Abstract

Penelitian ini dilakukan untuk mengetahui kecepatan timbulnya berahi dari dosis Gonadotropin Releasing Hormone (GnRH) yang berbeda dan pengaruh waktu pemberian GnRH terhadap jumlah korpus luteum pada sapi Pesisir. Materi yang digunakan yaitu 12 ekor induk sapi Pesisir, hormon PGF2α dan GnRH. Terdiri dari dua tahap, pertama Induk sapi disinkronisasi dengan 5 ml PGF2α dan kemudian diberi GnRH 50 µg, 100 µg dan 200µg. Kedua sapi disinkronisasi dengan 5 ml PGF2α dan kemudian diberi GnRH dosis hasil penelitian tahap pertama dan dibagi atas 4 waktu penyuntikan, yaitu A(16), B(32), C(48), dan D(64) jam. Peubah yang diamati adalah waktu terjadinya estrus setelah diberi dosis yang berbeda dan jumlah korpus luteum (CL) yang terbentuk dari waktu penyuntikan yang berbeda. Pengolahan data dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL). Hasil penelitian menunjukkan bahwa dosis GnRH yang tercepat menunjukan gejala estrus adalah 100 µg dengan rataan 45.42±2.24 jam, 200 µg rataan 51.33±4.51 jam dan 50 µg rataan 76.53±11.92 jam. Waktu pemberian GnRH berpengaruh terhadap jumlah CL yang terlihat dari jumlah CL dari masing-masing perlakuan. Perlakuan C(48) jam menunjukkan jumlah CL yang paling banyak dengan rataan 4.33±1.15 dan diikutiB(32) jam dengan rataan 3.67±1.53,perlakuan A(16) jam dengan rataan1.67±1.15perlakuan D(64) dengan rataan 1.67±0.58.

Abstract

This study is proposed to determine the heritability of growth traits of Simmental cattle in BPTU-HPT Padang Mengatas, West Sumatra. Data were collected by observations and measurements of the cattle in the field, completed by the secondary data from BPTU-HPT recorded from 2010 to 2016. Data for heritability were collected from 455 offsprings belonged to 15 sires. Weight and body size are two parameters used to estimate the heritability and breeding value, the heritability was analyzed between weaning and yearling age and breeding value was only calculated for weaning age. The data were analyzed by using paternal half-sib correlation method. The heritability between bulls and offspring was varied in weaning age, ranged from 0.24 to 0.97, and yearling age was showed high heritability in all parameters, ranged from 0.56 to 0.64. The heritability of weaning weight was 0.50 ± 0.19, weaning body size (shoulder height, body length and heart girth) was 0.97 ± 0.31; 0.24 ± 0.13; 0.44 ± 0.19, yearling weight was 0.56 ± 0.08 and yearling body size was 0.58 ± 0.08; 0.64 ± 0.09 and 0.56 ± 0.08 respectively. This result found that the highest heritability was shoulder height and the lowest heritability was body length. Relative breeding value above 0 represented by six sires with the highest breeding value was 37.12. Sires with the highest BV can be selected for future breeding in BPTU-HPT Padang Mengatas, while the lowest BV is recommended to be culled.

Abstract

This research aim to know the influence of depth veneering time of spermatozoa at media of TALP which was supplemented by 4% BSA to embryo gender in in- vitro and to get the time dissociation of spermatozoa which have the best chromosome Y. This research represent attempt of exsperiment in RBT by 4 time veneering of spermatozoa as treatment, times intake of semen of epididymis to groups. Variables measure are: 1). mortality of spermatozoa, 2). number of in vitro fertilzation, 3. masculine gender embryo ratio. Result shown, the veneering of cow spermatozoa during 90 minute in media of TALP which was supplemented by 4% bovine serum albumin (BSA) can dissociate spermatozoa which have chromosome Y with percentage of highest male sex (90.00±24.49), and highest percentage of mortality and fertility were obtained at veneering during 60 minute with scoring 73.33 ± 5.16 and 53.92±18.79 respectively.

Abstract

ABSTRAK. Studi ini bertujuan untuk mengidentifikasi keragaman gen FSH bagian ekson 2 menggunakan enzim restriksi TasI pada sapi Pesisir. Sampel darah yang digunakan yaitu 78 sampel darah dengan dua macam yaitu 13 jantan serta 65 betina yang berumur 1-5 tahun dari sapi pesisir yang didapat dari BPTUHPT Padang Mengatas. Protocol genomic DNA Purification system Kit dari Promega digunakan untuk mengisolasi DNA sampel darah. Amplifikasi gen FSH dilakukan dengan menggunakan teknik PCR atau Polymerase Chain Reaction. Enzim TasI digunakan untuk meretriksi produk amplifikasi. Hasil penelitian gen FSH bagian exon 2 pada sapi pesisir yaitu diperoleh terdapat dua macam genotip. Dua macam genotip tersebut yaitu heterozigot (+/-) serta homozigot tidak terpotong (-/-), untuk jantan berturut turut yaitu 0,38 dan 0,62 beserta frekuensi alel (+) dan alel (-) yaitu 0,19 dan 0,81, sedangkan untuk betina yaitu berturut-turut 0,43 dan 0,57 beserta frekuensi alel (+) dan alel (-) yaitu 0,21 dan 0,79. Sebaran genotip gen FSH|TasI exon 2 pada populasi Sapi Pesisir sifatnya polimorfik (beragam) dengan frekuensi alel yaitu kurang dari 0,99.Identification of Exon 2 FSH Gene Diversity Using TasI Restriction Enzymes in Coastal CattleABSTRACT. This study aims to identify the diversity of the exon 2 FSH gene using the restriction enzyme TasI in Coastal cattle. The blood samples used were 78 blood samples of two types, namely 13 males and 65 females aged 1-5 years, from coastal cattle obtained from BPTUHPT Padang Atas. The protocol genomic DNA Purification System Kit from Promega was used to isolate DNA from blood samples. The amplification of the FSH gene was carried out using the PCR or Polymerase Chain Reaction technique. The TasI enzyme was used to restrict the amplification product. The study results on the exon two portion of the FSH gene in coastal cattle showed two types of genotypes. The two types of genotypes are heterozygous (+/-) and untruncated homozygous (-/-); for males, 0.38 and 0.62, respectively, and the frequencies of allele (+) and allele (-) are 0.19 and 0. .81, while for females, it was 0.43 and 0.57, respectively, and the frequencies (+) and allele (-) were 0.21 and 0.79. Abstract

ABSTRAK. Penelitian ini bertujuan untuk menentukan kualitas oosit sapi Pesisir yang akan diproduksi secara in vitro dan untuk menentukan dosis penambahan Insulin pada media maturasi in vitro efektif untuk meningkatkan kematangan oosit sapi Pesisir. Sampel yang digunakan yaitu ovarium sapi Pesisir yang diambil di RPH kemudian dilakukan koleksi oosit dengan metode slicing di Laboratorium Bioteknologi. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) yang terdiri dari 4 perlakuan dan 4 ulangan. Perlakuan yang digunakan adalah penambahan insulin pada media maturasi in vitro yang terdiri dari tanpa insulin (P1), insulin 10 µg/ml (P2), insulin 15 µg/ml (P3) dan insulin 20 µg/ml (P4). Oosit sapi Pesisir dimaturasi dalam media maturasi selama 24 jam di dalam inkubator CO2 dengan suhu 38,5 ᵒC. Sampel yang sudah dimaturasi diwarnai dengan 2% aceto-orcein selama 5 menit kemudian zat pewarna dihilangkan dengan campuran (asam asetat, gliserin dan aquadest) selanjutnya pengamatan inti diamati dengan menggunakan mikroskop. Hasil penelitian menunjukkan bahwa oosit yang paling banyak ditemukan adalah oosit grade B (75,59%), A(13,53%), C (8,48%), dan yang paling sedikit adalah grade D (2,40%). Penambahan Insulin dengan dosis 10 µg/ml dan 15 µg/ml pada media maturasi in vitro dapat meningkatkan pematangan oosit sapi pesisir yang mencapai metaphase II (MII) daripada tanpa insulin, sedangkan penambahan dosis insulin 20 µg/ml menurunkan pematangan oosit sapi pesisir yang mencapai MII. Berdasarkan hasil penelitian tersebut disimpulkan bahwa oosit yang dimaturasi dengan penambahan insulin 10 µg/ml dan 15 µg/ml sangat berpengaruh (P<0,01) terhadap pematangan oosit mencapai MII dan hasil terbaik adalah penambahan insulin 15 µg/ml sebesar 84,58%. Kata kunci: Insulin, Oosit Sapi Pesisir, Pematangan Inti.Effect of Insulin Addition on In Vitro Maturation Media on Pesisir Cattle Oocyte MaturationABSTRACT. This study aims to determine the quality of Pesisir cattle oocytes to be produced in vitro and to determine that adding different insulin to in vitro maturation media effectively increases the maturity of pesisir cattle oocytes. The sample was bovine ovaries of Pesisir cattle taken at the RPH and then bovine oocytes collected at the Biotechnology Laboratory. This study used a randomized block design (RBD) which consisted of 4 treatments and 4 replications. The treatment used was adding of insulin to in vitro maturation media consisting of no insulin (P1), insulin 10 µg/ml (P2), insulin 15 µg/ml (P3) and insulin 20 µg/ml (P4). Pesisir bovine oocytes were matured in maturation medium for 24 hours in a CO2 incubator with a temperature of 38.5 ᵒC. Samples that had been matured were stained with 2% aceto-orcein for 5 minutes and then the dye was removed with a mixture (acetic acid, glycerin and distilled water) then the observations of the nucleus were observed using a microscope. The results showed that the most common oocytes were grade B oocytes (75.59%), A (13.53%), C (8.48%), and the fewest were grade D (2.40%). The addition of insulin at a dose of 10 µg/ml and 15 µg/ml in in vitro maturation media could increase the oocyte maturation of coastal bovines that reached metaphase II (MII) than without insulin, while the addition of 20 µg/ml insulin decreased the oocyte maturation of coastal bovines that reached MII. Based on the results of this study it was concluded that oocytes which were matured with the addition of insulin 10 µg/ml and 15 µg/ml had a very significant effect (P<0.01) on oocyte maturation reaching MII and the best treatmentin this study was an insulin dose of 15 µg/ml of 84.58%.

Abstract

ABSTRACT The purpose of this study was to identify the relationship between inseminator characteristics (internal and external) to the success of AI in Sawahlunto City. The objects of this research observation are all inseminators in Sawahlunto City. The data were analyzed with the simultaneous F test to see the effect of the variables simultaneously and the Partial T Test to see the effect of each variable. The results of the study found that the success of artificial insemination in Sawahlunto City was not very good, where an average S/C value of 3.04 was found. Internal characteristics (years of service, thoroughness, responsibility, technical ability in straw and thawing management, ability to understand the anatomy of the reproductive organs) and external (distance from home to work area, completeness of facilities and infrastructure, sanitation of equipment, communication of livestock reporting) have a significant relationship (P<0.05) on AI success, while internal (experience, training intensity) and external characteristics (IB postal conditions) did not have a significant relationship (P>0.05) to AI success in Sawahlunto City. From the results of this study it can be concluded that the characteristics of the inseminator (external and internal) influence the success of AI in Sawahlunto City. Abstract

The purpose of this study was to find out how the semen quality of pesisir cattle was after the addition of trehalose to egg yolk tris diluent. The research material in the form of fresh semen was obtained from pesisir cattle aged approximately 4 years and the quality of the semen had been tested by observing the quality of fresh semen before dilution.. This study used a randomized block design with 4 treatments and 4 replications. The treatment given was the addition of trehalose in egg yolk tris diluent. The treatment given consisted of control, the addition of trehalose 0.5%, 1%, and 1.5%. The variables in this study included motility, viability, intact plasma membrane (MPU), and intact acrosome cap (TAU) of spermatozoa of pesisir cattle. The results of this study indicated that the addition of trehalose had a significant effect (p<0.05) on the viability, MPU, TAU, and did not show a significant effect (p>0.05) on the motility of spermatozoa of pesisir cattle. The results of this study indicated that the dose of trehalose addition of 1.5% in egg yolk tris diluent showed the best results on the quality of spermatozoa of pesisir cattle, where motility was found to be 53%, viability 61%, MPU 63%, and TAU 63%. Abstract

This study aimed to find  the best combination between pre-freezing method and layer of post sexing on semen quality of Pesisir Bulls. This study used 2 Pesisir Bulls. The semen was evaluated using Computer Assisted Sperm Analyzer (CASA). This study used factorial randomized block design with 2 factors. Factor A was sexed semen layer of BSA column with 2 level, whereas Factor B was pre-freezing method with 3 level. Analysis of Variance (ANOVA) were used in this study for data Analysis. According to the analysis of variance, there is no interaction of the combination of two factors on each paramters. Pre-freezing method showed highly statistically significant effect (P<0.01) on the quality of motility on upper layer and bottom layer which was about (52.86-72.93%; 52.32-65.50%) and about (40.17-56.99%; 40.33-53.18%) on progressive motility. The value of Distance Curve Line on  (39.15-43.24 μm; 49.25-53.86 μm), Distance Average Path (22.38-24.60 μm; 26.52-29.94 μm), Distance Straight Line (17.32-19.07 μm; 20.93-24.25 μm). The value of Velocity Curve Line (88.67- 97.50 μm/s; 109.85-117.90 μm/s), Velocity Average Path (50.98- 55.63 μm/s; 59.32- 64.69μm/s), Velocity Straight Line (34.45-42.37 μm/s; 46.80-50.62 μm/s), Beat Cross Frequancy (23.91-25.17 Hz; 25.63-27.35 Hz), Straightness (0.75-0.77; 0.77-0.80), and Wobble (0.56-0.57; 0.53-0.55), highly statistically significant (P<0.01) on post sexing layers. Meanwhile, the value of Amplitude of Lateral Head (4.24-4.94 μm; 4.25-4.44 μm) showed highly significant effect (P<0.01) on factor A and factor B treatment, and value Linearity was about (0.42-0.44; 0.43-0.44) showed not significant effect (P>0,05) on every factor. According to the study, it can be concluded that the best treatment was treatment modified procedure layer by space the straw 16 cm every layers and The quality of post sexing X- and Y- spermatozoa motility effect on motility characteristic such as DCL, DAP, DSL, VCL, VAP, VSL, STR, WOB, ALH and BCF.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengidentifikasi karakteristik sifat kualitatif dan kuantitatif sapi silangan sapi Pesisir dan sapi Friesian Holstein (FH). Penelitian menggunakan 60 ekor sapi silangan sapi Pesisir dan sapi Friesian Holstein (FH). Metode Penelitian yang digunakan adalah metode berupa pengamatan dan pengukuran langsung dilapangan. Sifat kualitatif yang diamati adalah warna bulu, warna moncong, warna lingkar mata, tanduk dan gelambir. Hasil penelitian antara lain bahwa sifat kualitatif hasil persilangan dengan 3 variasi warna bulu induk, indukan berwan merah bulu merah menghasilkan bulu berwarna paling dominan coklat kemerahan hitam (40,00%), Warna moncong dominan hitam (91,11%). Lingkar mata berwarna dominan hitam (88,89), Muncul tanduk (100,00%) dan bergelamir (100,00%). Indukan berwarna hitam menghasilkan bulu berwarna dominan hitam putih (90,00%), Warna moncong berwarna dominan hitam (91,11%). Warma lingkar mata hitam (100,00%). Muncul tanduk (100,00%) dan bergelambir (100,00%). Indukan berwarna Putih menghasilkan warna bulu dominan berwarna hitam putih (80,00%), Warna moncong hitam (100,00%), warna lingkar mata hitam (100,00%). Muncul tanduk (100,00%) dan bergelambir (100,00%). Ukuran tubuh betina umur 0,3,6 bulan berturut-turut : Panjang Badan (56,49 cm ± 2,65, 76,11 cm ± 4,16, 92,22 cm ± 2,38), Tinggi Pundak (55,76 cm ± 3,90, 75,00 cm ± 3,95, 89,89 cm ± 2,62), Lingkar dada (69,14 cm ± 2,12, 90,32 cm ± 3,67, 100,41 cm ± 1,62), Panjang Kepala (20,41 cm ± 1,36, 22,03 cm ± 1,61, 25,35 cm ± 2,26), Lebar kepala (10,51 cm ± 1,04, 12,32 cm ± 0,97, 13,57 cm ± 1,09), Bobot badan (24,90 cm ± 1,43, 57,38 cm ± 5,69, 85,79 cm ± 3,61). Ukuran tubuh Jantan lebih tinggi dari pada betina.   Kata kunci: Kualitatif, Kuantitatif, Sapi Friesien Holstein (FH) dan Sapi Pesisir
