Abstract

Vibrio sp. is an opportunistic pathogenic bacterium that causes vibriosis in fish, shrimp, and seaweed. The use of synthetic antibiotics is everyday; however, resistance is difficult to prevent. The advantages of using natural antibiotics include their availability, environmental friendliness, and cost-effectiveness. This study aimed to utilize a natural material, namely rambutan (Nephelium lappaceum L.) leaf extract, as a candidate to be tested for its antibacterial activity against Vibrio sp. A phytochemical analysis of the rambutan leaf extract was conducted to identify the presence of bioactive compounds that inhibit bacterial growth. The extraction was performed through maceration using 70% ethanol as the solvent. The extract was then diluted in a series of concentrations ranging from 100% to 3.125%. Antibacterial activity was tested using the disk diffusion method on Thiosulfate Citrate Bile Salt Sucrose (TCBS) agar medium. The results showed a potent inhibitory effect at the 100% extract concentration, with an inhibition zone diameter of 24.675 ± 0.53 mm. Potent inhibition was also observed at 75% and 50% extract concentrations, with inhibition zones of 19.95 ± 1.56 mm and 19.225 ± 4.63 mm, respectively. Phytochemical analysis of the rambutan leaf extract revealed the presence of several bioactive compounds, including alkaloids, steroids, flavonoids, saponins, and tannins. The study concluded by calculating the extract yield to assess the efficiency of rambutan leaf extraction using ethanol, yielding 7.16 ± 0.14%.

Abstract

Study of the antibacterial ability of soursop leaf extract (Annona muricata L.) in inhibiting the growth of Vibrio sp. The maceration method with 70% ethanol solvent was used in this research to determine the yield, then the extract was diluted in stages from a concentration of 100% to 3.125%. The antibacterial test was done using the disc diffusion method in TCBS media and a phytochemical test was carried out on soursop leaves to analyze the secondary metabolite content. The extract yield obtained in this study was 3.77%. The test results showed that the largest zone of inhibition was found at 100% extract concentration of 11.45mm. The smallest inhibition zone was found in the 6.25% extract with an inhibition zone of 1.44mm, the diameter of the positive control (tetracycline) inhibition zone was 22.75mm, the diameter of 10% DMSO as a negative control was 0 mm. Phytochemical tests of soursop leaves show that they contain alkaloids, flavonoids, steroids, saponins and tannins.

Abstract

Pengobatan vibriosis dalam industri akuakultur memilih memberi makan ikan yang terinfeksi dengan obat antibiotik. Pemberian antibiotik sintesis bisa menyebabkan timbulnya resistensi antibiotik, dibutuhkan antibiotik baru dari bahan alam untuk mengurangi dampak bahaya bagi ikan, rumput laut, udang dan lingkungan sekitarnya. Salah satu tumbuhan yang dapat berfungsi sebagai antibakteri adalah tanaman belimbing wuluh. Penelitian ini bertujuan untuk memperoleh rendemen, mengukur kemampuan antibakteri Vibrio sp. dengan metode difusi cakram dan mengkaji kandungan metabolit sekunder dalam daun belimbing wuluh yang dilakukan maserasi dengan menggunakan pelarut etanol 70%. Tujuh perlakuan konsentrasi ekstrak yaitu ekstrak 100%, 75%, 50%, 25%, 12,5%, 6,25%, 3,125%. Hasil rendemen ekstrak etanol daun belimbing wuluh belimbing wuluh (Averrhoa bilimbi L.) sebesar 5%. Data hasil penelitian menunjukan adanya aktifitas antibakteri yang sedang dari ekstrak daun belimbing wuluh konsentrasi 100%, dengan zona hambat sebesar 16,95 mm. Aktifitas antibakteri yang lemah berada pada ekstrak konsentrasi 75% dengan zona hambat sebesar 11,75 mm. Konsentrasi ekstrak daun belimbing wuluh di bawah konsentrasi 75% tidak menghambat bakteri. Kajian fitokimia yang dilakukan dalam penelitian ini untuk mendukung sifat antibakteri Vibrio sp. Dari esktrak daun belimbing wuluh dan metabolit sekunder yang terkandung dalam daun belimbing wuluh adalah alkaloid, steroid, flavanoid, tanin dan saponin.
