Abstract

Hot springs or hot springs are springs that are produced as a result of the release of groundwater from the earth's crust after experiencing geothermal heating. Indonesia is one of the most active tectonic regions in the world with more than 70 active volcanoes, and has many geothermal areas. Hot springs are a suitable growth medium for thermophilic bacteria. Thermophilic bacteria are a group of bacteria that adapt to high-temperature environmental conditions, with temperatures ranging from 50°C-75°C. The natural habitat of thermophilic bacteria is widespread throughout the earth's surface, including in hot springs, volcanic craters or volcanic areas. One of the hot springs in North Sumatra province is sidebuk-debuk, which is located in Bandar Baru sub-district. is a hot spring, which has a temperature between 50°C – 75°C. At this time thermophilic bacteria are studied and researched intensively for reasons of developing basic research and biotechnology applications. Thermophilic bacteria have the potential to produce heat-resistant/ thermostable enzymes and can be used in industry, waste treatment processes and mineral weathering. Amylase is an important enzyme that is widely used in industrial applications through synthesis reactions and hydrolysis reactions, accounting for almost 65% of the total sales of enzymes in the world. Researchers are interested in knowing more about the role of thermostable enzymes in modern biotechnology today, therefore it is necessary to conduct research aimed at isolating and identifying local isolates of thermophilic bacteria as potential sources of enzymes, especially in North Sumatra. From this study, 19 isolates of Bacillus sp bacteria were obtained from Sidebuk-debuk hot springs based on phenotypic characters by looking at macroscopic, microscopic and biochemical characteristics, with 9 isolates of thermophilic bacteria that have the potential to produce amylase as indicated by the presence of a clear zone around the bacterial colonies. Keywords: Hot Springs, Bacillus sp Bacteria,

Abstract

Indonesia is a country that produces a lot of starchy materials such as cassava, sago, corn and others. Indonesia has the potential to develop an enzyme-producing industry, especially amylase. The utilization of starchy materials which so far has not been maximized and is still limited to conventional methods can be increased by enzymatic development processes. This will increase its economic value and at the same time increase state income. One of the enzymes that is currently very widely used in the food and beverage industry is amylase. Nin Suharti and Geminsyah Putra 2021 have succeeded in isolating and identifying thermophilic bacteria from the sidedust hot springs of North Sumatra. The advantage of thermophilic bacteria is that they can produce enzymes that are resistant to high temperatures / thermostable enzymes. These enzymes are able to survive and are active at high temperatures. Such properties are highly needed by enzyme-based industries. The use of enzymes that can withstand high temperatures in the field of biotechnology can reduce operating costs and increase reaction speed, this study uses various types of starch including potatoes, cassava and corn as suitable and easily available carbon sources for the production of amylase enzymes. Bacteria in the media with potato starch added can produce the highest amylase enzyme from the three treatments of 65 U/L. For bacteria added with corn starch, the result was 50 U/L of amylase enzyme. Meanwhile, the lowest of the three treatments was media with added sweet potato starch with a content of 39 U/L. The isolates generally had a round shape, jagged edges, grooved, and smooth, white colonies, microscopic bacilli-shaped, endospores and motile, positive for catalase, gelatin and citrate tests. With Gram staining, it generally shows a positive Gram reaction.

Abstract

First-time mothers may find childbirth difficult; they frequently feel intense anxiety, stress, and terror as the due date draws near, particularly in the third trimester of pregnancy when there is a higher chance of an early birth. The aim of the research was to ascertain the impact of Murottal therapy and husband support on the anxiety levels of expectant mothers during labor at Badrul Aini Mother and Child Hospital. This study uses a cross-sectional research approach and is quantitatively analytical. In August 2023, the study was carried out at the Badrul Ain Mother and Child Hospital. All of the pregnant patients in this study came to the Badrul Aini Mother and Child Hospital to give birth. The sampling technique was random, so the research sample was obtained from as many as 50 pregnant women. Data collection techniques in this study used questionnaires and observation techniques. Data analysis used correlation and multiple linear regression tests with SPSS software processing. The results showed that husband support (p-value = 0.006) and murrotal therapy significantly reduced the anxiety level of pregnant women (p-value = 0.001). However, the hypothesis that Murrotal therapy moderates the effect of husband support on anxiety levels was not supported. The importance of the husband's role in providing emotional and psychological support during labor and confirming the effectiveness of Murrotal therapy in managing maternal anxiety. Healthcare providers should consider involving husbands in prenatal care and labour, and Murrotal therapy can be a valuable tool in managing pregnant women's anxiety. Abstract

Indonesia is listed as the third largest shrimp producing country in the world. Shrimp is one of the leading export commodities. Shrimp is one of the main commodities in the fishing industry. Thus encouraging Indonesia to become one of the shrimp producing countries. In the shrimp processing process, usually the heads and shells of the shrimp are thrown away or used as animal feed. Traditionally, shrimp shell waste is generally used as a mixture for making crackers, paste, shrimp paste and as additional animal feed and returned to the shrimp cultivation itself. One use of shrimp shell waste that has prospects for development and has quite high economic value is processing it into chitin, chitosan and glucosamine. The aim of this research is to obtain isolates of chitinolytic bacteria from isolation and to utilize chitinolytic bacteria from white shrimp (Penaeus merguensis) shells to produce chitinase enzymes. Chitinolytic bacteria are bacteria that produce chitinase enzymes which play a role in degrading chitin into oligomers and chitin derivative compounds. Chitinolytic bacteria can be obtained from various sources such as the rhizosphere, phyllosphere, soil or from water environments such as seas, lakes, ponds or shrimp ponds and so on. Chitin that exists in nature is easily degraded by microorganisms. There are two pathways in the chitin degradation process. First, degradation by chitinolytic which hydrolyzes the ?-1,4-glycoside bond. Second, the polymer undergoes first deacetylation and then hydrolysis by chitosanase. Chitinase has many benefits, including as a bioinsecticide and biofunctional agent in controlling plant pests and is used to process chitin waste in the shrimp and crab freezing industry. It can also be used in the medical and food fields. From the test results, three types of bacteria were obtained, namely Vibrio sp, Bacillus sp, and Pseudomonas sp.

Abstract

Abstract

Limited access to and the high cost of commercial growth media have driven the development of alternative media based on locally available materials. Although several legume-based materials have been reported to support bacterial growth, the use of mung beans (Vigna radiata) as an alternative medium—particularly in the form of flour and infusion—and comparisons of their effectiveness against standard media remain limited. This study aims to evaluate the potential of these two forms as growth media for Staphylococcus aureus. This was an experimental study conducted from March to May 2024 at the Microbiology Laboratory of the Medan Public Health Polytechnic (Poltekkes Kemenkes). The method used was growth analysis via the Total Plate Count (TPC) method. The results showed that the average TPC of S. aureus on mung bean flour medium was 1.07 × 10⁷ CFU/mL, and on mung bean infusion medium it was 4.7 × 10⁶ CFU/mL, whereas TSA medium, as the standard medium, yielded 5.8Z× 10⁶ CFU/mL. This indicates that both forms of the mung bean alternative media were capable of supporting optimal bacterial growth, with the flour medium yielding a higher cell count compared to the standard medium (TSA). Morphologically, the colonies on the alternative media were smaller (±2 mm, yellowish-white) compared to those on TSA (±3 mm, yellow). Thus, it can be concluded that mung bean flour has the potential to be an effective alternative medium for growing S. aureus compared to the infusion medium, which is less optimal due to the possible degradation of nutrients during the heating process.

Abstract

Chronic kidney disease (CKD) is a disease characterized by a progressive decline in kidney function, often accompanied by anemia due to low hemoglobin (Hb) levels. Hemodialysis is a therapy used to replace kidney function and has the potential to affect Hb levels. This study aims to determine changes in hemoglobin levels before and after hemodialysis in CKD patients. The study was conducted at H. Abdul Manan Simatupang Kisaran Regional Hospital with a quantitative design to observe hemoglobin levels before and after hemodialysis. A total of 35 CKD patients undergoing hemodialysis were taken as samples using a purposive sampling technique with certain inclusion criteria. Hb level data were collected from medical records and analyzed using a paired t-test. The results showed that Hb levels before hemodialysis had an average of 8.217 g/dL and increased to 8.831 g/dL after hemodialysis. The statistical test showed a p value = 0.000 (p < 0.05), which means there was a significant difference between Hb levels before and after hemodialysis. The conclusion of this study is that hemodialysis has a significant effect on increasing hemoglobin levels in patients with chronic kidney failure.
