Abstract

Chrysanthemum cinerariifolium (C. cinerariifolium) is an empirically proven medicinal plant that has anticancer activities. This study aimed to profile metabolites, and cytotoxic activity of root, leaf, stem, and root extracts C. cinerariifolium on T47D cells and to determine the correlation of metabolite content with cytotoxic activity. The metabolite profile was carried out using UPLC-QToF-MS / MS, and cytotoxic activity was carried out using the MTT method. The results obtained in the form of a chromatogram were processed with the application Masslynk so that a metabolite profile data obtained. The data is then analyzed statistically using Principal Component Analysis (PCA). The results obtained on metabolite profiling showed that there were differences in metabolite profiles in the roots, stems, leaves, and chrysanthemums. The characteristic compounds in the flower section are D - (-) - Morphine and in the leaf part of genistein  and N - [(5-Chloro-1,2,3-thiadiazol-4-yl) methyl] -1- (2-isopropyl-4 -methyl-1,3-thiazol-5-yl) -N-methyl ethanolamine. The major compounds in the roots, stems, and leaves are Orphenadrine with successive percentages of 9.11%, 10.16%, and 3.24%, and the major compounds in the flower section are D - (-) - Morphine with a percentage of 10, 86%. Furthermore, the results of the cytotoxic activity test showed differences in anticancer cytotoxic activity in the parts of the flower, leaf, stem, and root of C. cinerariifolium. Besides, there is a relationship between the metabolite content and anticancer cytotoxic activity of each part of C. cinerariifolium. The higher the level of orphenadrine compounds in the plant, the higher the potential for anticancer. Keywords: C. cinerariifolium, UPLC-QToF-MS/MS, T47D cells.

Abstract

Leaf of Anting-Anting (Acalypha indica) is one of the plants that contain potent flavonoid compounds as antibacterial Staphylococcus aureus. The purpose of this research is to know physical characteristic of microemulsion of A. Indica leaf extract and to know antibacterial activity against Staphylococcus aureus bacteria. The microemulsions in this study used isopropyl myristate as the oil phase, tween 80 and span 80 as surfactant, isopropanol as kosurfactant and free water from CO2. Microemulsion was made by varying the concentration of the extract into 3 formulas, there are 5%, 10% and 15%. A. indica extract obtained using ultrasonic maseration extraction method with 70% ethanol solvent. All the formulas were evaluated for the characteristics of the preparations and the antibacterial activity test against Staphylococcus aureus bacteria with the sumuran diffusion method. The research results showed that the microemulsion of A. indica leaf extract has good physical characteristic with pH value between 4.9-5.8, deep oil type, that particle size F1 9.34 μm, F2 14.22 μm and F3 9.68 μm and stable physically at 25°± 2°C and 40°± 2°C. The result of antibacterial activity test showed that F1 did not differ significantly with positive control. However, the inhibitory power of F1 is still under positive clindamycin control of 12.98 mm and 15.05 mm. So it can be collected that F1 is an ideal formula which has good physical characteristics and has optimum drag power.

Abstract

The high morbidity or mortality due to diabetes mellitus becomes the world health problem. That fact encourages the study about antidiabetes drugs from natural material especially plants. This study aims to analyze the effect of ethyl acetate extract from Chrysophyllum cainito L. leaves in decreasing of blood sugar level of alloxan-induced male male wistar rats. This research is considered as experimental research with pre and post test randomized controlled group design. The experimental animals in this study were 25 male male wistar rats divided into 5 treatment groups, treatment with CMC-Na (negative control), metformin with the dose of 9 mg/200gBW (positive control), and leaves extract C. cainito dose 25; 50; 75 mg/kgBW. All treatment groups had significant decrease of blood sugar levels at 7, 10 and 14 day observations with p 0.05. While in the day 3 had insignificant decrease of blood sugar level with p 0.05. Every C. cainito leaves extract doses group have the ability to decrease sugar blood level better than negative control, but still below positif control. The optimum dose in decreasing sugar blood level in this study is 75 mg/kg BW.
