Abstract

Community activities around the Pelus River cause pollution which affects the bacterial community found in the river. The aim of this research was to determine the characteristics of bacteria isolated from the Pelus river environment in Banyumas Regency. The method used in this research is an exploratory method where bacterial sampling is carried out at 3 different stations. The samples taken were then isolated, observed colony morphology, as well as biochemical and physiological characterization of the bacteria. The research results obtained 10 different isolates. A total of 10 isolates were obtained based on morphological differences. The characteristics of most of the Pelus River bacteria show variations ranging from enzymatic to environmental resistance which describes the waters of the Pelus River. Bacteria isolated from the water have characteristics that identify them as bacteria capable of degrading compounds

Abstract

The catfish is a freshwater cultivated fish that can be found all over Indonesia. However, the problems that occur in general are usually attacks by pathogens from the Aeromonas sp group. This research aims to optimize the rapid detection of the Aeromonas sp group using PCR at the Aquaculture Technology Development Center, Cangkringan, Sleman, special region of Yogyakarta. The research was conducted by survey with purposive sampling. Isolation was carried out using two growth media, namely Tryptone Soy Agar (TSA) and Glutamate Starch Phenile (GSP). Colonies that had characteristics of the Aeromonas sp group were subjected to PCR amplification using specific primers for the Aeromonas sp group. The results of the amplification resulted in DNA that matched the target, namely 953bp, which showed that the sample belonged to the Aeromonas sp group. Based on this technique, the level of accuracy in detecting pathogens is higher than conventional methods.

Abstract

The presence of vibrio bacteria in the breeding pond is one of the important factors that affect the activities of vaname shrimp breeding. Vibrio sp. bacteria in waters can cause losses if the number is above the threshold that should be. The high abundance of Vibrio sp. bacteria in the cultivation environment is an indicator that shows the potential for disease that can reduce the growth rate and survival of shrimp. The purpose of the study was to determine the abundance and dominance of vibrio bacteria in vaname shrimp breeding ponds using the TPC (Total Plate Count) calculation technique. The methods used include bacterial sampling, preparation of tools and materials, making bacterial culture media (NA and TCBS), dilution, bacterial inoculation, and calculating the number of bacterial colonies. The results of the calculation of bacterial abundance showed that there were 9 breeding ponds with different abundances. The highest abundance was found in pond BV1 with a total bacterial value of 9.6 x 105 CFU/ml. Meanwhile, the highest vibrio dominance was found in pond BV4 with a percentage of 43.69%.

Abstract

Peminatan konsumsi ikan sejak dini terutama pada anak yang menginjak sekolah dasar sangatlah minim. Hal ini disebabkan oleh minimnya pengetahuan mengenai pentingnya konsumsi ikan dan kecenderungan olahan ikan yang kurang menarik. Dalam menanggulangi hal ini diperlukan sosialisasi gemar makan ikan sejak dini pada lingkungan sekolah dasar. Target luaran dari pengabdian ini adalah peningkatan pengetahuam mengenai pentingnya konsumsi ikan, dan menarik minat anak anak SDN Rejasari dan MI Cokroaminoto Rejasari untuk mengkonsumsi olahan ikan. Metode pengabdian meliputi survey lokasi pelaksanaan kegiatan pengabdian, kegiatan penyampaian materi sosialisasi gemar makan ikan, kegiatan sesi tanya jawab, dan kegiatan pemberian sampel nugget ikan. Hasil pengabdian yang dilakukan di SD dan MI di desa rejasari berhasil menambah pengetahuan dan minat siswa/i terhadap konsumsi ikan. Hal ini dibuktikan pada sesi tanya jawab dengan rasa keingintahuan yang mendalam seputar gizi dan berbagai jenis olahan ikan. Selain itu sampel nugget ikan lele yang diberikan tim pengabdi juga sangat disenangi oleh siswa/i di desa rejasari. Olahan sampel nugget ikan lele yang tim pengabdi berikan juga membuat siswa/i di desa rejasari ingin mencoba olahan lainnya yang terbuat dari ikan.

Abstract

Amylolytic bacteria play an important role in the ecosystem, especially as probiotic and bioremediation agents in cultivation, as examples can be found in Mina Padi culture. The purpose of this study was to determine the amylolytic bacteria in the waters of the Mina Padi pond. Bacterial isolation began with bacterial sampling, inoculation and isolation of bacteria, calculation of the total abundance of bacteria, observation of bacterial morphology and bacterial purification, and isolation of amylolytic bacteria. The results of the isolation of amylolytic bacteria obtained 3 bacterial isolates capable of producing amylolytic enzymes, namely BA5, BA6, and BA7. The highest index of amylolytic activity was obtained by isolates of BA6 with a medium category of 2.3 cm, and the lowest index was obtained by BA5 with a weak category of 0.3 cm. The average bacterial abundance from each dilution was 2.5 x 103 CFU/mL.
