Abstract

Cellulolytic bacteria are bacteria that produce cellulase enzymes capable of degrading cellulose substrates. This study aimed to isolate and identify as well as measure the cellulase enzyme activity of cellulolytic bacteria isolated from the digestive tract of swans (Cygnus olor) in Mataram. The bacteria were isolated using de Man Rogosa Sharpe (MRS) selective media with the spread plate method. Then, the extracellular enzyme activity test was conducted by growing selected pure isolates on 1% Carboxy Methyl Cellulose (CMC 1%) media, followed by pouring 0.1% congo red solution and rinsing with 1M NaCl solution to determine cellulolytic activity (cellulolytic potential indicated by the appearance of a clear zone around the colony) and measuring the cellulolytic index. The bacterial isolation results obtained 4 isolates with the potential to support fiber degradation in animal feed. The highest cellulolytic index was produced by the isolate coded S1, reaching 34.5 mm, while the lowest cellulolytic index was produced by 2 isolates coded J21 and I2, reaching 18.8 mm.

Abstract

Diseases in goats reduce productivity and may cause reproductive disorders and zoonotic risks. Salmonellosis, caused by Salmonella spp., is an important livestock disease that can infect ruminants and poultry and contaminate animal products, affecting animal and human health. The purpose of this study was to determine the presence and the prevalence of Salmonella in Peranakan Etawa (PE) goats and to evaluate the antibiotic resistance profile of the isolates. Fecal samples were collected from 20 Peranakan Etawa goats and examined for the presence of Salmonella. Data on Salmonella cases and inhibition zone diameters (mm) obtained from antibiotic susceptibility testing were analyzed descriptively. The goats were managed under a semi-intensive system. They were fed odot grass supplemented with concentrate and silage additives, provided with well water as a drinking source, with feed troughs cleaned once daily and manure removed weekly. No anthelmintic treatment had been administered during the previous eight months. The results indicated that Salmonella infection was detected in 5% of the PE goats. The isolates showed the highest sensitivity to ciprofloxacin (25 mm inhibition zone), followed by chloramphenicol (8 mm) and tetracycline (7.33 mm), while resistance was observed against ampicillin.

Abstract

EIxceissivei eithanol consumption is associateid with impaireid malei reiproductivei function through its deitrimeintal eiffeicts on teisticular structurei and speirmatogeineisis. Howeiveir, thei meichanisms undeirlying eithanol-induceid teisticular damagei, particularly thosei involving Leiydig ceills, Seirtoli ceills, and androgein reiceiptor (AR) eixpreission, reimain incompleiteily undeirstood. This study aimeid to eivaluatei thei eiffeicts of eithanol administration on speirmatogeineisis and teisticular histopathological damagei in Wistar rats by asseissing Leiydig ceills, Seirtoli ceills, and AR eixpreission. A post-teist-only control group deisign was conducteid using 32 malei Wistar rats randomly assigneid to onei control group and threiei eithanol-treiateid groups reiceiiving diffeireint doseis of eithanol for 28 days. Teisticular tissueis weirei eixamineid histologically to asseiss Leiydig and Seirtoli ceills, whilei AR eixpreission was meiasureid using an einzymei-linkeid immunosorbeint assay (EILISA). Data weirei analyzeid using onei-way analysis of variancei (ANOVA) followeid by Tukeiy's post hoc teist, with statistical significancei seit at p < 0.05. EIthanol administration significantly reiduceid thei numbeirs of Leiydig and Seirtoli ceills in a dosei-deipeindeint manneir (p < 0.05). Histopathological eixamination reiveialeid progreissivei teisticular damagei characteirizeid by geirm ceill deigeineiration, reiduceid ceillular deinsity, cytoplasmic vacuolization, and narrowing of thei seiminifeirous tubulei lumein. Although AR eixpreission showeid a deicreiasing treind, no significant diffeireinceis weirei obseirveid among thei groups (p > 0.05). EIthanol eixposurei compromiseis teisticular inteigrity by disrupting speirmatogeineisis through thei deipleition of Leiydig and Seirtoli ceills and inducing histopathological alteirations without significantly affeicting AR eixpreission. Theisei findings providei insight into thei meichanisms of eithanol-induceid malei reiproductivei toxicity and may support futurei strateigieis for preiveinting alcohol-reilateid malei infeirtility.
