Abstract

Lung cancer is genetic changes in the V-Ki-ras2 gene Kirsten rat sarcoma viral oncogene homolog(KRAS), which is involved in signaling pathways related to proliferation, cell differentiation, andapoptosis. Point mutations in the KRAS gene were detected in 15% to 20% of all Non-Small Cell LungCarcinoma (NSCLCs) and about 30% of lung adenocarcinomas, with the most common mutation beingat codon 12. This study aimed to determine the binding reaction between diterpene-type cembranoidcompounds from tobacco leaves (Nicotiana tabacum L.) and KRAS in human lung cancer. We foundthe presence of a cembranoid-type diterpene active compound in the form of thunbergol (C20H34O)with Gas Chromatography and Mass Spectroscopy (GC-MS) examination. Based on the resultsof molecular docking, it was found that the diterpene-type cembranoid ligand binds to the KRASreceptor with a yield of G -7.0 kcal/mol, pKi 7.35 M, one hydrogen bond with type ILE36 (1,937 Å).In conclusion, diterpene-type cembranoid can be considered an anticancer compound because of themolecular interaction with cembranoid-type diterpene through cell proliferation and differentiationpathways, as well as apoptosis.

Abstract

Abstrak: Diabetes mellitus (DM) merupakan penyakit dengan prevalensi tertinggi di dunia, tidak hanya menyerang para lansia namun pada usia yang lebih muda. Riwayat keluarga, aktivitas fisik, umur, stres, tekanan darah serta nilai kolesterol berhubungan dengan terjadinya DM tipe 2, dan orang yang memiliki berat badan dengan tingkat obesitas berisiko 7,14 kali terkena penyakit diabetes mellitus tipe 2 jika dibandingkan dengan orang yang berada pada berat badan ideal atau normal. Tujuan : Meningkatkan pengetahuan masyarakat tentang ciri-ciri pengidap penyakit Diabetes Mellitus dan faktor-faktor pencegahannya. Metode: pengabdian masyarakat dilakukan dengan cara edukasi online berbasis zoom meeting dengan menggunakan power point. Pre test dilakukan sebelum edukasi dimulai, post test dilakukan setelah pemaparan edukasi selesai dilakukan. Materi edukasi terdiri dari definisi, etiology, klasifikasi, faktor-faktor resiko, gejala klinis, pemeriksaan penunjang, diagnosis, terapi farmakologi dan non farmakoterapi serta cara pencegahannya Hasil: 50 audiens telah mendapatkan informed consent sebelum mengikuti kegiatan pengabdian masyarakat, 27 audiens mengikuti pre test dan 41 audiens mengikuti post test. Didapatkan peningkatan pengetahuan peserta mengenai definisi diabetes mellitus dari 74,1% di pre test dan sesudah post test 100%, Penyebab diabetes mellitus dari 48,1% pre test dan sesudah post test 92,7%, Kebiasaan yang menyebabkan diabetes mellitus dari 40,7% di pre test dan sesudah post test 87,8% , Pencegahan diabetes mellitus dari 55,6% di pre test dan sesudah post test 85,4 %, faktor pencegahan diabetes mellitus dari 81,5 % pre test dan sesudah post test 92,7 % Kesimpulan: terjadi peningkatan pengetahuan responden tentang penyakit diabetes mellitus

Abstract

Cervical cancer is the fourth most common cancer among women, with resistance to therapy often linked to Bcl-2 overexpression. This study evaluated the anticancer activity of durian peel extract (Durio zibethinus Murray) against HeLa cells through Bcl-2-mediated apoptosis and investigated apigenin and luteolin interactions with Bcl-2 via molecular docking. The extract was prepared by ethanol maceration, characterized by phytochemical screening and GC-MS, and tested for cytotoxicity using the MTT assay, while apoptosis was assessed through Bcl-2 immunofluorescence. Phytochemical screening confirmed flavonoids, alkaloids, saponins, tannins, and triterpenoids, with GC-MS identifying over 93 bioactive compounds. The extract exhibited significant cytotoxicity with an IC₅₀ of 5.92 μg/mL, and cell viability decreased concentration-dependently (p < 0.05), accompanied by apoptotic morphological changes and reduced Bcl-2 expression. Molecular docking revealed favorable binding of apigenin and luteolin to Bcl-2, with binding energies of −6.01 and −5.55 kcal/mol, respectively. In conclusion, durian peel extract demonstrates promising anticancer activity against HeLa cells by inducing apoptosis through Bcl-2 suppression, with apigenin and luteolin serving as potential natural candidates for cervical cancer therapy.
