Abstract

health problem, especially due to population growth, aging, poor lifestyle and diet, and high morbidity rates indicated by the increasing number of deaths each year. This study was conducted with the aim of determined the antidiabetic activity of samples used the Nelson-Somogi method. Samples were soxhleted used 96% ethanol solvent. The results of phytochemical screening, the extract showed positive content of phenol, flavonoid, saponin, tannin and steroid compounds. The in vitro glucose level reduction test used a UV-Vis spectrophotometer. The wavelength used was 740 nm and the incubation time was 25 minutes. The percentage of sugar level reduction after added the extract at a concentration of 1, 2 3; 4; 5 ppm was 28.56%; 40.48 percent; 51.99%; 60.74%; 70.72 percent. The linear regression equation obtained y = 10.458x + 19.128 R2 value = 0.9964. It can be concluded that the 96% ethanol extract of the sample has the potential to reduce blood sugar levels with an EC50 value of 2.95 ppm.

Abstract

ABSTRAK Nutraseutikal adalah produk suplemen makanan atau herbal yang dapat memberikan manfaat bagi kesehatan dalam pencegahan dan pengobatan penyakit yang berasal dari bahan-bahan alami. Gummy candies yaitu suatu sediaan produk manisan yang berbentuk lunak seperti jelly. Tekstur permen jelly dipengaruhi oleh bahan pembentuk gel atau hidrokoloid yang digunakan Salah satu tanaman yang mengandung antioksidan tinggi adalah belimbing wuluh. Belimbing wuluh digunakan sebagai zat aktif dalam sediaan gummy candies. Penelitian ini memiliki tujuan untuk mengetahui karakteristik gummy candies berdasarkan gelling agent alami yang digunakan dari sari daun cincau hijau (Cyclea barbata Miers.). Sediaan gummy candies dibuat dengan variasi konsentrasi yaitu 10% (F1), 20% (F2), 30% (F3), dan 40% (F4). Sediaan gummy candies dilakukan uji evaluasi meliputi organoleptis, pH, kadar air, keseragaman bobot, elastisitas, dan uji hedonik berdasarkan bentuk, warna, rasa, dan tekstur. Evaluasi hasil uji organoleptis berwarna merah, memiliki rasa pisang ambon, aroma khas, dan tekstur sangat kenyal. Uji pH (5,13-5,20), Uji kadar air (66%-72%), Uji keseragaman bobot kurang (1,093-1,269), Uji elastisitas (1,06-1,76). Berdasarkan hasil uji hedonik diketahui bahwa formulasi 3 dengan kadar sari daun cincau hijau sebanyak 30% adalah formulasi paling optimal karena lebih disukai oleh responden dan diterima dengan baik, dari sisi bentuk, warna, rasa, dan tekstur. Formulasi terbaik pada penelitian ini yaitu formula 3. Kata Kunci : Belimbing wuluh, Gummy candies, Cincau hijau, Pektin, Gelatin.

Abstract

Background: Diabetes mellitus remains a major global health issue, with rising prevalence due to population growth, aging, and lifestyle factors. Melastoma malabthricum L. has potential as an antidiabetic agent based on empirical data and in vivo study. Objective: To determine the content of secondary metabolite compounds contained in the extract of Karamunting leaves and to determine the activity and EC50 value of the extract for antidiabetes was evaluated using the Nelson-Somogyi method. Methods: This study used the phytochemical screening approach to identify the active chemicals. 96% ethanol was the solvent used in the maceration process to extract the leaves of M. malabathricum. The Nelson-Somogyi method was used to test the antidiabetic activity by calculated the sample's EC50 value on decreased glucose levels. The Nelson Somogyi method's antidiabetic activity was tested with a UV-Vis Spectrophotometer. It operated for 25 minutes at a wavelength of 740 nm. Results: Phenolic chemicals, flavonoids, saponins, steroids, and tannins all exhibited positive results from the phytochemical screening. After the addition of 96% ethanol extract of M. malabathricum leaves at gradually higher concentrations, the percentage decrease in glucose levels was 25.32%, 36.90%, 51.39%, 70.87, and 82.83% at concentrations of 1, 2, 3, 4, and 5 ppm. R2 = 0.9927 with y = 14.898x + 8.7747. Conclusion: 96% ethanol extract of M.malabathricum leaves contains several active substances, can reduce blood glucose levels and has an EC50 value of 2.76 ppm. Clinically, this implies a promising therapeutic potential with lower required dosages.
