Abstract

Inflammatory cells infiltration after eugenol administration in Sprague Dawley rat’s molars. The purpose of this study was to determine effect of eugenol on inflammatory cells infiltration (neutrophils, macrophages and lymphocytes) in the inflamed dental pulp. Thirty male Sprague Dawley rats aged 3-4 months and weighed 300-350 g were randomly divided into 2 groups: treated and control groups. Rats were injected with ketamine HCl i.m. (0.1 ml/100 grams BW) before cavity preparation at occlusal surface of the upper molars. Cavity preparation was made using a round bur No.010 to pulp perforation. At the cavity base of treated group, 15 rats were given eugenol and of control group 15 rats were given aquadest, before filled with temporary filling. Three rats from each group were sacrificed at 1, 3, 5, 7 and 14 days after the cavity preparation. Rats were injected with ketamine HCl im (0.1 ml / 100 g BW) on the thigh then tissues were collected for histological process and stained using haematoxylin eosin. The number of inflammatory cells (neutrophils, macrophages and lymphocytes) was analysed by Two-Way Anova. The result of the study showed number of inflammatory cells infiltration of treated group was lower than control and showed significant differences between groups (p <0.05). The conclusion from this study is eugenol administration in Sprague Dawley rats’s inflamed dental pulp can reduce the number of inflammatory cells (neutrophils, macrophages and lymphocytes).ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh eugenol terhadap jumlah sel inflamasi (netrofil, makrofag dan limfosit) pada pulpa gigi terinflamasi. Tiga puluh ekor tikus Sprague Dawley jantan berumur  3-4 bulan dengan berat badan 300-350 g dibagi secara acak dalam 2 kelompok yaitu perlakuan dan kontrol. Tikus diinjeksi ketamine HCl i.m. (0,1 ml/100 gram BB) sebelum dilakukan preparasi kavitas pada permukaan oklusal gigi molar satu rahang atas. Preparasi kavitas dibuat dengan menggunakan bur bulat No.010 hingga kedalaman pulpa. Pada dasar kavitas kelompok perlakuan (15 ekor) diberi eugenol dan kelompok kontrol (15 ekor) diberi akuades, kemudian kavitas ditumpat sementara. Tiga ekor tikus dari masing-masing kelompok dikorbankan pada 1, 3, 5, 7 dan 14 hari pasca preparasi kavitas. Tikus diinjeksi ketamine HCl i.m. (0,1 ml/100 gram BB) pada paha kemudian jaringan diambil, diproses secara histologis dan dicat dengan hematoxylin eosin. Data jumlah sel inflamasi (netrofil, makrofag dan limfosit) dianalisis dengan Two Way Anova. Hasil penelitian menunjukkan rerata jumlah infiltrasi sel inflamasi kelompok perlakuan lebih rendah dibandingkan kontrol dan berbeda bermakna (p<0,05). Kesimpulan dari penelitian ini adalah pemberian eugenol pada pulpa gigi tikus Sprague Dawley yang mengalami inflamasi dapat menurunkan jumlah sel inflamasi (netrofil, makrofag dan limfosit).

Abstract

A good sealing ability of apical part of root canal system is needed to prevent microleakage thus resulting long-lasting successful treatment. Chitosan nanoparticles as nanofillers could be added to epoxy resin sealer to increase adaptation to dentinal wall. The purpose of this study was to evaluate the apical sealing ability of chitosan nanoparticles addition to epoxy-resin-based sealer. Thirty mandibular premolars were used in this study. The root canals were prepared using rotary files up to #30/0.09. The canal was irrigated with 2.5% NaOCl and 17% EDTA, then was rinsed with sterile water. All teeth were randomly divided into 3 groups (n=10). Group I was obturated with gutta-percha, Group II was obturated with gutta-percha and epoxy resin sealer, group III was obturated with gutta-percha and epoxy resin sealer that was added with chitosan nanoparticles. All specimens were stored in an incubator for 7 days at 37 °C. After that, specimens were tested with centrifuging dye penetration method with methylene blue 2% solution. The specimens were longitudinally sectioned, observed under stereomicroscope (8x magnification) and measured in millimeters (mm). The data were analyzed using one-way Anova. The results of one-way Anova showed that dye penetration in apical part of the root canal in all groups was statistically significant difference. The addition of chitosan nanoparticles to epoxy resin sealer increases the apical sealing ability of root canal obturation material.

Abstract

Bleaching merupakan prosedur pemutihan gigi yang pada saat ini banyak diminati oleh masyarakat. Salah satu efek bleaching adalah terjadinya peningkatan porusitas email sehingga gigi lebih rentan terhadap terjadinya karies. Remineralisasi diperlukan untuk meningkatkan kekerasan email, salah satunya dengan pemberian pasta gigi. Terdapat beberapa jenis pasta gigi yang beredar di pasaran, namun belum diketahui pasta gigi manakah yang terbaikuntuk digunakan pada pasien pasca bleaching. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan pasta gigi fluorida dan pasta gigi low-abrasive fluoridated terhadap karakteristik permukaan gigi pasca aplikasi home bleaching menggunakan karbamid peroksida 20%. Penelitian ini menggunakan spesimen berupa 20 gigi premolar rahang atas yang sudah dilakukan ekstraksi, kemudian diberi perlakuan penyikatan dengan akuades, pasta giginonfluorida, pasta gigi fluorida, dan pasta gigi low-abrasive fluoridated. Uji kekasaran permukaan dilakukan sebelum bleaching, setelah bleaching, serta penyikatan setelah bleaching. Hasil uji ANAVA dua jalur menunjukkan nilai p < 0,05 maka terdapat perbedaan yang signifikan pada kelompok perlakuan penyikatan gigi dan kelompok waktu pengujian kekasaran permukaan. Uji post-hoc multiple comparison test LSD menghasilkan terdapat perbedaaan kekasaran permukaan gigi yang bermakna antara kelompok penyikatan gigi dengan pasta gigi fluorida dan pastagigi low-abrasive fluoridated. Proses bleaching pada penelitian ini menunjukkan peningkatan kekasaran permukaan email gigi secara signifikan. Perlakuan penyikatan gigi menurunkan kekasaran permukaan email secara bermakna. Berdasarkan hasil penelitian dapat diambil kesimpulan bahwa proses penyikatan dengan pasta gigi pasca bleaching dapat mengembalikan kondisi kekasaran permukaan email yang mendekati saat sebelum dilakukan bleaching.Penggunaan pasta gigi fluorida dan pasta gigi low-abrasive fluoridated direkomendasikan untuk digunakan pada pasien pasca perawatan bleaching.

Abstract

ABSTRACTIntroduction: Root canal treatment (RCT) of mandibular third molars is often considered challenging due to anatomical variability, limited accessibility, and difficulty achieving proper isolation. As a result, extraction is frequently chosen. This case report outlines a successful single-visit RCT on a right mandibular third molar with a curved canal and Class II disto-occlusal caries. Case Report: A 30-year-old female presented with a large cavity on her right lower molar and requested a restorative treatment. Three weeks earlier, devitalization and temporary restoration had been performed by another dentist. Clinical examination revealed no periapical lesion, and percussion testing was negative. Radiographic analysis showed a slight curvature in the distal canal. A pre-curved K-File and the One Curve System were used for instrumentation. The canal was obturated in a single visit, followed by restoration using direct composite and short fiber-reinforced composite. Matrix and PTFE tape were utilized for proper adaptation in the distal area. Conclusion: With accurate diagnosis, understanding of anatomical complexity, and appropriate technique, single-visit RCT on mandibular third molars is achievable. This case supports the viability of conservative treatment for necrotic third molars with asymptomatic apical periodontitis, avoiding unnecessary extraction.Keywords single visit, root canal treatment, third lower molar, curved canal, composite restoration. ABSTRAKPendahuluan: Perawatan saluran akar (RCT) pada molar ketiga mandibula menantang karena variasi konfigurasi saluran, kesulitan akses ke area posterior, dan tantangan dalam isolasi. Akibatnya, pencabutan sering menjadi pilihan. Laporan kasus ini mendeskripsikan protokol RCT kunjungan tunggal pada molar ketiga kanan mandibula dengan saluran melengkung dan karies disto-oklusal kelas II. Laporan Kasus: Seorang wanita 30 tahun datang dengan kavitas besar pada molar ketiga kanan bawah dan menginginkan perawatan tanpa pencabutan. Tiga minggu sebelumnya, dokter lain telah melakukan devitalisasi dan restorasi sementara. Pemeriksaan klinis menunjukkan tidak ada lesi periapikal dan uji perkusi negatif. Radiograf menunjukkan kelengkungan ringan pada saluran distal. RCT dilakukan menggunakan K-file pra-lengkung dengan sistem One Curve. Obturasi dilakukan dalam satu kunjungan, diikuti restorasi komposit langsung yang diperkuat dengan komposit serat pendek. Area distal direstorasi menggunakan matriks dan pita PTFE. Simpulan: RCT kunjungan tunggal pada molar ketiga mandibula dengan pulpa nekrotik dan periodontitis apikal asimptomatik dapat berhasil dilakukan dengan diagnosis akurat dan teknik yang tepat. Pendekatan konservatif ini menunjukkan bahwa pencabutan bukan satu-satunya pilihan.Kata kunci sekali kunjungan, perawatan saluran akar, molar ketiga, saluran melengkung, restorasi komposit.
