Abstract

Penurunan produksi anggrek di Indonesia disebabkan oleh penyediaan bibit yang rendah di tingkat petani maupun industri. Teknologi perbanyakan secara konvensional melalui kultur in vitro menggunakan benih belum mampu memenuhi permintaan bibit anggrek dalam jumlah yang besar. Teknologi perbanyakan bibit anggrek secara klonal menggunakan bioreactor dapat menghasilkan bibit klonal dalam waktu yang cepat karena meningkatkan kecepatan proliferasi sel. Pengenalan teknologi bioreactor kepada petani plasma dan industri anggrek penting dilakukan untuk mendapatkan bibit anggrek dengan kualitas ekspor. Bimbingan teknis diikuti oleh petani plasma dan karyawan DD Orchid Nursery. Kegiatan bimtek dilakukan dengan metode ceramah, diskusi, dan demonstrasi tentang teknik perbanyakan bibit anggrek secara in vitro serta pengenalan teknologi bioreactor menggunakan organ vegetatif tanaman untuk produksi masal anggrek. Hasil kegiatan bimtek menunjukkan bahwa petani plasma dan karyawan DD Orchid Nursery sangat antusias pada saat demonstrasi bioreactor serta saat berdiskusi tentang penggunaan bioreactor dari skala kecil hingga besar. Aplikasi bioreactor menjadi salah satu solusi dalam memproduksi bibit anggrek secara masal dalam waktu yang singkat.

Abstract

Vanilla (Vanilla planifolia Andrews.) is one of the plantation crops that has a high selling price, namely wet vanilla IDR 200,000-300,000/kg, regular quality dry vanilla around 1-3 million/kg, and export quality reaching 5-7 million/kg. The opportunity for farmers to develop vanilla commodities in Indonesia is very large, but limited planting materials still hamper it. In-vitro propagation is one technique that can be used to overcome the problem of vanilla propagation in Indonesia. This research aimed to determine the interaction of BAP and coconut water on the growth of vanilla nodal explants in vitro. The method used in this research was a factorial Completely Randomized Design (CRD) with a combination of Benzyl Amino Purine (BAP) and coconut water. The BAP concentration consists of 0 mg/L, 1 mg/L, and 2 mg/L. Coconut water has 0%, 15%, and 30% levels. Data were analyzed using ANOVA and further tested with the DMRT test at a confidence level of 95%. The results showed an interaction between BAP and coconut water on the growth of vanilla explants. The combination treatment of 1 mg/L BAP + 15% coconut water showed the best results with the fastest response time of 8.76+1.53 DAP, average shoot length 2.84±0.70 cm/explant and average root length 0.75±0.07 cm/explant

Abstract

The Dendrobium variety orchid is a plant that has rare potential and growth. This requires large amounts of seeds that can be propagated in vitro. This study aims to determine the effect of TDZ on the direct induction of somatic embryogenesis. The study was conducted using a completely randomized design (CRD) with one factor, namely media consisting of basal Murashige and Skoog (MS) media with growth regulator TDZ. Each treatment out of a total of seven treatments was repeated four times. The research was carried out from March 2022 to August 2022. The results showed that leaf explants (clonal) could only be induced in 2 treatments (1 mg/L and 1.5 mg/L) out of 28 experimental media treatment units. The earliest callus appeared in the 1 mg/L treatment with a growth time of 90 days after planting (DAP) and the earliest PLB appeared in the 1.5 mg/L treatment with a growing time of 45 days after planting (DAP), the percentage of explant growth that became PLB with 1.5 mg/L treatment resulted in a percentage of 20 % and the percentage of callus growth at 1 mg/L yields a percentage of 20%.
