril medicinal chemicals in samples from brands A, B, C, and D
