Abstract

Microbiology laboratories play an important role in supporting diagnosis and clinical decision-making, so the quality of test results must be guaranteed. The complexity of the testing process requires the consistent application of quality management through Quality Assurance (QA) and Quality Control (QC) at all stages of pre-analytical, analytical, and post-analytical. This study aims to analyze the effectiveness of QA and QC implementation in improving microbiology laboratory test results at Grandmed Lubuk Pakam Hospital. This study uses a qualitative approach with a case study design. Data were collected through in-depth interviews, observations, and quality document reviews of the head of the laboratory, the doctor in charge, microbiology laboratory analysts, and administrative staff. Data analysis was conducted thematically to examine the QA and QC implementation process and factors affecting the effectiveness of quality management. The results showed that QA and QC had been implemented through SOPs, internal quality control, result validation, and quality documentation. However, their implementation has not been optimal due to limited human resources, overlapping tasks, and inconsistent evaluation and follow-up of quality audits. Nevertheless, the implementation of QA and QC has contributed positively to improving the reliability of test results and clinician confidence. The conclusion of this study is that the effectiveness of microbiology laboratory quality management through the implementation of QA and QC is influenced by the adequacy of human resources, management commitment, and consistency of implementation at the operational level

Abstract

Background: Microbiology is the study of microorganisms, which are microscopic organisms that can only be observed through a microscope. These microorganisms have an important role in various aspects of life, including in the health sector. Objective: This study aims to evaluate whether alternative media, such as potato and corn, can be used as a substitute for Nutrient Agar in supporting the growth of Staphylococcus aureus bacteria. Methods: This research is experimental, by comparing the effectiveness of potato and corn media as an alternative to Nutrient Agar for the growth of Staphylococcus aureus. Results: The results of the research in the One-way Anova test obtained a significant value (0.00) which means (p <0.05), so it can be concluded that there is a significant difference between the ability of Nutrient Agar media, potato media and corn media in growing Staphylococcus aureus bacteria. To find out each group that has a significant difference, the Post-hoc follow-up test is carried out. The results of the post-hoc follow-up test shown in Figure 4.2.2 obtained a significant value between Nutrient Agar media and potato media (0.00), Nutrient Agar media with corn media (0.558), and between potato and corn media (0.00). Conclusion: The results showed that potato media was significantly different in its ability to grow Staphylococcus aureus compared to Nutrient Agar and corn. Potato media shows potential as an alternative to Nutrient Agar, despite its different effectiveness.

Abstract

Gastritis and gastroenteritis are common digestive disorders often associated with infections caused by the bacteria Helicobacter pylori (H. pylori) and Escherichia coli (E. coli). Rapid and accurate diagnosis is crucial for determining appropriate management, but conventional methods still have limitations in terms of sensitivity and turnaround time. This study aims to identify the presence of H. pylori DNA in gastritis patients and E. coli DNA in gastroenteritis patients using electrophoresis following DNA amplification via PCR (Polymerase Chain Reaction). This is a laboratory experimental study utilizing 10 patient blood samples, comprising 5 patients diagnosed with gastritis and 5 patients with gastroenteritis. The research procedure included DNA extraction, amplification of target genes (ureA for H. pylori and stx1 for E. coli), and visualization of the results using agarose gel electrophoresis. The results showed that of the 5 samples from gastritis patients, 4 samples (80%) exhibited a 294 bp DNA band, indicating the presence of the H. pylori gene. Meanwhile, all gastroenteritis patient samples (100%) showed a 302-bp DNA band, indicating the presence of the E. coli gene. The electrophoresis results showed clear and consistent DNA bands in identifying both bacteria. The conclusion of this study indicates that the PCR-based electrophoresis method is effective in rapidly and accurately detecting the presence of H. pylori and E. coli in clinical samples, thus having the potential to serve as a reliable molecular diagnostic method to support the diagnosis of gastritis and gastroenteritis.

Abstract

Gastritis and gastroenteritis are gastrointestinal infectious diseases that remain significant public health concerns, particularly in areas where Clean and Healthy Living Behavior (PHBS), personal hygiene, and environmental sanitation have not been optimally implemented. Gastritis is commonly associated with Helicobacter pylori infection, whereas gastroenteritis is often caused by pathogenic bacteria, including Escherichia coli. Limited public knowledge regarding the causes, modes of transmission, risk factors, and preventive measures for these diseases may increase the risk of infection. Therefore, this Community Service Program aimed to improve public knowledge and awareness of gastritis and gastroenteritis prevention through the introduction of Helicobacter pylori and Escherichia coli. The activity involved 30 participants and was conducted through health education sessions, interactive discussions, question-and-answer sessions, and the use of educational media, including posters and leaflets. The effectiveness of the program was evaluated using pre-test and post-test assessments to measure participants' knowledge before and after the educational intervention. The results showed that the mean knowledge score increased from 61.3 (moderate category) in the pre-test to 91.7 (very good category) in the post-test. These findings indicate that the educational intervention was effective in improving participants' understanding of the causes, transmission routes, symptoms, risk factors, and preventive measures for gastritis and gastroenteritis through the implementation of Clean and Healthy Living Behavior (PHBS), proper hygiene, environmental sanitation, and food safety practices. This community service activity is expected to enhance public health literacy and encourage sustainable healthy behaviors to prevent gastrointestinal infectious diseases.
