Abstract

Atherosclerosis is a vascular pathological condition characterized by active, chronic, progressive inflammation and plaque formation in the arterial walls Interleukin 1-beta (IL-1β) cytokine influences every stage of atherosclerosis, from destroying monocytes and other immune cells to forming plaques. Caspase-1 protein plays a role in activating the inflammatory cytokine Pro-Interleukin-1 Beta (Pro IL-1β) to IL-1β. One of the plants that has the potential to treat atherosclerosis is Andrographis paniculata. Andrographolide and its analogs, such as neoandrographolide and deoxyandrographolide, are the main bioactive compounds with various pharmacological activities. This study aimed to screen the activity of the compounds through the description of the stability of the compounds in protein caspase-1. This type of research was an in-silico exploratory study of the caspase-1 protein (PDB ID: 1RWK), with compound preparations, protein preparations, validating the molecular docking method to docking andrographolide neoandrografolid and deoksiandrografolid compounds on the target protein. The compounds has an affinity for the target protein with binding energy values ​​of -5.95 kcal/mol, -6.02 kcal/mol, and -6.81 kcal/mol respectively, which are smaller than the native ligand -4.77 kcal/mol. Docking results show that andrographolide, neoandrografolid, and deoxyandrografolid can potentially inhibit caspase-1, which activates the pro-inflammatory cytokine IL-1β in atherosclerosis.

Abstract

Miana (Coleus scutellarioides) contains flavonoids as its primary active components and is widely used in traditional medicine, particularly in Indonesia. In Bali, which is renowned for its traditional healing practices (Usada Bali), miana is commonly utilized as a herbal remedy. The leaves of miana possess various pharmacological activities, including anti-inflammatory, antiproliferative, antidiabetic, anthelmintic, and antipyretic effects. Geographical conditions, such as the altitude of the growing region, are among the factors that influence the flavonoid content in miana leaves. This study aimed to determine the total flavonoid content in miana leaves from three different altitudes in Bali: Bedugul (1273 masl), Klungkung (393 masl), and Sanur (11 masl). An experimental laboratory design was employed, using maceration with 96% ethanol as the extraction method. The total flavonoid content was quantified using UV-Vis spectrophotometry, with quercetin as the reference standard. The results showed that miana leaves from Klungkung exhibited the highest total flavonoid content (540.85 ± 7.97 mgQE/g), followed by Bedugul (488.68 ± 6.07 mgQE/g) and Sanur (368.53 ± 11.2 mgQE/g). Statistical analysis revealed that the total flavonoid content of miana leaves from Klungkung was significantly higher (P<0.05) than that from the other two regions. These findings indicate that the altitude of the growing region affects the total flavonoid content in miana leaves, with the highest levels observed at an intermediate altitude (393 masl).

Abstract

Penerimaan barang yang baik dalam distribusi farmasi merupakan proses penting untuk memastikan bahwa obat yang diterima sesuai jenis, jumlah, kondisi fisik, dan berasal dari pemasok resmi, sesuai regulasi Cara Distribusi Obat yang Baik (CDOB). Penelitian ini bertujuan mengevaluasi implementasi SOP penerimaan barang di PBF X  berdasarkan pedoman CDOB. Metode yang digunakan adalah deskriptif observasional melalui wawancara semi-terstruktur dengan Apoteker Penanggung Jawab. Hasil menunjukkan bahwa PBF X  telah menerapkan sebagian besar prinsip CDOB dengan baik, termasuk pemeriksaan fisik, pencocokan dokumen, dan dokumentasi digital melalui sistem SAP. Namun demikian, ditemukan bahwa aspek pelaporan kerusakan belum sepenuhnya terintegrasi dengan sistem, serta belum ada evaluasi berkala terhadap efektivitas SOP. Diperlukan meningkatkan sistem pelaporan kerusakan secara digital dan melakukan audit SOP secara berkala guna menjamin keberlanjutan mutu distribusi.

Abstract

Background: Peptic ulcer disease (PUD) was a persistent gastrointestinal disorder characterized by mucosal damage in the stomach or duodenum. Cyclea barbata leaves had been reported to contain bioactive compounds with gastroprotective potential. Objectives: This study investigated the protective effects of 70% ethanolic extract of C. barbata leaves on ethanol-induced gastric ulcers in rats. Methods: Thirty male rats were randomly allocated into six groups. The groups consisted of a normal control, a negative control (Na CMC 0.5%), a positive control (ranitidine 13.5 mg/kg BW), and three treatment groups that received C. barbata ethanolic extract at doses of 100, 200, and 400 mg/kg BW. Treatments were administered orally for 14 consecutive days, and the animals were sacrificed on day 15. Ulcer count, ulcer diameter, ulcer index, and gastric pH were assessed. Results: The negative control group exhibited an average of 6.4 ulcers. Administration of ranitidine and all tested concentrations of C. barbata extract significantly reduced the number of ulcers compared to the negative control. The most pronounced effect was observed at 400 mg/kg BW, yielding an average of 1 ulcer and a significantly lower ulcer index (p < 0.05). In contrast, gastric pH values showed no significant differences among the treatment and negative control groups (p > 0.05). Conclusions: The findings indicated that the ethanolic extract of C. barbata leaves exerted gastroprotective activity, with the 400 mg/kg BW dose demonstrating the strongest ulcer-preventive effect.

Abstract

ABSTRACTPomegranate (Punica granatum L.) peel is a major agro-industrial byproduct rich in bioactive compounds, particularly polyphenols, which have attracted increasing interest for functional food and phytopharmaceutical applications. However, studies specifically analyzing the total polyphenol content of the local Balinese red pomegranate peel are still limited. This study aimed to determine the total polyphenol content and perform phytochemical screening of the red pomegranate peel collected from Bali, Indonesia. The research employed an experimental laboratory design. Red pomegranate peels were collected from Jadi Village, Tabanan Regency, Bali. The peels were dried, powdered, and extracted by maceration using 96% ethanol. Phytochemical screening was performed to detect major secondary metabolites. The total polyphenol content was determined using the Folin-Ciocalteu method with gallic acid as the standard, measured by UV-Vis spectrophotometry at 765 nm. All analyses were conducted in triplicate. The extraction yielded 25.78% of viscous ethanol extract. Phytochemical screening confirmed the presence of flavonoids, tannins, saponins, and steroids/triterpenoids. The standard gallic acid calibration curve showed good linearity with a correlation coefficient (R²) of 0.9916. The total polyphenol content was found to be 147.953 mg GAE/g at 1,000 ppm concentration and 34.136 mg GAE/g at 100 ppm. The findings demonstrate that the local Balinese red pomegranate peel contains significant polyphenolic compounds and secondary metabolites with antioxidant potential. This supports its potential for valorization as a source of natural antioxidants for functional food, nutraceutical, or sustainable packaging applications. Further studies are recommended to assess its antioxidant activity and to compare seasonal and varietal variations.Keywords:  Punica granatum L.; polyphenol content; Folin-Ciocalteu; UV-Vis spectrophotometry; phytochemical screening.ABSTRAKKulit delima (Punica granatum L.) adalah produk sampingan agro-industri utama yang kaya akan senyawa bioaktif, terutama polifenol, yang semakin menarik minat untuk aplikasi makanan fungsional dan fitofarmasi. Namun, studi yang secara khusus menganalisis total kandungan polifenol dari kulit buah delima merah Bali lokal masih terbatas. Studi ini bertujuan untuk menentukan kandungan total polifenol dan melakukan skrining fitokimia pada kulit buah delima merah yang dikumpulkan dari Bali, Indonesia. Penelitian ini menggunakan desain laboratorium eksperimental. Kulit delima merah dikumpulkan dari Desa Jadi, Kabupaten Tabanan, Bali. Kulit buah delima dikeringkan, dihaluskan, dan diekstraksi dengan cara perendaman menggunakan etanol 96%. Skrining fitokimia dilakukan untuk mendeteksi metabolit sekunder utama. Kandungan total polifenol ditentukan menggunakan metode Folin-Ciocalteu dengan asam galat sebagai standar, diukur dengan spektrofotometri UV-Vis pada 765 nm. Semua analisis dilakukan dalam triplikat. Ekstraksi menghasilkan 25,78% ekstrak etanol kental. Skrining fitokimia mengkonfirmasi keberadaan flavonoid, tanin, saponin, dan steroid/triterpenoid. Kurva kalibrasi asam galat standar menunjukkan linearitas yang baik dengan koefisien korelasi (R²) sebesar 0,9916. Kandungan total polifenol ditemukan sebesar 147,953 mg GAE/g pada konsentrasi 1.000 ppm dan 34,136 mg GAE/g pada 100 ppm. Temuan tersebut menunjukkan bahwa kulit delima merah Bali lokal mengandung senyawa polifenolik yang signifikan dan metabolit sekunder dengan potensi antioksidan. Ini mendukung potensinya untuk valorizasi sebagai sumber antioksidan alami untuk makanan fungsional, nutraceutical, atau aplikasi kemasan berkelanjutan. Studi lebih lanjut direkomendasikan untuk menilai aktivitas antioksidannya dan membandingkan variasi musiman serta varietas. Kata kunci:Punica granatum L.; kandungan polifenol; Folin-Ciocalteu; spektrofotometri UV-Vis; skrining fitokimia.
