Abstract

Free radicals can cause many diseases, such as cancer. Antioxidant is a compound that could scavenge free radicals. One of the natural source antioxidants is guava. The application of gibberellic acid (GA3) is known to affect the physicochemical properties and antioxidant content of the fruit. The goals of this research were to investigate the antioxidant activity of skin, flesh and fruit of crystal guava were given additional GA3 treatment with concentration 0; 60; 120; 180; 240; and 300 ppm by determining the value of the Antioxidant Activity Index (AAI) using DPPH; evaluate the total phenolic content (TPC), ferric reducing antioxidant power (FRAP) and physicochemical properties; analyse the correlation between the TPC with AAI DPPH by Pearson’s method. Extraction was performed by the maceration method using acetone 50% mixed with a magnetic stirrer for 1 h. Determination of AAI DPPH, FRAP and the TPC was performed by UV-Visible spectrophotometry. TPC in the flesh and fruit fractions tended to increase and FRAP values in all fractions decreased with GA3 application. The highest TPC and FRAP was given by the G3 treatment on the skin fraction and G0 treatment on the skin fraction. The highest AAI in all fractions was given by the G3 treatment. The TPC was significantly positively correlated with the AAI DPPH of the flesh and fruit fraction extracts (0.250 ≤ r ≤ 0.880), and the skin fraction extracts showed a strong negative correlation between TPC and AAI DPPH (-0.813 < r < -0.973).Keywords: Antioxidant activity; DPPH; FRAP; Gibberellic acid; Psidium guajava

Abstract

Natural products and their structural analogs are the main sources of pharmacotherapy. Various Aglaia species have been investigated since the 1960s for their phytochemical constituents and biological properties. This research focuses on isolating the terpenoid group from Aglaia shawiana, to provide a basis for further research to identify lead compounds for drug development. The hexane extract of Aglaia shawiana leaves was isolated using vacuum liquid chromatography and gravity column chromatography, employing solvents with varying polarities. The isolation process yielded two compounds: compound 1 and compound 2. A total of 15 mg of a pure isolate was obtained for compound 1, and 4.5 mg for compound 2. Their structures were characterized using 1D NMR (¹H, 13C, DEPT 135) and 2D NMR (HMQC, HMBC, 1H-1H COSY) and by comparing them with previously reported spectral data. It was concluded that compound 1 has the molecular formula C30H50O3, is a triterpenoid type cycloartane, and is named cycloartan-3β,29-diol-24-one, while compound 2 has the molecular formula C30H52O, is a steroid compound, and is named 23a-homostigmast-5en-3β-ol. These compounds were discovered for the first time in this species.

Abstract

With about 120 species, Aglaia is one of the largest genera of the plant family Meliaceae (the mahogany plants). Various Aglaia species have been investigated since the 1960s for their phytochemical constituents and biological properties. This research objective was to find secondary metabolites that have activity as anti-breast cancer compounds from endemic Indonesian Aglaia, such as Aglaia angustifolia Miq. Two cholestan type steroids, stigmast-5en-3α-acetat (1), as a new steroid with α-sterochemistry of acetyl moiety at C-3 and 23a-homostigmast-5en-3β-ol (2), with unusual side chain were isolated for the first time from the stem bark of Aglaia angustifolia Miq or known as segara tree in Kalimantan. The chemical structures of two steroids were identified with spectroscopic data, including IR, NMR (1H, 13C, DEPT 135°, HMQC, HMBC, NOESY, 1H-1H COSY) and HRTOF-MS, as well as by comparing with previously reported spectral data. These two steroids were isolated for the first time from this genus. Steroids 1 and 2 were evaluated for cytotoxic activity against MCF-7 breast cancer cells and showed weak activity with IC50 values of 829.0 and 903.0 µg/mL, respectively.

Abstract

The Bogor Regency dairy cattle livestock business area (KUNAK) is in Cibungbulang District, one of the livestock areas, and the product is cow's milk. The cow's milk produced is sold directly to consumers or milk producers. The selling value of fresh cow's milk is lower than processed cow's milk, and fresh cow's milk has the risk of being easily damaged, resulting in losses for farmers. This Community Service (PKM) activity aims to assist farmers in handling their processed milk production by making yogurt for the Cattle Farmers Group (KTTSP). The approach is in the form of an initial survey regarding partner problems, formulating solutions, providing assistance, and analyzing the partner's level of understanding. This community service activity results in an increase in partners' knowledge of dairy products and how to make them before and after the activity. Apart from that, partners can also increase the selling value of pure cow's milk through processed products in the form of yogurt with a delicious taste and attractive packaging, thereby increasing the selling value of the product.

Abstract

ABSTRACT. The Aglaia species, which contains triterpenoids, is the most numerous in the Meliaceae family. Aglaia foveolata (A. foveolata) is a type of plant that has many benefits, as medicinal ingredients. The potential of this plant is inseparable from the content of various bioactive compounds. This study aims to isolate, characterize the active compound from the twigs of A. foveolata and test its activity as an antibacterial. Three dammarane-type triterpenoids were isolated from the A. foveolata twigs which is, namely dammar-24-en-3β,20-diol (1), an epimeric mixture of shoreic and eichlerianic acid (2, 3). Their chemical structures were determined based on spectroscopic data using infrared, high-resolution mass spectrometry, and including one and two-dimensional NMR techniques, as well as through data comparison of the reported compound. Compound 1 was reported for the first time to be successfully isolated from this species. All these substances were tested for the first time for their antibacterial activity against two Gram-positive bacteria Staphylococcus aureus and Bacillus subtilis and two Gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa, through this study. Compound 1 was inactive, the epimeric mixture of 2 and 3 showed moderate antibacterial activity with a minimum inhibitory concentration (MIC) value ranging from 31.7 to 126.6 ppm, particularly against S. aureus with a MIC value of 31.7 ppm. Abstract

Aglaia merupakan salah satu genus terbesar dari famili tanaman Meliaceae (tanaman mahoni). Sejak tahun 1960-an, genus ini banyak diteliti untuk mengetahui kandungan fitokimia dan sifat biologisnya. Tujuan penelitian ini adalah untuk menemukan metabolit sekunder dari Aglaia Aglaia shawiana, yang memiliki aktivitas sebagai senyawa anti kanker payudara. Isolasi dan identifikasi struktur senyawa kimia telah dilakukan dari ekstrak metanol daun A. shawiana. Ekstrak metanol daun A. shawiana telah diisolasi, difraksinasi menghasilkan sebuah isolat dengan berbagai metode kromatografi kolom gravitasi. Penentuan struktur senyawa isolat dilakukan dengan metode spektroskopi resonansi magnetik inti 1 dimensi (1H-NMR, 13C-NMR dan DEPT-1350) dan perbandingan dengan referensi terdahulu. Isolat yang telah berhasil diisolasi dalam studi ini merupakan campuran senyawa β-sitosterol dan stigmasterol, dimana β-sitosterol lebih dominan jumlahnya dibandingkan stigmasterol. Isolat telah diuji aktivitas sitotoksiknya terhadap lini sel kanker payudara MCF-7 dengan metoda dimetiltiazol difeniltetrazolium bromid (MTT) dan diperoleh nilai IC50 sebesar 135,9 µg/mL.Mixture of β-Sitosterol and Stigmasterol Compounds from Methanol Extract of Aglaia shawiana Leaves and Its Cytotoxic Activity Against MCF-7 Cancer Cell. Aglaia is one of the largest genera of the Meliaceae plant family (mahogany plants). This study aimed to find secondary metabolites from Aglaia shawiana that have activity as anti-breast cancer compounds. Isolation and identification of the structure of chemical compounds have been carried out from the methanol extract of A. shawiana leaves. The methanol extract of A. shawiana leaves has been isolated and fractionated, resulting in an isolate by various gravity column chromatography methods. Determination of the structure of pure isolate compounds was carried out by 1-dimensional nuclear magnetic resonance spectroscopy (1H-NMR, 13C-NMR, and DEPT-1350) and comparison with previous references. The isolated fraction successfully isolated in this study is a mixture of β-sitosterol and stigmasterol compounds, where β-sitosterol is more dominant than stigmasterol. The isolate was tested for its cytotoxic activity against the MCF-7 breast cancer cell line using the dimethylthiazole diphenyltetrazolium bromide (MTT) method and obtained an IC50 value of 135.9 µg/mL.
